The records of 21 horses with an adenoma of the pars intermedia of the pituitary gland were reviewed. The animals comprised nine Dutch warmblood horses and 12 ponies. They ranged in age from 12 to 30 years (mean +/- sem 21 +/- 1.3 years) for 20 of the horses, and comprised 13 geldings and eight mares. All the animals showed hirsutism. In 1991, nine horses were diagnosed as having the adenoma, equivalent to 0.5 per cent of the horses examined in 1991. The mean +/- sem survival time of four of the horses (from discharge to the development of complications which required euthanasia) was 192 +/- 59 days (range 120 to 368 days). Routine biochemical measurements were within their normal ranges except for a high alkaline phosphatase activity (768 +/- 487 iu/litre) and a high plasma glucose concentration (7.8 +/- 0.9 mmol/litre) in 16 of the horses; only six had a glucose concentration above 9.0 mmol/litre. The basal plasma concentration of adrenocorticotrophic hormone (ACTH) in the diseased horses was at least 191 pg/ml. Eight of the horses had mean plasma cortisol concentrations before and after the administration of 25 iu of synthetic ACTH1-24 of 168 +/- 21 (range 74 to 248) and 478 +/- 58 nmol/litre (range 248 to 771), respectively. Analysis of the urine of five of the horses revealed a mean specific gravity of 1.034 +/- 0.0034 (range 1.025 to 1.045).(ABSTRACT TRUNCATED AT 250 WORDS)

Idiopathic hepatic fibrosis was diagnosed by liver biopsy in 15 young dogs, of which nine were German shepherds. Clinical signs included ascites, anorexia, weight loss and hepatic encephalopathy. Erythrocyte microcytosis was a consistent clinical feature, and clinical chemistry generally revealed hypoproteinaemia and high serum activities of alkaline phosphatase and, to a smaller extent, alanine aminotransferase. Fasting blood ammonia and serum bile acid concentrations were increased in most dogs examined, and all the dogs tested had prolonged retention of sulfobromophthalein at 30 minutes. Multiple acquired portosystemic shunts were revealed by laparotomy and/or portography. Non-inflammatory fibrosis was present to different degrees in all the dogs' livers, and on the basis of its predominant location these were classified as having central perivenous fibrosis, diffuse pericellular fibrosis or periportal fibrosis. The response to symptomatic treatment and anti-fibrotic therapy with glucocorticosteroids or colchicine was variable. Seven dogs died or were euthanased shortly after diagnosis, but one dog survived two-and-a-half years, and three dogs were still alive more than four years after the initial diagnosis.

Reference values for some haematological and plasma biochemical constituents were established in Spanish ibex (Capra pyrenaica hispanica) restrained either physically or chemically with tiletamine-zolazepam. The following variables were studied: haematocrit, haemoglobin concentration, total erythrocyte and leucocyte counts, haematological indices, erythrocyte dimensions, differential count of leucocytes, glucose, urea, uric acid, cholesterol, triglycerides, creatinine, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, creatinine phosphokinase, lactic dehydrogenase, gamma-glutamyl transpeptidase, total plasma protein, albumin, globulins, albumin-globulin ratio, sodium, potassium, calcium, magnesium, total phosphorus, chloride and osmolality. No haematological data have been published before but the values observed were in the general range of other artiodactyls, with the exception of the number and size of the erythrocytes which were respectively larger and smaller than in most other ruminants. Significant differences were found for a number of the variables between the values recorded in physically restrained animals and the values recorded in anaesthetised animals; they included the number of erythrocytes and related parameters, the plasma proteins and some inorganic ions.

Urinary enzyme activities of alanine aminopeptidase, gamma-glutamyl transpeptidase, alkaline phosphatase, N-acetyl-beta-D-glucosaminidase and beta-glucuronidase were determined in 15 dogs with leishmaniasis and in a group of eight normal dogs. Serum creatinine and blood urea nitrogen concentrations were also measured and renal histology was examined. All the affected dogs had renal lesions. However, no significant differences in blood urea nitrogen and creatinine concentrations were found between the control group and the affected group. The urinary enzyme activities of gamma-glutamyl transpeptidase (P < 0.01), N-acetyl-beta-D-glucosaminidase (P < 0.01) and beta-glucuronidase (P < 0.05) were significantly higher in the affected dogs. Urinary enzymes therefore seem to be a more sensitive and reliable test for assessing early renal damage in canine leishmaniasis than serum creatinine or blood urea nitrogen concentrations.

Plasma chemistry and haematological studies were conducted on chickens with coccidiosis. Male White Leghorn chickens, of two weeks old, were inoculated with 5 x 10(4) Eimeria tenella sporulated oocysts or with 1 x 10(6) E acervulina sporulated oocysts. Blood samples were taken four, seven and 11 days after inoculation. A wet chemistry system was applied to measure the plasma activities of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, gamma glutamyltransferase, creatine kinase, amylase and lactate dehydrogenase and the concentrations of creatine, total bilirubin, urate, total cholesterol, total protein, albumin, glucose and triglycerides. A dry chemistry system was applied to measure sodium, potassium, chloride and calcium. The number of red blood cells and packed cell volume were determined by a micro cell counter and blood pH was measured with a blood gas analyser. The erythrocyte count, packed cell volume, sodium and chloride levels in the chickens infected with E tenella were significantly (P < 0.05) lower than those of the uninfected controls. The significant decrease in blood pH of the chickens infected with E acervulina suggests malabsorption associated with duodenal lesions induced by the infection.

Tissue nonspecific alkaline phosphatase (AP) plays a well-known role in bone mineralization. This role was first suggested by a human AP deficiency disease, hypophosphatasia. Further studies with AP gene knockout mice have also suggested a role for AP in mineralization. However, AP is also expressed in other human tissues besides bone and cartilage, and this raises a question as to whether AP may also play a role in pathological mineralization such as dystrophic and vascular calcification. In vitro studies carried out in our laboratory indicate that a variety of cell types stably expressing membrane-bound AP can affect extracellular mineralization regardless of the tissue from which the cell lines originated (e.g. fibroblasts, vascular endothelial cells, or renal epithelial cells). This AP-mediated extracellular mineralization is both substrate/dependent and culture environment/dependent and may be consistent with a putative role for AP in pathological mineralization in tissues other than bone and cartilage. In this regard, it is interesting to note that high levels of AP are observed in vascular endothelia of small arterioles in brain and heart. It is probable that expression of AP in small arterioles of brain and heart may also contribute to the vascular hardening and calcification observed in humans. This in turn could be related to vascular aging, vascular disease, and the resultant weakening of and/or rupture of vessel walls.

Six weimaraner puppies, five of which were genetically related, showed systemic signs associated with hypertrophic osteodystrophy, including fever and involvement of the gastrointestinal, respiratory or nervous systems, in addition to the metaphyseal lesions. In five of the dogs the clinical signs developed less than 10 days after they had been vaccinated with a modified live virus vaccine. Radiographic findings suggested that both the hindlimbs and forelimbs were equally involved in the disease process. Abnormal haematological findings included leucocytosis with neutrophilia and monocytosis, and there was a consistent increase in the activity of alkaline phosphatase. Serum protein electrophoretic studies of three of the dogs revealed hypogammaglobulinaemia and abetaglobulinaemia in two of them. Conservative treatment with rest and non-steroidal anti-inflammatory drugs had little effect, and treatment with corticosteroids appeared to give the best results.

Pluripotent cells from the periosteal layer adjacent to cortical bone attain an osteoblast-like phenotype in culture when reaching confluence in monolayer. It is unknown whether such newly differentiated osteoblast-like cells preserve the chondrogenic potential characteristics for stem cells derived from the periosteum. Primary osteoprogenitor cells derived from bovine metacarpal periosteum were differentiated into alkaline phosphatase-positive osteoblast-like cells by an established monolayer culture protocol. After transfer into suspension culture in agarose gels, the cells differentiated into chondrocytes demonstrated by the production of collagen II, but not of collagen I, as well as alkaline phosphatase activity was abated. Contrarily, with continuation of monolayer culture, the cells maintained their osteoblast-like phenotype and secreted large amounts of collagen I and a minor quantity of collagen III and V. The alkaline phosphatase activity steadily increased during the entire culture period of 2 weeks. Thus, our culture techniques can serve as useful tools to study mechanisms of differentiation by modulating the phenotypic potential of osteogenic cells. The results presented here support the notion that the extracellular environment strongly influences the cell type and its metabolism.

A clinical biochemistry analyser designed specifically for veterinary use was used to analyse plasma samples from 24 vervet monkeys (Cercopithecus aethiops). Two millilitres of heparinised blood was collected from each of the 24 monkeys on four occasions at intervals of one week. Plasma was separated and analysed for the concentrations of triglycerides, cholesterol, total proteins, albumin, globulins, creatinine and blood urea nitrogen (BUN) and the activities of alkaline phosphatase (AP), lactate dehydrogenase (LDH), alanine aminotransferase (ALT), aspartate aminotransferase (AST), and creatine kinase (CK). The tests were easy to perform, used small volumes of plasma, and yielded consistent results for most of the analytes. The activities of CK and AP, but not AST, appeared to be influenced by haemolysis, and there were significant individual variations in the activity of LDH.

Fourteen calves were used to investigate the changes from birth to 83 days of age in the concentrations of serum albumin, alkaline phosphatase, beta-hydroxybutyrate, plasma cortisol, serum creatine kinase, creatinine, iron, plasma fibrinogen, serum gamma-glutamyl transferase, plasma glucose, haptoglobin, serum non-esterified fatty acids, total protein, transferrin, triglycerides, urea and gamma globulin; the haematological variables measured were: basophils, eosinophils, haematocrit, haemoglobin, lymphocytes, mean cell haemoglobin, mean cell haemoglobin concentration, mean cell volume, monocytes, band neutrophils, neutrophils, platelets, red blood cells and white blood cells. The changes are presented as a series of graphs and the values are discussed in relation to the published reference ranges for adult cattle. Two populations of calves were identified which gave rise to a bimodal distribution for some of the variables. Differences in haematocrit, haemoglobin and red blood cell counts were apparent at birth, with raised values for these measurements being associated with an increased white blood cell and neutrophil count between three and 27 days of age.
