This is a review of historical facets of research in cutaneous neurohistology. The silver stains, used for some 150 years, led to the discovery of the neurone theory and to contemporary comparative studies. The
cholinesterase
methods, used since 1950, are the most convenient for general studies of nerves. The fluorescence technique of Falck (1962) is valuable for the study of adrenergic fibers. The electron microscope techniques used since 1950 have allowed the comprehensive description of myelinated and unmyelinated nerve fibers and of corpuscles. The most important unsettled questions pertain to the physiology of cutaneous nerves and precise definitions of cutaneous sensibility.

Alpha-fetoprotein (AFP) was determined by a new radioimmunoassay in the sera of patients with different liver diseases. Compared to a normal group (n = 140, AFP levels below 15 ng/ml), most elevated AFP concentrations were found in 18 patients with primary liver cancer (PLC), 7 of whom showed Ouchterlony-positive levels (above 10,000 ng/ml). In 3 cases with liver cirrhosis, PLC was first suggested by high AFP levels between 1000 and 3600 ng/ml and later confirmed by histology. On the other hand, only 6 from 15 patients with other primary tumors and liver metastasis had AFP concentrations between 20 and 111 ng/ml. In 90% of 102 patients with liver cirrhosis AFP levels below 20 ng/ml were determined, while 13 cases showed elevated values up to 134 ng/ml. A transitory AFP increase between 20 and 238 ng/ml was seen in 32% of 63 cases in the early stage of acute hepatitis but 65% of 31 these cases in follow-up studies. 3 of 7 cases of chronic hepatitis gave similar results. The maximal AFP levels developed following the maximal transaminase activities by 5-18 days and coincided with a decrease of
cholinesterase
activity. The radioimmunological determination of AFP is recommended for earlier detection of the development of PLC in liver cirrhosis patients.

An investigation was undertaken to demonstrate the neural elements of the lung of Francolinus pondicerianus, from the point of view of neurohistology and histochemistry. The staining of the neural elements was done by the
cholinesterase
technique with a maintained pH of 5.2, temperature 40 degrees C and incubation period of 19 h. Distribution of nerves in association with bronchial cartilage, pulmonary vessels and bronchi has been described and discussed. The distribution of the ganglia in association with blood vessels, bronchi, cartilage, various plexuses and the neural terminal terminal network has also been described. The innervation of the bronchi and their branches, and formation of the neural terminal network has been studied, as well as the distribution of
cholinesterase
in bronchi, blood vessels, muscles, ganglia, and nerve fibres.

In order to investigate the effect of nutrient deprivation on brain development, mouse cerebellum cultures were grown in nutrient media with reduced serum concentration. After 9--18 days in vitro, experimental cultures grown in media with low serum concentration exhibited a delayed and retarded myelination. Electron microscopic examination of experimental cultures revealed profiles of deficient myelination, but showed normal neuronal and synaptic structures. The amount of myelin basic protein measured by radioimmunoassay was markedly reduced in experimental cultures. Activity of
cholinesterase
in these experimental cultures was also decreased.

Phosfolan, chlorpyrifos, and stirophos when applied to white mice at sublethal doses did not induce any delayed neurotoxic effect. On the other hand, Leptophos and EPN when administered orally at sublethal or lethal levels clearly produced a delayed neurotoxic ataxia in treated mice. The five tested organophosphorus insecticides were compared for their ability to inhibit
cholinesterase
, neurotoxic esterases and monoamine oxidase. I50 values were estimated for each case. The results revealed that all five compounds were inhibitors of
cholinesterase
, but only Leptophos and EPN were shown to be potent inhibitors for both neurotoxic esterase and monoamine oxidase in the mouse brain. Additional particular properties of both Leptophos and EPN were found in their ability to cause delayed neurotoxic ataxia in chickens and sheep fed once on sublethal doses of these compounds. It is believed that the phosphonate ester configuration of EPN and Leptophos has a specific mode of toxic action which is mainly located at the central nervous system. It is also postulated that these delayed neurotoxic agents might inhibit postganglionic sympathetic neurons, thus resulting in chronic paralytic effects.

In 1976, epidemic organophosphate insecticide poisoning due to malathion occurred among 7500 field workers in the Pakistan malaria control programme. In July, the peak month of the epidemic, it is estimated that there were about 2800 cases. In field studies low red-cell
cholinesterase
activities were associated with the signs and symptoms of organophosphate insecticide intoxication. Toxicity was seen with 3 different formulations of the insecticide and was greatest with the products containing increased amounts of isomalathion, a toxic malathion degradation product. Poor work practices, which had developed when D.D.T. was the primary insecticide for malaria control, resulted in excessive skin contact with and percutaneous absorption of the pesticide. Airborne malathion concentrations were very low. Implementation of good work practices and proscription of use of the 2 pesticide formulations most contaminated with isomalathion halted the epidemic in September. An extensive training programme and surveillance system for pesticide toxicity preceded 1977 spraying operations.

activity in the microfilaria of the nocturnally periodic Wuchereria bancrofti has been described. Using histochemical methods,
cholinesterase
activity has been demonstrated in the amphids (Mundgebilde), phasmids (Schwanzgebilde), nerve-ring, excretory and anal vesicles of the microfilaria. It has been found that the enzyme activity disappeared completely after treatment with the specific
cholinesterase
inhibitor eserine. Evidence is presented that the greater amount of the
cholinesterase
activity detected is related to
acetylcholinesterase
and the functional significance of the enzyme in the enzyme reactive structures is discussed.
