Embryos immobilized with neuromuscular blocking agents for differing periods between 4.5 and 9 days of incubation had an increased number of motoneurons in the brachial and lumbar lateral motor columns. Treatment with alpha-cobratoxin (alpha-CTX) on days 4--9, for instance, was able to prevent virtually all natural cell death during this period; control embryos had an average of 22,500 lumbar motoneurons on day 5.5, and 13,500 on day 10, whereas treated embryos had approximately 21,000 cells on day 10. Curare, alpha-CTX, alpha-bungarotoxin (alpha-BTX) and botulinum toxin were all about equally effective in preventing cell death. Similar treatment begun after day 12, however, had no effect on cell number. If even a partial immobilization was continued after day 10 (in embryos totally immobilized earlier) most of the excess neurons were maintained, in some cases right up to hatching, at which time the embryos died due to respiratory failure. In contrast, when administration of the immobilizing agents was stopped, allowing the embryos' motility to return to control levels, the excess neurons underwent a delayed cell death and total cell number fell to below control levels by days 16--18. Limb muscles from embryos with excess motoneurons exhibited relatively normal differentiation and had acetylcholinesterase (AChE) stained endplates which were innervated. Following curare treatment the two wing muscles, anterior and posterior latissimus dorsi, were found to have an increased number of AChE-stained endplates, whereas the only leg muscle examined quantitatively--the ischioflexorius (IFL)--did not; the IFL, did, however, have a markedly reduced variance in endplate distance, as well as other apparent differences suggesting an altered pattern of innervation. Our findings imply that the number of motoneurons undergoing natural cell death is closely related to muscle activity. Thus, functional interactions at the developing neuromuscular junction seem to be critical in controlling cell death. If a retrograde trophic factor is involved its action is somehow related to muscle activity.

Intrastriatal transplantation of fetal striatal (STR), cortical (CTX), or ventral mesencephalic (VM) tissue into the normal striatum has been shown to produce behavioral deficits (38). Here, we have examined the cellular elements of the transplants and their connectivity with the host using histochemistry for cytochrome oxidase (CO) and acetylcholinesterase (AChE), immunocytochemistry for glial fibrillary acidic protein (GFAP), OX42, tyrosine hydroxylase (TH), dopamine beta-hydroxylase (DBH), serotonin (5-HT), and cholecystokinin (CCK). Autoradiography for dopamine D1 and D2, muscarinic cholinergic, and serotonin 5-HT1 and 5-HT2 receptors at 5-15 months after transplantation was also investigated. CO staining showed that all transplants were metabolically active. The STR and VM transplants contained AChE-positive neurons and fibers. The CTX transplants exhibited AChE terminals with an appearance similar to that of the host cortex. AChE staining within the STR transplants was patchy. 5-HT-, TH-, and DBH-immunoreactive (IR) fibers were found in the STR and CTX transplants. In two of six CTX transplants, many TH-IR neurons were present. The VM transplants contained many TH-IR, 5-HT-IR, and DBH-IR cell bodies and fibers. CCK-IR stain was found in the VM transplant and was coextensive with regions containing TH-IR cell bodies. Fibers stained by all markers crossed the transplant and host border. Receptor autoradiography revealed that muscarinic cholinergic and 5-HT2 receptors were present in the STR, CTX, and VM transplants. In addition, dopamine D1 and D2 receptors were present in the STR transplants. Intermittent heavy staining for GFAP and OX42 were observed along the border of most transplants and the hosts. It was noted that high densities and hypertrophy of GFAP- or OX42-stained astrocytes or microglia, respectively, were present in the transplants and adjacent host. OX42-stained macrophages were found in many transplants. The present results indicate that intrastriatal transplants into the intact normal brain express numerous histochemical, immunocytochemical, and receptor features characteristic of the appropriate adult tissues. The afferents from the host extend into the STR and CTX transplants, and neural fibers from the VM transplants grew into surrounding host tissue, suggesting possible anatomical connection. Ultrastructural evidence is needed to determine if these fibers form synaptic connections. The results from GFAP and OX42 immunocytochemical staining support the possibility suggested by behavioral studies that damage to the host brain is induced by neural transplantation.

The Authors have evaluated the serum cholinesterase activity in patients subjected to 2 different schemes of polychemotherapy: COP (CTX, VCR, Prednisone) and C-MOPP (CTX, VCR, PCZ, Prednisone). The two schemes involve the administration of cyclophosphamide that, as has already been proved, significantly lowers serum cholinesterase level after only a few hours. The Authors have observed that the combination of cyclophosphamide with other antiblastic drugs or a longer treatment have little influence on the down curve of the serum cholinesterase in relation to what they have observed after treatment with cyclophosphamide alone.

As the Authors and Others have already demonstrated, cyclophosphamide leads to a rapid and prolonged decrease in the serum cholinesterase activity. In this work, the behaviour of the serum cholinesterase in patients subjected to C M F polychemotherapy (CTX, VCR, FU) has been studied. The grouping of patients on the basis of serum cholinesterase values before treatment has demonstrated that the enzymatic activity decrease arrives at level practically homogeneous. A month after interruption of therapy values equal to those at starting point have been observed.
