Silymarin has been claimed to have a benificial effect in various types of liver injury. In a prospective study in patients with acute viral hepatitis (n = 151) the effectiveness of this drug on the cause of the disease was tested. The groups with and without Silymarin (Legalon) were comparable concerning age and sex distribution and the frequency of HBs-antigen positive hepatitis; Laboratory findings (total serum bilirubin, activity of GOT, GPT and alkaline phosphatase and prothrombin time) were determined in intervals of 5 to 7 days over a period of 5 weeks beginning with the onset of jaundice. There were no statistical significant differences between both groups in the decrease of mean values of all parameters tested. The frequency of nearly normalized values of transaminases and serum bilirubin after 10, 20 and 30 days was not higher in the group treated with Silymarin as compared to the controls. It is concluded that Silymarin has no favourable effects on the cause of acute viral hepatitis.

Fasting serum bile acid (SBA) was measured by the enzymic fluorimetric method coupled with the NAD-resazurin system in 23 controls, 35 asymptomatic carriers of HIs antigen including 4 e antigen carriers and 91 patients with various liver diseases. All GHBs and e antigen carriers showed SBA within the normal range. SBA was most significantly correlated with serum bilirubin (gamma=0.74) and was a more sensitive index for impaired liver function than bilirubin or alkaline phosphatase in 164 radomly chosen samples from the liver disease group. In serial determinations of SBA with reference to GOT, GPT, changing patterns of these two parameters were classified into the parallel type and the discrepant type. Thirty two out of 40 cases with chronic liver disease belonged to the parallel type. SBA remained abnormal even after the normalization of transaminase in 12 out of 20 resolving episodes in cases of the parallel type, regardless of diagnosis. Since SBA changes according to the stage of the disease activity, serial and simultaneous estimation of SBA and GOT, GPT was found to be helpful in the observation of liver diseases. Maximum values of SBA elevation in an endogenous bile acid tolerance test after eating two egg yolks were higher than controls in 4 out of 7 cases with liver disease, who were associated with normal fasting SBA.

Non-antimicrobial actions of oleandomycin (triacetyloleandomycin and oleandomycin phosphate) were studied in patients with bronchial asthma. Twenty-one cases of the disease without associating infections entered the study, and they were given 750mg of oleandomycin or triacetryloleandomycin in three divided doses daily for two weeks. Clinical manifestations and laboratory findings were compared to assess the effectiveness of the antibiotic therapy between the three 2-week periods before, during and after the therapy. Improvements in clinical manifestations were attained in 11 of 21 cases (52.3%), and last after discontinuance of the therapy in 8(38.1%). The blood level of 11-OHCS as determined by the Demoorr's fluorescence method increased by greater than 20% at the end of thearpy in 7 of 18 cases (38.9%). In 5 of the 7 cases favorable responses were seen clinically to the oleandomycin therapy. The serum IgE level determined by the radioimmunosorbent test was compared before and after the therapy to reveal that oleandomycin caused decrease of IgE in 10 and increase in 9 of 20 cases examined. The oleandomycin therapy resulted increases by greater than 20% of the vital capacity and FEV 1.0 in 2 and 3, respectively, of 15 cases. Jaundice in association with elevations of the GOT, GPT and alkaline phosphatase developed in one patient, and generalized skin eruption in another. Both of these cases were given triacetyloleandomycin.

Experiments were carried out with a total of 25 chickens of the Leghorn breed divided into two test groups and one control group. The test chickens were given daily the phosphorus organic pesticides timet and lebaized at rates of 0.05 mg anen mg per kg. The treated birds were kept under observation in the course of 72 days recording their cholinesterase activity (ChEA), serumglutamatoxal-acetate transaminase (SGOT), serumglutamatpyruvate transaminase (SGPT), and alkaline phosphatase (APh), and blood sugar level. It was found that as early as the third day from the beginning of the experiment ChEA dropped abruptly--by 62.72 per cent following treatment with timet, and by 29.26 per cent when treatment was carried out with lebaizid. SGOT and SGPT rose up to the 36th day by 78.74 and 59.42 per cent as against the controls. The blood sugar level increased by 20.44 per cent following the administration of timet, and by 6.95 per cent following that of lebaizid. APh on the 12th day was by 60.84 per cent higher as compared to the initial level in the case of timet, and by 39.15 per cent in the case of lebaizid. Results showed that the continuous giving of phosphorus organic insecticides (timet and lebaized) at low rates to chickens leads to the drop of ChA and the rise of SGOT, GPT, APh, and blood sugar, these changes being more clearly manifested in chickens that have been offered timet.

Rats and hamsters were fed ETU at levels of 0, 5, 17, 60, 200 mg/kg in the diet. Body weights, food consumption, seric enzyme activities (GPT, alkaline phosphatase), hepatic enzyme activities (GPT, alkaline phosphatase G6PDH), cholesterolemia, thyroid weight and others organs, histology were the criteria studied. ETU was found causing hypercholesterolemia for the 2 species at 5 mg/kg dietary levels. Thyroid impairement is predominant in rat and hepatic impairment is predominant in hamster. ETU was found to be not carcinogenic for hamsters even at 200 mg/kg level and carcinogenic for rats at 60 mg/kg level for males and 200 mg/kg level for females.

Investigations were carried out on the serum enzymes, total proteins and some electrolites (potassium and sodium) in eight spleenectomized sheep experimentally infected with Babesia organisms. It was found that babesiasis in sheep was accompanied by a considerable rise in the activity of GOT and GPT (from 80JU up to 260IU and from 4 IU up to 19 IU). The amount of inorganic phosphorus dropped, and that of potassium sodium and alkaline phosphatase showed no changes. It is believed that the changes in the activity of GOT, GPT and inorganic phosphorus depend on the gravity and the duration of the disease and can be used as a criterion in determining the pathologic process.

The influence of Uden, exerted on the activity of GOT, GPT, LDH acid phosphatase and alkaline phosphatase in the serum, is investigated upon 4-day-long treatment. The experiment is conducted in three groups of white male rats with mean weight 270 +/- 20 g. The first group of animals underwnt poisoning with 1/20 LD50, the second--with 1/4 LD50 while in the third, control group, the animals were treated with an oil solvent at the respective days. It is found that acid phosphatase, GOT and alkaline phosphatase activity in group two is virtually unchanged, whilst GPT activity in both experimental groups, and acid phosphatase in group one are reliably increased (P less than 0,01), with 20,17 and 53 per cent respectively. LDH activity is reliably lowered with 60 per cent in group two, whereas in group one a tendency for a reduction is noted (P less than 0,05).

Blood samples from rats, rabbits and beagles were subjected to simultaneous analyses for ten parameters using the RaBA-System and the results were compared with those obtained by the routine manual methods. Although the mean values measured by the two methods differ in several items, a significant correlation between the two methods in the majority of the determinations makes the use of the RaBA-System acceptable. The RaBA-System is thus considered to be applicable to evaluation of hemoglobin, total protein, glucose, BUN, total cholesterol, GOT, GPT, and alkaline phosphatase in blood samples from rats, rabbits and beagles. Plasma LDH and albumin, however, cannot be determined with accuracy by the system.

In order to establish the normal baseline of blood values in rats using the RaBA-System, the following eight parameters were analyzed in 8-22-week-old Wistar-Imamichi rats of both sexes: hemoglobin, total protein, glucose, BUN, total cholesterol, GOT, GPT, and alkaline phosphatase. With the exception of glucose, all these parameters were significantly different depending upon ages. Significant sex differences were observed in the values of hemoglobin, total cholesterol, GOT, and alkaline phosphatase.

In an attempt to clinico-pathologically examine asymptomatic HBs AG carriers, follow-up studies were made on 57 HBAg-positive blood donors with the S-GPT within normal range and the following conclusions were arrived at: 1) The results of liver function tests made in the present studies revealed the following rates of abnormalities: the S-GPT was abnormal in 14.5% of the subjects, the S-GOT was abnormal in 9%, the serum total bilirubins were abnormal in 12.2%, the serum alkaline phosphatase level was abnormal in 24.5%, the TTT was abnormal in 4.4%, the ZTT was abnormal in 2.2%, the gamma-globulin was abnormal in 21.2%, and the ICG retention was abnormal in 25.6%. It was thus necessary to make a follow-up study of the results of liver function test. 2) Anti-HBs was negative in all subjects, the rate of lymphocytic blastogenesis in the peripheral blood (tested by the addition of PHA) was low in 7 (36.8%) of 19 patients, and the MIF test by the addition of the purified HBs Ag revealed that 17 subjects, excluding one subject with a histologic picture of acute hepatitis, were not susceptible to HBs Ag. It was, therefore, surmised that immunological insufficiency would be involved in the development of asymptomatic carriers. 3) Histologic examinations, made on 20 subjects, revealed A.V.H. in one subject, N.S.R.H. in seven, N.S.R. in ten and fatty liver in two, and further revealed mild, diffuse inflammations in 8 subjects in the first two group (40% in total). Further, pleomorphism was noted in the hepatocytes of 8 (40%) of these 20 subjects, and a study is under way of the significance of the pleomorphism.
