The liver was normal microscopically, with no evidence of bile duct obstruction. alpha-Fetoprotein in the tumor tissue was detected by immunoprecipitation reaction in agar. were also detected in the tumor tissue and total
alkaline phosphatase
activity of the tissue was very high. These findings suggested their tumor origin.

(CPALP) were detected simultaneously in the sera and body fluids of two male patients with gastric carcinoma matestatic to the liver. At autopsy, widely disseminated
gastric cancer
of Borrmann III type with liver metastases was revealed in both bases. Histologically, they were moderately differentiated tubular and papillary adenocarcinomas with marked cellular atypia and necrosis. In Case 1, the properties of CPALP were identical to Nagao type CPALP, and in Case 2 the Variant type CPALP. Using immunofluorescence, CEA and CPALP were demonstrated in both primary and metastatic cells. However, only in Case 2 was AFP observed in some of the primary tumor cells.

and comparisons of characteristics of highly differentiated adenocarcinoma (24 observations) and nondifferentiated cancer (51 observations) revealed no differences in the character of the secret and in the activity of
alkaline phosphatase
in tumoral cells of glandular and nondifferentiated cancer. At the same time different localization of primary cancer was established: nondifferentiated cancer--in the region of glandular necks and basal parts of the mucosa, adenocarcinomas--in the surface regions. Comparison of the data obtained with features of physiological regeneration of the epithelium of the gastric mucosa justifies the assumption that foveate epithelium of nondifferentiated cancer, epithelium of the glandular neck, was the source of origination of adenocarcinoma. A different histogenesis of adenocarcinoma and nondifferentiated cancer accounts for their morphological differences and characteristics of the clinical course.

obtained from gastric carcinoma tissues of 23 patients was separated into 3 bands (ALPa, ALPb, and ALPc) by polyacrylamide-gel disc electrophoresis. Ten cases showed ALPa only, 4 cases showed ALPa and ALPb, 5 cases showed ALPa and ALPc, and the other 4 cases showed ALPa, ALPb, ALPc, respectively. It was concluded that ALPa was similar in its enzymological and immunological properties to hepatic-type

, ALPb probably originates from the cancer cell itself. As for the other enzymes, ALPa and ALPc, further investigation should be made to determine whether they come from the cancer cell itself or not.

Glycylprolyl beta-naphthylamidase activities in sera from 40 normal subjects (18-81 years) were: 22.6 +/- 0.9 (S.E.) (11.8-38.2) I.U./1 serum at 37 degrees C. The enzyme activities did not differ significantly with age between the younger group under 40-years-old and the older group over 40-years-old. Males, especially under 40-years-old, had slight but significantly higher activities than females. The levels were decreased in patients with

. The levels were elevated in patients with hepatobiliary diseases, and had significant correlations with the results of the serum tests in hepatic diseases such as glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase,

with a "diffuse carcinomatosis of the bone marrow" (DCMB) showing a widespread osteoplastic bone metastasis. In a DCBM, a widespread invasion of cancer cells to the bone marrow occurs, causing leukoerythroblastosis, anemia, and disseminated intravascular coagulation. The local reaction in the bone, namely being osteoplastic or osteolytic, has not been discussed in previous papers. Our cases were characterized by an extremely high serum

, a "superscan" view of the bone scintigram, and a diffuse osteoplastic bone metastasis with an osteoid increase. Thus we think there are DCBM subtypes with a diffuse osteoplastic bone metastasis.

An unusual
alkaline phosphatase
(AP), named HuG-AP, was found in a newly established cell line (HuG-1) derived from a patient with
stomach cancer
. The enzyme was purified about 300-fold by affinity chromatography. On polyacrylamide gradient (4-30%) gel electrophoresis, the one band with the enzyme activity was observed. The enzymic properties of HuG-AP did not conform to those of liver, intestinal, placental, and germ cell AP isoenzymes which were recognized as homodimeric structure. Thermostability of the HuG-AP showed an intermediate value between intestinal and placental APs. Immunologically, the HuG-AP reacted with both anti-intestinal and anti-placental AP monoclonal antibodies. Dot blot analysis showed that both intestinal and placental AP mRNAs were expressed in HuG-1 cells concurrently. Therefore, we concluded that this novel AP had a hybrid form, namely heterodimeric structure, consisting of one subunit each of intestinal and placental APs. However, all of the properties of this hybrid AP did not conform to the intermediate enzymic properties between intestinal and placental APs which would be shown when they both coexist. Because an AP identical to HuG-AP had already been found in the metastatic lesion of the liver of the same patient, the expression of this novel AP seemed to occur in the patient's original cancer cells but did not result from spontaneous transformation of cultured cells.
