The stabilities of different mRNA species were analyzed in a reticulocyte lysate system under protein-synthesizing conditions. In all cases examined the relative mRNA degradation by reticulocyte ribonucleases as well as by the interferon-modulated (2'-5') (A)n-dependent endonuclease correlated with the extent of (U)nA sequences within the 3' non-coding region. The experimental data presented indicate that according to their stabilities at least three major mRNA groups may be identified: (a) (U)nA-poor mRNAs (e.g. globin) are essentially stable and are only slightly degraded by the (2'-5')(A)n-dependent endonuclease; (b) mRNA species with intermediate (U)nA levels (e.g. Ig alpha and Ig mu heavy-chain mRNAs) are partially degraded by general
ribonuclease
activity and further degraded by the (2'-5')(A)n-dependent endonuclease and (c) (U)nA-rich mRNA species (such as c-myc and non-skeletal actin mRNAs) are inherently unstable and are extremely sensitive to degradation by general
ribonuclease
activity. A survey of mRNA nucleotide sequences demonstrated that without exception (U)nA-rich stretches appeared more frequently within the 3' non-coding region than in the coding or 5' non-coding regions. A comparison of 3' non-coding region sequences from 92 different mRNAs revealed that transiently expressed mRNAs, such as the interleukins, nerve growth factor,
epidermal growth factor receptor
, c-myc, c-fos, c-myb and several other oncogenes as well as interferons alpha, beta and gamma were exceptionally (U)nA-rich. It is postulated that differential mRNA stability may be partly determined by the primary nucleotide sequence and in particular by (U)nA sequences within the 3' non-coding region. This may represent a novel post-transcriptional strategy employed by the cell to selectively retain or destroy discrete mRNA species.

Transforming growth factor alpha (TGF-alpha) is expressed in the brain and affects cells by binding to the
epidermal growth factor receptor
(EGF-R). Using a
ribonuclease
protection assay, we found that TGF-alpha steady state mRNA levels in the mouse striatum peak during the first week of postnatal life. Temporally this peak correlates with the height of gliogenesis in the subependymal layer (SEL), which lies along the striatal border of the lateral ventricle. In vitro studies demonstrate that TGF-alpha can stimulate the proliferation of astrocytes, so glial fibrillary acidic protein (GFAP) mRNA levels were measured as well and it was observed that the peak of GFAP expression followed that of TGF-alpha by 1 week. Furthermore, in a TGF-alpha deficient mouse, waved-1 (wa-1), a significant reduction of GFAP mRNA levels and immunostaining for GFAP was found in the striatum. Bromodeoxyuridine labeling combined with immunohistochemistry of normal postnatal day 6 brain showed that the proliferating cells in the SEL are EGF-R immunoreactive. In the waved-1 SEL, there were fewer BrdU positive cells and there was a reduced level of [3H]thymidine incorporation. EGF-R immunoreactive cells were found in the SEL of the adult mouse brain. Taken together, our data suggest that the TGF-alpha/EGF-R signaling pathway is involved in postnatal mitogenic events in the brain.

The molecular mechanisms by which endometriosis persists in locations outside the uterus are unclear. Recently, the
epidermal growth factor receptor
(EGF-R) has been postulated to have a role in the disease process of endometriosis. To explore this, we determined the levels of EGF-R protein and messenger ribonucleic acid (mRNA) expression in endometriotic tissues and compared the levels to that of eutopic endometrium. Using rabbit anti-EGF-R antibody, we found more intense immunohistochemical staining for EGF-R in glandular cells than in stromal cells of both endometriomas and endometriotic implants. No difference in staining intensity was noted between endometriotic tissues and eutopic endometrium. A
ribonuclease
protection assay was used to determine mRNA levels for EGF-R. PhosphoImager analysis revealed the following levels of mRNA for EGF-R; eutopic endometrium, 1.00 +/- 0.27 (arbitrary units; mean +/- SEM; n = 6 patients); cyst walls of endometriomas, 0.21 +/- 0.12 (n = 10 patients); endometriotic implants, 0.29 +/- 0.13 (n = 9 patients); and pelvic adhesions, 0.03 +/- 0.03 (n = 5 patients). Endometriotic tissues had significantly less mRNA for EGF-R than eutopic endometrium (P < 0.05, by Newman-Keuls test). Our findings support the hypothesis that EGF-R may be associated with the disease process of endometriosis.

The effects of estrogen and progesterone on the expression of
epidermal growth factor receptor
(
EGFR
) in human endometrium were studied in hypogonadal women under conditions that simulated a normal menstrual cycle. All women received the same regimen of estrogen and progesterone and underwent serial biopsies. In one group of women (group I), a biopsy was obtained before receiving estrogen (CD0) and after 11 days (CD11) of estrogen replacement. A second group of women was biopsied on CD11 and CD21 to assess the combined effects of progesterone and estrogen (group II). Immunohistochemistry was used to test for the presence of
EGFR
, and a
ribonuclease
protection assay was used to assess the amounts of
EGFR
messenger ribonucleic acid (RNA) relative to ribosomal RNA in the tissue. In group I, a significant increase in
EGFR
messenger RNA from CD0 to CD11 was observed. A similar increase was observed to occur between CD11 and CD21 in group II. Immunostaining for
EGFR
was absent in all CD0 biopsies, but was present in all estrogen-exposed endometrium. No difference in immunostaining was noted between CD11 and CD21. We conclude that estrogen stimulates the synthesis of
EGFR
in human endometrium and that progesterone does not appear to modulate this effect. The examination of other parameters in hormone-replaced hypogonadal subjects will be valuable in understanding the complex physiological regulation of the human endometrium.

Wnt genes are involved in mouse mammary cancer, but their role in human cancer is unknown. Human Wnt5a was cloned from a placental cDNA library and used to assess expression by
ribonuclease
protection and in situ hybridization in human breast cell lines and in normal, benign, and malignant breast tissues. Human Wnt5a shows over 99% homology at amino acid level with mouse Wnt5a, and 90% with Xenopus Wnt5a. It was expressed only at low levels in breast cell lines and normal breast tissue. Benign proliferations and invasive cancer respectively showed 10-fold and 4-fold higher Wnt5a than normal breast tissues. The greater up-regulation in benign conditions suggests a role in aberrant differentiation. In situ hybridization localized the signal to the epithelial component. Wnt5a is the first member of the Wnt family to demonstrate overexpression in human breast cancer. It was not associated with factors known to affect breast cancer prognosis such as lymph node status or
epidermal growth factor receptor
status.

We have conjugated the murine monoclonal antibody (528) against the human
epidermal growth factor receptor

EGFR
) to mammalian pancreatic ribonuclease (
RNase
) via N-succinimidyl-3-(2-pyridyldithio) propionate (SPDP) and 2-iminothiolene (2-IT). The conjugate showed dose-dependent cytotoxicity against
EGFR
-producing squamous cancer cells (A431, TE8, TE5, Ca9-22) and no detectable cytotoxicity against
EGFR
-deficient small-cell lung cancer cells (H69). The cytotoxicity of the conjugate was positively correlated with the
EGFR
numbers of each cell line. The addition of excess 528 antibody to the medium protected A431 cells from the conjugate cytotoxicity. This immunoconjugate might be useful for targeted treatment of squamous cell carcinomas hyperexpressing
EGFR

Proto-oncogenes are involved in the regulation of gene expression, for example after ligand binding to growth factor receptors. Expression of the proto-oncogenes c-fos, c-jun, c-ha-ras and c-myc was studied in in vivo grown and in vitro cultured bovine preimplantation blastocysts employing RT-PCR,
ribonuclease
protection assay and immunohistochemistry. Thirteen- and 14- day-old preimplantation blastocysts, i.e. stages before and during trophoblast elongation, were used. In in vivo-grown blastocysts c-fos, c-jun and c-ha-ras transcripts as well as c-Fos, c-Jun and c-Myc proteins were detected in all stages studied. Cultured blastocysts were treated with 10 nM epidermal growth factor and 10 nM transforming growth factor-alpha simultaneously. Epidermal growth factor and transforming growth factor-alpha treatment induced c-fos mRNA and c-Myc protein expression. The induction of downstream targets of the
epidermal growth factor receptor
by epidermal growth factor and transforming growth factor-alpha indicates a functional epidermal growth factor signal transduction pathway in elongating bovine blastocysts.

The aim of this study is to assess the expression of
epidermal growth factor receptor
(
EGFR
) and c-erbB2 and their correlation with human papillomavirus (HPV) status and prognosis in squamous cell carcinoma of the cervix. The expression of
EGFR
and c-erbB2 was studied at the protein level using the immunohistochemical (IHC) staining method, at the RNA level using the
ribonuclease
protection assay and at the DNA level using Southern blot and hybridization method. One hundred and one patients with squamous cell carcinoma of the cervix were recruited. Fifty-one patients were of stage 1B/2A and 50 patients were of stage 2B and above. Positive IHC stainings of
EGFR
and c-erbB2 proteins were found in 74.2 and 19.8% of cases, respectively. DNA amplifications of
EGFR
and c-erbB2 genes were detected in 35.4 and 17.2%, respectively. Of the patients showing positive
EGFR
and c-erbB2 staining, only 39.2 and 25%, respectively, showed DNA amplifications. RNA overexpression of
EGFR
or c-erbB2 was only detected in 2% of cervical cancers and was associated with positive staining and DNA amplifications. HPV was detected in 79.2% of the cases by HPV consensus primers L1, in 57.4% for HPV 16 and 27.7% for HPV 18. The abnormal expression of
EGFR
and c-erbB2 had no correlation with HPV detection and had no prognostic significance on survival.

ERalpha-negative breast tumors tend to overexpress growth factor receptors such as

or c-erbB-2. Raf-1 is a key intermediate in the signal transduction pathways of these receptors. High levels of constitutive Raf kinase (Deltaraf) activity imparts ERalpha- positive MCF-7 breast cancer cells with the ability to grow in the absence of estrogen. Deltaraf transfectants maintained in estrogen-depleted media showed greatly diminished responses to 17beta-estradiol or the pure antiestrogen ICI 182,780. Western blotting, ligand binding, and immunohistochemistry assays revealed a loss of ERalpha protein expression, and

Transient expression of Deltaraf and double-label immunostaining showed ERalpha was lost in those cells that transiently expressed Deltaraf. - or c-erbB-2-overexpressing breast cancer cells is directly responsible for generation of an ERalpha-negative phenotype and, more importantly, that this process may be abrogated by inhibiting these pathways, thus restoring ERalpha expression.
