The hexane extract of Ferula jaeschkeana aerial parts was studied at an oral dose of 25 mg/kg per day for its postcoital effects in pregnant rats. Ovaries of treated rats remained in a cyclic state rather than undergoing pregnancy as demonstrated by constant ovulation accompanied by newly formed corpora lutea. Follicles were present in different stages of development. Uterine histoarchitecture of treated rats appeared non-receptive for implantation. No decidcoma were observed on day 5 of pregnancy and the luminal epithelium remained unresponsive. Uterus was non-oedematous and lumen was considerably wider. Administration of the extract caused increases in the protein and glycogen content of ovary and uterus, while the activity of acid phosphatase remained essentially unchanged and the activity of alkaline phosphatase was increased. The volume of uterine fluid in the treated rats was increased considerably on day 5 post coitum. It appears that the histological and biochemical modifications in the ovary and uterus of treated pregnant rats do not support the preparation of uterus for implantation.

Variants of the Cx43 gap junction protein have been detected on Western immunoblots by using an antipeptide antibody to the N-terminus of the protein. In heart ventricle, atrium, brain, retina, and uterus, different yet characteristic ratios of a broad 43-kDa band and a 39- to 40-kDa doublet were observed. These proteins (in lens epithelium, testes, and spleen) or their messages (in stomach, duodenum, kidney, and lung) were also detected in several nonexcitable systems but at consistently lower levels than found in electrically excitable tissues. The reproducible heterogeneity in electrophoretic mobility of Cx43 seen in different tissues does not appear to be due to proteolysis, since both the 43-kDa band and the 39- to 40-kDa doublet were recognized by an N-terminal as well as a C-terminal antibody. Furthermore, Northern (RNA) blots from different tissues show that both polypeptide profiles arise from indistinguishable transcripts. The conversion by alkaline phosphatase treatment of a predominantly 43-kDa profile (in heart) to a 39- to 40-kDa profile (characteristic of brain and protein translated in vitro from the RNA) suggests that the observed electrophoretic heterogeneity arises from tissue-wide differences in the phosphorylation state of Cx43.

Forty-five ovariectomised gilts in early gravidity received injections of progesterone doses between 120 mg and 40 mg as well as 250 micrograms/die of oestradiol-benzoate and oral application of 12 mg/die of norgestrel, the latter being administered in addition to the 40 mg dose of progesterone. The animals were killed between the 20th and 24th or on the 35th days of gravidity, before endometrial activities were determined of alkaline and acid phosphatases at various points of the uterus (centre and sides of ampullae and in-between ampullae). Under conditions of sufficient progesterone supply (120 mg/die and 100 mg/die), endometrial activities of both alkaline and acid phosphatases in the centre of ampullae were found to be higher than those in-between them. Activities were lower with inadequate progesterone supply (40 mg/die), and differences between points of sampling were less strongly pronounced for alkaline phosphatase. Endometrial glycogen concentrations in the centre of ampullae were lower than those in-between. Values in response to inadequate progesterone administration were lower than those following sufficient supply. With regard to glycogen concentrations, differences between points of sampling in the myometrium were below those in the endometrium.

The aim of this study was to produce large liver tumors reliably, and to diagnose the tumors during development. Therefore, New Zealand white rabbits were treated with N-nitrosodiethylamine orally three times per week by gavage and were examined by clinical-chemical assay at regular intervals during the average treatment period of 14 months. The total cumulative dose was 1200 mg N-nitrosodiethylamine over 14 months. After a short treatment period the initial dose of 3 mg/kg had to be reduced to 1.5 mg/kg. In all 11 treated animals (100%) liver tumors were seen at the end of the study. Four control animals did not show any neoplastic changes. Clinical parameters investigated were for an assessment of liver function, total protein, urea, creatinine, alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, albumin and neuraminic acid as well as some serum electrolytes. The in vivo diagnosis of liver tumors based on changes in these parameters proved to be relatively unreliable. The liver enzyme tests and urea concentration only yielded significant changes when the liver tumors were very large. Changes in neuraminic acid levels were the most reliable indicator for the presence of a liver tumor in this animal model. In the 11 treated animals, serum values of this marker increased towards the end of the study by an average of 300 mg/dl. The induced tumors were mainly hepatocellular carcinomas. Only in 1 animal was a hepatocellular adenoma found. Further primary tumors diagnosed were six adenomas in the kidneys and two uterus adenomas, as well as nasal cavity tumors (two papillomas, one carcinoma, one adenoma and one adenocarcinoma). In 70% of the treated rabbits the hepatocellular carcinomas had metastasized to the lungs.

Sixteen pregnant gilts were ovariectomised (Groups 1 and 2: 11th to 15th days of gravidity, Group 3: 19th to 22nd days of gravidity, Group 4: 34th to 45th days of gravidity). Daily intramuscular injections of 120 mg of progesterone and 250 micrograms of oestradiol benzoate were applied to them, beginning on the day of ovariectomy to preserve gravidity. All experimental animals were sacrificed between the 21st and 25th days of gravidity (Group 1) and between the 31st and 37th days (Groups 2 and 3) as well as between the 52nd and 61st days (Group 4). Living and dead embryos were numerically recorded, and histological als well as biochemical studies were conducted in all uteri. Embryo survival rates were normal, that is between 62.8 and 80 percent in Groups 1, 2, and 3, with the numbers of living embryos being 9.5, 7.5, and 8.0. Gravidity-specific uterus alterations were typical, despite constant hormone substitution. The thickness of endometrium declined with significance over the period under review. Plication was intensified, while the surface epithelium was flattened. Vascularisation increased, particularly in the subepithelial region, and the endometrial stroma became more oedematous. Full functionality was retained by the uterine glands in the period under review (increase in glandular epithelium as well as rising activity of alkaline phosphatase). Significant increase was recorded from the activity of acid phosphatase, whereas glycogen concentrations went down in the myometrium.

Previous studies have shown that fast homoarginine-sensitive alkaline phosphatase (FHAP) is roughly equivalent to CEA (Roche) as a marker in colon cancer. The present study compares FHAP with CEA-EIA (Abbott) as a marker in cancer of the colon, breast, lung, ovary, uterus, skin, and lymph nodes. Comparison is made with regard to sensitivity, specificity, predictive value of a positive test, and diagnostic efficiency. It was determined that FHAP and CEA-EIA were comparable as markers for cancers of the colon, breast, and lung. FHAP was more sensitive and specific and had a higher predictive value and diagnostic efficiency for cancers of the ovary, uterus, skin, and lymph nodes.

Toxic effects of beryllium salts on the reproductive organs of cyclic adult female albino rats have been studied. An attempt was made to overcome these effects using an Ayurvedic medicine Liv-52 (Himalaya Drug Co., Bombay). Liv-52-primed rats (1 mL/rat/day for 15 days) were exposed to beryllium nitrate intravenously and were sacrificed at different time intervals. At autopsy ovary, uterus, cervix, and vagina were processed for biochemical and histopathologic examination. Histoarchitecture of the ovary, uterus, cervix, and vagina revealed severe necrotic changes with beryllium nitrate treatment. Tissue glycogen content and the activity of alkaline phosphatase were inhibited significantly after beryllium treatment. Total and esterified cholesterol levels increased significantly in these organs when exposed to beryllium salts. However, a significant improvement was observed in the biochemical parameters and histoarchitecture of these organs when beryllium was injected into Liv-52-primed animals.

We have examined the potential for using calf uterine progesterone receptor (PR) as a substrate for phosphorylation by cAMP-dependent protein kinase (cAMP-PK), PR was found to interact with anti-PR monoclonal antibody alpha PR6 (Sullivan et al., 1986), which was to immunopurify the receptor. Protein staining of the purified preparation revealed the presence of two major bands corresponding to 114 kDa and 90 kDa peptides; only 114 kDa peptide could be photoaffinity-labeled with R5020. The 90 kDa peptide co-migrated with 90 kDa heat shock protein (hsp-90) precipitated by anti-hsp-90 monoclonal antibody AC88 (Riehl et al., 1985). Incubation of the immunopurified protein-A-Sepharose-adsorbed PR with the catalytic subunit of cAMP-PK in the presence of gamma-[32P]ATP and divalent cations resulted in a Mg++-dependent incorporation of 32P-radioactivity into both the 114 kDa and the hsp-90 peptides. Small 32P-incorporation was also seen in the 114 kDa peptide in the presence of Mn++. A 60 degrees C preincubation of immunopurified PR increased the extent of phosphorylation of the hsp-90 peptide. A pretreatment with alkaline phosphatase reduced the ability of PR to act as a substrate while the steroid occupancy of PR appeared to enhance the phosphorylation of the 114 kDa peptide. The differential cation requirement for the phosphorylation of 114 kDa and hsp-90 peptides and a selective hormone-dependent increase in the phosphorylation of the 114 kDa peptide suggest a possible role of phosphorylation in mediating progesterone action in the calf uterus.

In the experiment performed on 15 rabbits, action potentials as a group of spikes and separate pick fluctuations have been revealed electrophysiologically in the round ligament of the uterus and the proper ligament of the ovary. In various parts of the myometrial supravascular layer and in the ligaments of the uterus under normal conditions and at leiomyoma nicotinamidedinucleotide-tetrazolium reductase (NADH), nicotinamidedinucleotide-phosphate reductase (NADPH) and alkaline phosphatase activity has been determined both in the myometrial supravascular layer and in the ligaments of the uterus. At leiomyoma NADH activity is elevated, and that of NADPH--decreased in comparison with that in the intact organ. Capillary density in various parts of the myometrial supravascular layer at leiomyoma of the uterus does not noticeably++ differ from that under normal conditions. The data obtained prove the conclusion on a morphofunctional unity of the myometrium and the ligaments of the uterus. By the aggregate of a higher parameters of the enzymes investigated in the area of the posterior part of the isthmus of the uterus, a conclusion is made on a specific functional importance of this part in comparison with others.

Sixty spare human embryos at various stages of preimplantation development were prepared for cytogenetic analysis. Fluorescent staining of those with metaphases allowed scoring for the presence of a Y chromosome. In situ hybridization was then performed using a biotinylated Y-specific sequence, and the probe was detected by a standard streptavidin-linked alkaline phosphatase system. This enabled comparison of the chromosomal sex with that obtained after in situ hybridization in 28 embryos, and the sexing result obtained by the two methods was concordant in all cases. A further 21 embryos in which no metaphase chromosomes were obtained were sexed by biotinylated in situ hybridization only. Overall, 66 per cent of male interphase nuclei demonstrated a Y-specific hybridization signal. Results were obtained in under 24 h, which may permit the sexing of an embryo biopsied during cleavage and the transfer of sexed embryos at the blastocyst stage to the mother's uterus in the same cycle as oocytes are collected for in vitro fertilization.
