Author followed up the activity of the three enzymes involved in the catabolism of nucleic acids--acid deoxyribonulease (
DNase II
), alkaline ribonuclease (RNase I), and acid ribonuclease (
RNase II
)--in the denervated gastrocnemius and soleus muscles of rats for 28 postoperative days. The activity of both acid nucleases increased in both types of denervated muscles, compared with the respective controls. Up to the 14th postoperative day, the activity excess of both acid nucleases was more significant in the m. gastrocnemius than in the m. soleus. The RNase I ran below the control activity during the whole period in the m. soleus and up to the 14th day in the m. gastrocnemius. The role of nucleases and nuclease inhibitors in the changes of nucleic acid catabolism in neurogenic muscular atrophies is discussed.

Effect of alcohol consumption on the activity of two lysosomal nucleases,
deoxyribonuclease II
(DNAase II) and
ribonuclease II
(RNAase II) and calcium concentration have been studied during liver regeneration of Sprague-Dawley rats over a period of 10 days following 70% partial hepatectomy. Liver weight was completely restored in partially hepatectomized rats at 8 days in both sexes, but ethanol treatment resulted in only a partial restoration of liver weight at 10 days. Specific activity of DNAase II in partial hepatectomized animals increased by 50-75% at 6-12 hrs above sham operated controls, and the specific activity of RNAase II increased 2.3 fold at 6 hr, while calcium concentration decreased by 50% at 6-12 hrs. Ethanol treatment masked and/or delayed the increase in the specific activity of both enzymes at early stages of liver regeneration and also masked the decrease in calcium concentration. These results indicate that ethanol consumption delays the process of liver regeneration by altering the activity of lysosomal nucleases and calcium concentration.

The activities and androgenic regulation of seven lysosomal enzymes viz. acid phosphatase, N-acetyl hexosaminidase, alpha-mannosidase, beta-glucuronidase,

RNase II
and phospholipase A was established in caput, corpus and cauda segments of monkey epididymis. Estimation of enzyme activities in the the epididymis of control, castrated and castrated-androgen replaced monkeys revealed that all the enzymes except
RNase II
showed higher activity in caput and corpus as compared to cauda. The enzymes were reduced markedly after castration and on subsequent androgen replacement there was a significant stimulation of the repressed activities, but the control levels were not restored.
RNase II
showed highest activity in cauda which was further elevated after castration. The possible role of these enzymes in sperm maturation and disposal is discussed.

Effect of repeated (20 days) exposure to picrotoxin (PTX) on rat liver lysosomal function was evaluated by measuring the free and total activities of acid phosphatase, cathepsin D,
ribonuclease II
(RNAse II) and
deoxyribonuclease II
(DNAse II). The free activities of the nucleases (both RNAse II and DNAse II) were increased following PTX exposure. The total DNAse II activity was increased by 2.2-fold whereas the total acid phosphatase activity was decreased by 28%. Consequently, the ratios of total activity / free activity were low in the PTX exposed groups, implying loss of membrane integrity. Cathepsin D activity was completely abolished. The results show that repeated exposure to PTX can lead to lysosomal dysfunction in liver.

Effects of treatmentin vivo with the antimalarials:chloroquine (CQ), primaquine (PQ) and quinine(Q) on lysosomal enzymes and lysosomal membrane integrity were examined. Treatment with the three antimalarials showed an apparent increase in the membrane stability. CQ treatment resulted in increase in both the 'free' and 'total' activities of all the enzymes i.e. acid phosphatase,

and cathepsin D. PQ treatment lowered the 'free' and 'total' activities of acid phosphatase and cathepsin D, but the
DNase II
activities increased. Treatment with Q resulted in increased 'free' and 'total' activities of
RNase II

DNase II
. While 'free' activities of acid phosphatase and cathepsin D were low; the 'total' activities increased significantly. Our results suggest that a generalized increase in free nucleases activities following prolonged treatment with antimalarials may lead to cell damage and/or necrosis.
