Cardiac hypertrophy was produced in rats by constriction of the ascending aorta. Removal of the constricting band 10 days after operation resulted in rapid decline in left ventricular (LV) weight and total ventricular RNA. Activities of acid RNase and beta-glucuronidase were elevated 3 days after aortic constriction. Activities of cathepsin D and alkaline RNase were unchanges. Activities of cathepsin D and acid RNase were unchanged 1 and 3 days after removal of constricting band. Ca2+-activated, neutral protease (CAF) isolated from postmitochondrial muscle supernatant was partially purified and characterized. CAF specifically degrades alpha-actinin when incubated with isolated myofibriles in the presence of Ca2+.

The following enzymatic activities were measured in serum of patients with benign and malignant ovarian tumors before treatment: alkaline and acid phosphatases, aspartyl (AspAT) and alanyl (AlAT) aminotransferases, leucyl (LAP) and alanyl (AAP) aminopeptidases, lactate dehydrogenase (LDH), gamma-glutamyl transpeptidase, cathepsin, alkaline ribonuclease (RNase) and beta-glucuronidase. It was shown that at least three determinations (phosphatases and LAP) are practically useless in a discrimination between the examined groups. RNase in combination with AspAT (AlAT) or RNase with AAP and LDH were found to give the best results as marker enzymes.

Several newly synthesized boron betaine analogs had antitumor activity in Ehrlich ascites, Walker 256 ascites carcinosarcoma, and Lewis lung screens and marginal activity in the B-16 melanotic melanoma screen. In vivo testing demonstrated that trimethylamine-cyanoborane inhibied Ehrlich ascites cell DNA and protein syntheses as well as gene modulation by chromatin protein phosphorylation and methylation. Trimethylamine-cyanoborane increased cyclic-AMP levels. In vitro testing showed that nuclear DNA polymerase, thymidylate synthetase, S-adenosylmethyltransferase, nonhistone chromatin methylation, deoxyribonuclease, ribonuclease, and cathepsin were inhibited by the boron analogs. These compounds did not demonstrate high antitumor activity at the doses employed, but blockage of methyl transfer from S-adenosylmethionine was established as a feasible method for controlling cell proliferation.

The effect of dieldrin on the bound and free activity of cathepsin, acid ribonuclease, acid phosphatase and aryl sulphatase has been investigated. Administration of dieldrin increased the activity of free cathepsin, acid ribonuclease, and acid phosphatase in rat liver whereas it did not affect the activity of aryl sulphatase. This indicates that dieldrin labilizes the lysosomes.

In rats treated with DOCA plus high salt or with high salt alone, hypertensive rats with renal vascular lesions showed an incomplete suppression of KRA. Cathepsin activity of rat kidney was higher under high salt loading than in the control. Beta-glucuronidase activity was greatest in rats with renal vascular lesions and smallest in rats fed on normal chow. RNase and DNase activities were greater in rats with renal vascular lesions than in rats without renal vascular lesions under high salt loading. 2) In rats of both sexes SHR showed greater KRA and cathepsin activities than WK rat under high salt loading. In female rats DNase, RNase and beta-GPase activities were greater in SHR than in WK rat under high salt loading. 3) KRA was higher in SHRSP aged 10 months than in SHRSR, though KRA of SHR was smaller than KRA of WK rat. Cathepsin activity was greater in SHRSP than in SHRSR. DNase and beta-NAGA activities were greater in SHR than in WK rat. 4) In 7 weeks of age SHRSR showed more PRC than SHRSP. At the age of 10 months SHRSP showed higher PRC than WK rat. The roles renin and lysosomal enzymes in hypertensive renal vascular lesions were discussed to some extent.

The effects of HPD plus light on lysosome membrane and release of hydrolysis enzymes from lysosomes of normal rat liver and hepatoma cells were investigated. HPD was bound to lysosome either in vitro or in vivo. Lysosomes bound HPD plus light increased lipid peroxidation of unsaturated fatty acid of lysosome membrane and enhanced the activity of DNase, RNase, cathepsin and acid phosphatase. These effects were related to HPD concentration and exposure time but it was unchanged in the control. It was concluded that the enhancement of hydrolysis enzymes from lysosomes was due to the lysosome membrane damage under the action of siglet oxygen produced by HPD bound lysosomes following light activation.

The metabolic activity of lysosomes was monitored by assaying the lysosomal enzymes. DNase, RNase, acid phosphatase and cathepsin in rat liver regenerating after partial hepatectomy at various intervals after the operation. Results of the present study demonstrate that the activities of lysosomal enzymes are increased significantly during the process of rapid regeneration, especially at 24 h and 48 h. A small rise in enzymatic activities was noticed at 36 and 72 h of regeneration, leading towards normalisation at 168 h. Administration of tin(II) tartrate (25 mu mol Sn2+/100 gm body wt.) significantly increased the DNAse and RNAse activity at 24 h of the regeneration, while a decrease was noticed in acid phosphatase activity. An increase was also found in the catheptic activity of tin-treated hepatectomized rats. It can thus be concluded that an increased metabolic activity of lysosomes is reflected by the increased activity of its hydrolytic enzymes during the regeneration, and tin at low concentrations is helpful to this process. Further work is in progress to gain insight into the other metabolic activity of the lysosomes during the process of regeneration.

1. Chinese-hamster ovary fibroblasts were cultured to provide a homogeneous cell population. Homogenates obtained from these cells were fractionated by centrifugation techniques and the resulting fractions were analysed for protein and for enzymes representative of certain subcellular particles. 2. Unlike those in rat liver homogenates, the mitochondrial and lysosomal populations proved impossible to separate by differential centrifugation owing to the similarity of their sedimentation properties. Their resolution was possible by using isopycnic centrifugation in a continuous sucrose density gradient. 3. The mitochondrial population equilibrated at a density of 1.17g.cm(-3) as in rat liver homogenates. However, the lysosomal population equilibrated at a lower rather than a higher density position than the mitochondria and the probable reasons for this are discussed. 4. The lysosomal population subdivided into two groups characterized by differences in acid hydrolase content and equilibrium densities. The fraction with a density of 1.15g.cm(-3) contained the majority of arylsulphatases A and B, of cathepsin and of beta-acetylglucosaminidase activities, whereas that with a density of 1.09g.cm(-3) contained the majority of the acid phosphatase and acid ribonuclease activities. The probable division of the lysosomal population of a single cell into a number of distinguishable subgroups is suggested.

1. Acid and alkaline protease activities in bovine anterior and posterior pituitary lobes were reinvestigated by measurement of u.v. and Folin-Ciocalteu colour values of trichloroacetic acid-soluble digestion products of denatured haemoglobin. 2. Both lobes of the pituitary gland contain a cathepsin with a pH optimum at 3.8. 3. When release of u.v.-absorbing material was used as the assay there was also an optimum at pH8.3-9.7, but this proved to be due to the release of nucleosides from an endogenous substrate. 4. The presence of a ;cyclizing' ribonuclease active at alkaline pH on endogenous RNA was confirmed by the inhibitory effects of phosphate, arsenate and bentonite. The activity was unaffected by heat, EDTA or metal ions. The enzyme also acted on exogenous RNA. 5. A purified preparation of neurosecretory granules from fresh bovine posterior pituitary lobes was free from alkaline ribonuclease activity. Most of the activity present in the tissue was recovered in the supernatant plus microsomal material. 6. The distribution of RNA did not follow that of the alkaline ribonuclease.

Rat mammary tumours induced by 7,12-dimethylbenz[a]anthracene can undergo repeated growth and regression during successive pregnancies. In a 10-day period after birth about half of the tumours regressed 50% or more. The concentrations of the lysosomal enzymes increased in regressing mammary tumours to the following multiples of the initial values: beta-glucuronidase, 7.7; beta-galactosidase, 3.9; cathepsin, 2.9; acid ribonuclease, 2.1; arylsulphatase A, 1.5; acid phosphatase, 1.4. In contrast, several non-lysosomal enzymes failed to increase. Activities in the post-partum uterus increased to the following multiples of the initial values: beta-glucuronidase, 5.8; cathepsin, 5.5; acid ribonuclease, 4.3; beta-galactosidase, 2.2; acid phosphatase, 1.8. Arylsulphatase A in the post-partum uterus decreased significantly, suggesting a non-lysosomal distribution or a special function related to pregnancy. No other significant changes were observed in the lysosomal or non-lysosomal enzymes in the hormone-independent liver or hormone-dependent normal mammary gland. The ratio of free to bound arylsulphatase A and acid ribonuclease decreased slightly 1-3 days after birth because of problems in homogenizing the tumours. At days 4-8, however, there was a dramatic increase in the ratio of the free to bound activities. The results can be explained in terms of the lysosomal theory of intracellular digestion.
