Erythrocyte and serum enzyme system in the Gagu of the Ivory Coast have been investigated. Some systems (e.g. PGM) differ little between the subdivisions of the Gagu group, but others differ considerably (e.g. G6PD variants). In general the red cell enzyme frequencies fall within the range of variation characteristic of African populations. Serum cholinesterase variants are present only at low frequency, and the distribution of alkaline phosphatase phenotype shows the expected correlation with ABO blood groups.

The effect of daily dermal spray of malathion for four weeks in recommended (0.5 and 1.0 per cent) and higher (5.0 per cent) concentration on various enzymes in Bubalus bubalis species were studied. The higher concentration of 5.0 per cent showed lethal effect after 2 to 3 exposures. The cholinesterase activity in both RBC (RChE) and plasma (PChE) were inhibited with all the concentrations. There was also significant (P less than 0.05) elevation in the activities of serum aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase with 1.0 and 5.0 per cent spray and enzyme activities remained altered even during post-medication. The extent of various biochemical changes were dose and time dependent.

The influence of high intake of vitamin C in the young growing rats under administration of nickel sulphate in toxic doses has been studied. Ingestion of nickel sulphate depresses the growth rates of rats, alters the vitamin C status in different tissues, inhibits certain enzymes of vitamin C metabolism and changes the activities of alkaline phosphatase and succinic dehydrogenase in the liver and kidney tissues. The acid phosphatase activity of liver, kidney and brain tissues of rats and glucose-6-phosphatase activity in liver, and serum GOT activity were stimulated, with reduction in the in the liver GOT activity. There is stimulation in the activities of rat brain inorganic pyrophosphatase and cholinesterase. Kidney tissues of rats were found to be more susceptible towards nickel toxicity as compared to the hepatic tissues in respect of morphological alterations. There is almost no alteration in the hepatic lipid composition. Administration of vitamin C in high doses to rats fed nickel salts in toxic doses can restore not only the growth rates but also certain enzyme activities to a significant extent.

For the evaluation of certain differences in the diminution of export proteins of the liver we examined some exactly defined groups of liver diseases with the aim of further differentiation of the pathogenetic mechanisms. We measured the activity of glutamate-oxalacetate transaminase, glutamate-pyruvate transaminase, glutamate dehydrogenase, lactate dehydrogenase, alkaline phosphatase, cholinesterase and lecithin-cholesterol acyltransferase, the Quick value, the coagulation factors I, II, V, VII, VIII, IX and X. Clotting factors were determined by a Schnitger-Gross Coagulometer. Prothrombin, antithrombin III, plasminogen, factor VIII associated antigen and activated factor XIII were measured by immunoelectrophoresis according to Laurell. Lipoprotein electrophoresis in agarose gel was performed to evaluate changes in lecithin-cholesterol acyltransferase activity. Except of the rising diminution of export proteins in the course of liver disease from acute hepatitis to cirrhosis we found also specific changes of the patterns of the plasma specific enzymes. These proteins were diminished dependent on their half life time and the inflammatory activity--measured as the height of the transaminases. Lecithin cholesterol acyltransferase and factor VIII did not participate in the general diminution of the most export proteins; some details were found to explain this differing behaviour. Results are critically discussed with regard to new aspects in the biochemistry of the damaged liver cell.

After intraperitoneal administration of acrylonitrile into rats, fructose monophosphate aldolase was found in blood serum, activity of butyryl cholinesterase was inhibited but activity of alkaline phosphatase was unaltered in rat blood serum; these data demonstrate the necro-bystrophic type impairment. Decrease in the hepatotoxic effect of acrylonitrile after preadministration of antioxidants appears to be a result of stimulation of lipid peroxidation, preventing the toxic effect of this industrially important substance.

The methodology of a large prospective study on the influence of repeated anaesthetics on liver function is reported and the problems involved are discussed. The most suitable patients were those presenting for endoscopic examination of the bladder and urethra, for urethral dilatation and for cervical implantation of radium. Blood samples were taken immediately before induction of anaesthesia and on days 3-4 and 13-15 after operation, when a clinical assessment of the patient was also carried out. The concentrations of six enzymes (lactate dehydrogenase, alkaline phosphatase, aspartate aminotransferase, alanine aminotransferase, serum cholinesterase and gamma glutamyl transpeptidase) werechosen specifically as indices of liver function. The eosinophil count was measured to reflect any hypersensitivity reaction. The non-Gaussian distribution of these necessitated using appropriate non-parametric tests together with parametric tests on logarithmic transformed data. In addition a quantal method was used to measure the frequency of patients showing an "abnormal" increase in enzyme concentrations.

Lectins from Canavalia ensiformis, Phaseolus vulgaris, and Triticum vulgare react with arylamidase, alkaline phosphatase, gamma-glutamyltransferase, and cholinesterase of human sera by formation of enzymatically active, mostly insoluble complexes. Arylamidase, alkaline phosphatase, and cholinesterase react more intensely in sera of healthy people than in sera of patients with liver and neoplastic diseases. Arylesterase is bound to a distinct degree only by concanavalin A. The enzymes mentioned above also react slightly with the following lectins in order of decreasing intensity: Ricinus communis, Arachis hypogaea, Helix pomatia, Glycine max, Dolichos biflorus, and Ulex europaeus. Though multiple forms containing less sialic acid are favourably bound, preincubation with neuraminidase does not improve the reaction except with soybean lectin. Since higher concentrations of lectins react also with fast moving fractions of high sialic acid content, no steric hindrance of the binding between lectins and sialoenzymes is supposed, as concluded from determination of the total enzyme activity.

The approximative lethal dose of triacetin and diethylene glycole acetate is determined after the method of Deihmann and Leblanc. Experiments are conducted on white rats to establish the acute and subacute oral, dermal and inhalatory toxicity of the two substances. Changes in weight, liver and kidneys weight coefficient, hematopoiesis and hepatic function (biochemical and pathomorphological), as well as the stimulating effect on mucosa and skin are studied. The results of the study show a weak stimulating action on mucosa and skin, and insignificant cumulation. Emphasis is laid on the functional character of changes in the values of some enzymes -- alkaline phosphatase, cytochrome oxidase, cholinesterase -- and of the pathomorphologically established parenchymatous dystrophy. Presumably, it is a matter of changes more strongly manifested in imported triacetin. The conclusion is reached that imported triacetin may be substituted for lokally produced diethylene glycoldiacetate which proves to be with a lower acute and subacute toxicity.

The presentation deals with the enzymatic spectrum of the blood serum (aminotransferase-glutamino-pyruvic and glutamino-oxaloacetic acid, sera cholinesterase, histidase, acid alkaline phosphatase) in 100 patients with transient disorders of cerebral circulation in the form of ischemic and hemorrhagic strokes. These disorders of circulation appeared on the background [corrected] of atherosclerosis and hypertensive disease or in other combination along with vasculitis of a different etiology. The most significant were changes of histidase and acid phosphatase activity and an inhibition of cholinesterase activity in ischemic and hemorrhagic strokes with expressed focal disorders of cerebral circulation. In an improvement of the clinical state following medicative therapy there was a normalization of these indices. The only exclusion was histidase the content of which in some cases remained cunhanged.

The localization of non-specific alkaline phosphatase and cholinesterase activities evidenced in capillaries endothelium of the Rat brain have been compared in cerebral areas with or without the blood-brain barrier (BBB). There is a good topographical correlation: structures devoid of BBB (caudal part of the subfornical organ, choroid plexus, area postrema) do not display any non-specific cholinesterase activity with photonic microscope methods and an intense alkaline phosphatase activity; whereas structures with BBB (cortex, subcommissural organ, rostral part of the subfornical organ) show both enzymatic activities. At the ultrastructural level, more sensitive methods allow to disclose a non-specific cholinesterase activity in the reticulum endoplasmic cisternae and in the nuclear membrane in all endothelial cells. Alkaline phosphatase activities are present in the capillaries endothelium and in the conjonctive cells of pericapillaries spaces which exist in the circumventricular organs. Besides, these activities are closely related with the lateral and basal plasmalemmas of the choroidal cells responsible for the blood-cerebro-spinal fluid barrier. The apical plasmalemma is almost devoid of enzymatic activity. This result confirms the noticeable polarity of the choroidal cells, both morphologically and cytochemically.
