The activity was determined of sorbitol-dehydrogenase (SDH),
cholinesterase
(CE), leucine-aminopeptidase (LAP),
acid phosphatase
(AP), and glutamate-dehydrogenase (GDH) in blood plasma of lambs irradiated with 200, 320, and 450 Rad. Results showed that with the mild form of acute radiation sickness (treatment with 200 Rad) the activity of the blood enzymes did not change. With the severe (320 Rad) and the peracute (450 Rad) forms of the disease within the first 24 hours the activity of SDH and CE rose 3 to 5 times, and that of GDH and AP--twice within a period of 1 to 3 days following irradiation. In the case of a threefold increased activity of SDH and CE within 15 to 24 hours of treatment the diseased animals died. Parallel hematologic investigations revealed that well manifested leukopenia with these two forms of the sickness took place after the 24th hour. Activity determination of the investigated plasma enzymes within the first 15-24 hours following irradiation could be used as an adjunct test to the study of the hematologic indices for the early diagnosis of the severe and peracute forms of irradiation sickness as well as for the exit of the disease.

This work examines ultrastructural and histochemical changes in the human nasal respiratory mucosa resulting from regular use of oral contraceptives (OCs). 25 healthy women aged 20-25 years were treated with Anovlar (4 mg norethisterone and .05 mg ethinyl estradiol) for 9 months. Biopsies were taken from 5 each of the 15 who did not develop nasal symptoms at 3, 6, and 9 months and from the 10 who developed symptoms at 9 months. A punch biopsy was taken from the lower border of the inferior turbinate, 1 cm behind its anterior end, using 4% xylocaine without adrenalization. The biopsy was divided into 2 parts for electron microscopic study and histochemical study. All symptom-free subjects developed the same changes: glandular hyperactivity, increased acid mucopolysaccharide content of the ground substance, and an increased defensive mechanism due to increased phagocytic activity. The ultrastructural changes and histochemical reactions were similar to those of symptom-free pregnant women. The 10 women developing nasal symptoms showed squamous metaplasia, interepithelial edema, glandular hyperplasia, histiocytic proliferation, and fibrous tissue deposition, all attributed to the action of estrogen. The morphologic changes in the liver were investigated along with those in the activity of the specific liver enzymes in the blood plasma of rats in the case of the severe form of acute radiation disease. The rats were treated with 670 Rad at the rate of 90 Rad/min. The studies were carried out on the 1st, 8th , 15th, 22nd, and 30th day following irradiation. It was found that under the conditions of the experiment the activity of the cytoplasmic enzymes sorbitoldehydrogenase ,
cholinesterase
, and leucinaminopeptidase strongly rose on the 1st day after treatment (the activity of sorbitoldehydrogenase increased 11 times). The activity of
acid phosphatase
and glutamate dehydrogenase also increased strongly, whereupon there was deterioration of the mitochondrial and lysosomal structures. Seen were well expressed processes of fatty and parenchymal dystrophy. The studies on the changes in the activity of these enzymes can be used as an adjunct, resp., an auxiliary test to the haematologic indices in the evaluation of the severity of radiation disease.

The present study demonstrates that under conditions of iso or hyperosmolarity, P. aeruginosa utilized carnitine as the carbon, nitrogen or carbon and nitrogen sources. As occurred in the case of choline, the bacteria synthesized
cholinesterase
(ChE),
acid phosphatase
(Ac.Pase) and phospholipase C (PLC) under any of these conditions and in the presence of high or low Pi concentrations. Carnitine acted as an osmoprotectant when the cells were grown in the presence of preferred carbon and nitrogen sources and high NaCl concentrations. Under these conditions the three enzyme activities were not produced. The osmotically stressed bacteria grown under any of the above conditions accumulated betaine. Its presence indicated that carnitine may be metabolized by P. aeruginosa to produce betaine which could account for the induction of the three enzyme activities or its action as an osmoprotectant. The phosphatidylcholine encountered in the host cell membranes allows the bacteria to obtain free choline by the coordinated action of PLC and Ac.Pase. Since the consequence of this action may be cell disruption, the increase of free carnitine in the natural environment of the bacteria is also possible. These two compounds, choline and carnitine, acting in conjunction or separately, may increase the production of PLC and Ac.Pase activities by P. aeruginosa and thus enhance the degradative effect upon the host cells.

Neutral endopeptidase 24.11 (NEP; "enkephalinase") may inactivate a number of centrally active neuropeptides including the enkephalins and substance P. In most areas of the central nervous system, the cell types which express NEP activity are not known. The hypoglossal nucleus (N.XII) was selected as a model system to characterize the cytochemical localization of NEP. The effect of hypoglossal nerve axotomy upon the distribution of NEP activity in the hypoglossal nucleus was compared to the effect upon cholinergic markers, the mu opiate receptor, and the enkephalins. By use of a fluorescence histochemical method, NEP was localized at all levels of N.XII to the soma and proximal processes of the majority of the apparent motor neurons in the nucleus. Fluorescent double-labeling studies revealed the presence of numerous enkephalinergic varicosities which localized to the neuropil surrounding NEP-stained motor neurons. To determine whether NEP was synthesized by these motor neurons, 18 rats received a unilateral transection of the hypoglossal nerve. A pronounced decrease in NEP staining in N.XII was observed on the operated side as early as 3 days following axotomy. This decrease persisted at all levels of the nucleus for about 5 weeks. By 7 weeks, the staining between the control and operated sides was indistinguishable. By contrast, there was no apparent change in the density or distribution of enkephalin-immunoreactive varicosities in five animals examined 6 to 32 days following axotomy. Radioligand binding of [3H]DAMGO to the mu-opiate receptor in N.XII was studied in 20 animals by quantitative autoradiography at 2, 6, and 11 days after axotomy. No significant changes in the level of radioligand binding to the mu-receptor were detected in response to axotomy. pseudocholinesterase
showed a coordinate decrease in motor neuron-associated staining on the operated side of N.XII at 3, 6, and 11 days following axotomy which paralleled the decrease in NEP staining. By contrast, the lysosomal enzyme marker,

, showed a pronounced increase in staining on the operated side. The results of this study are consistent with the synthesis of NEP by cholinergic N.XII motor neurons and indicates that the enkephalins and NEP in N.XII are closely associated, but derive from separate neuronal populations. "Contractile" arterial smooth muscle cells (SMC) return to a less differentiated "synthetic" state during adaptative and proliferative processes in vitro and in cell cultures. At present, the enzyme expression of the modulation of cultured SMC is partially unknown. In order to define metabolic events associated with cell modulation in vitro, we studied 16 enzyme activities in primary and secondary (P1-P3-P10) SMC cultures in comparison to in situ SMC in fetal and adult rat aorta. The "contractile" SMC in aorta of 2 months old rat showed very high nucleotide hydrolase activities (5'-nucleotidase, Mg-ATPase, Ca-ATPase), and naphthylesterase activities and weak lysosomal

activity; the glycolysis-linked dehydrogenases were expressed with higher activities than Krebs cycle-linked enzymes. In primary cultures, the SMC near the explant expressed a "contractile-like" enzyme behaviour, in opposite to cells in the peripheral part of growing area enzymatically similar to sub-cultured SMC. Proliferating SMC in secondary cultures were characterized by increased lysosomal activities and by the decrease or disappearance of Ca-ATPase, Mg-ATPase, 5'-nucleotidase, and
butyrylcholinesterase
activities like fetal SMC in vivo. These enzyme changes in subcultures might be related to a deficiency of nucleotide ester hydrolysis, abnormal adenosine and AMP levels, lowered lipolytic capability and increased lysosomal reactivity. In conclusion, subcultured "synthetic" SMC expressed enzyme cytochemical patterns different from those of "contractile" SMC and similar to those of fetal immature SMC. Their enzyme behaviour is unfavourable to contractile function and favourable to cell proliferation and lipid accumulation, two characteristic features of SMC in atherosclerotic plaques.

Our results suggest that the various histochemical changes in retina induced by intraocular pressure elevation were compensatory in the early stage and were beneficial to the supply of energy needed in retinal tissue and cellular metabolism; while in the late stage, the lesion of retina cells developed due to decompensation. SOD-CCE could alleviate the retinal lesions caused by intraocular pressure elevation, and can be used as auxiliary drug for the treatment of intraocular pressure elevation.
