Polycations, including ribonuclease A,
ribonuclease
S protein and peptide, spermine, spermidine, and polylysines, enhance unstimulated and stimulated adenylate cyclase activity of beef thyroid membranes at low concentrations and inhibit these activities at high concentrations. Peak polylysine stimulation occurs with degrees of polymerization of 6 to 14, and for large polymers a potency limit for this maximum is reached at 4 X 10(-5) M expressed as lysine residues. Both enhancement and inhibition appear to be due to charge-charge interactions and are abolished by KC1. Polyanions are inhibitory only. The biphasic effect of polycations is seen on basal cyclase activity, occurs with
prostaglandin E1
- and 5'-guanylyl-imidodiphosphate-stimulated cyclase, but is most striking with thyrotropin. There is little enhancement of F--activated cyclase. The enhancement is not sensitive to changes in pH, Mg2+, or regenerating system and does not correlate with the stability constants between polycations and ATP. We suggest that the polycation effect is a general, electrostatic effect on membrane conformation and is not restricted to a particular receptor domain.

Liver fatty acid binding protein (L-FABP) binds avidly the arachidonic acid metabolites, hydroperoxyeicosatetraenoic acids (HPETEs) and hydroxyeicosatetraenoic acids (HETEs). Binding of 15-[3H]HPETE was specific, saturable, reversible, and rapid. Ligand was recovered from the protein complex and confirmed to be 15-[3H]HPETE by TLC. L-FABP bound HPETE with a dissociation constant of 76 nM,5-HETE at 175 nM, and 15-HETE at 1.8 microM, and the reference fatty acids oleic acid at 1.2 microM and arachidonic acid at 1.7 microM. Thus, the affinity was approximately 16-fold greater for 15-HPETE, and 7-fold higher for 5-HETE, than for oleic acid. The need exists for studies of complexes of L-FABP with the HPETEs and HETEs in hepatocytes, especially since L-FABP has previously been associated with mitosis in normal hepatocytes, and shown to be the target protein of two liver carcinogens, and these arachidonic acid metabolites have been found to be able to modulate activities related to cell growth.

A number of cytokines and their receptors are abundantly expressed at the materno-fetal interface and are thought to have a function in the regulation of placentation. Granulocyte-colony stimulating factor (G-CSF) is expressed by stromal cells in both placental tissue and maternal decidua throughout placentation. In this study, we examined the expression of placental G-CSF receptor (G-CSFR) mRNA and protein throughout gestation by

protection assays, Western blotting, and immunohistochemistry. The major placental form of G-CSFR mRNA, corresponding to a membrane-bound form of the protein, was present in first-trimester placental tissues; levels decreased in second- and were highest in third-trimester placental tissues. Two placental G-CSFR molecules, 120 kDa and 150 kDa, were detected in first- and third-, but not second-, trimester tissues. The level of the 150-kDa G-CSFR was greater in the third- than in first-trimester samples. These differences were irrespective of whether or not the patients had received

analogues and oxytocin, oxytocin alone, or mifepristone before labor. We demonstrated by immunohistochemistry that interstitial cytotrophoblast in first- and second-trimester decidual tissue and cytotrophoblast in term fetal membranes express G-CSFR. These data demonstrate that the expression of specific forms of placental G-CSFR is strictly cell type- and developmental stage-specific, and they suggest that G-CSFR may be important in decidual invasion of cytotrophoblast and in trophoblast function during placentation.
