Testis
-specific genes are essential for spermatogenesis in mammalian male reproduction. We have identified a novel gene, Tsc21, exclusively expressed in mice and human testes from the results of the Affymetrix Genechip analysis in the six developmental stages of testis of postnatal Balb/C mice. The full cDNA length of Tsc21 was 810 bp, with a 543 bp open reading frame encoding a 180 amino acids protein with a predicted molecular weight of 21.040 kDa. A Blast search in the mouse genome database localized the Tsc21 gene to mice chromosome 6C3. Multiple amino acid sequence alignment of human, mouse, and rat homologous genes showed that mice Tsc21 protein was highly homologous with the human Tsc21 gene (70%) and rat Tsc21 gene (86%). -polymerase chain reaction analysis showed that the mice Tsc21 is exclusively expressed in the testis and epididymis of mice, and its expression is only detected after the mice is 35 days old. Human Tsc21 is also exclusively expressed in testis of human. Considering the expression profile Tsc21 in mice and human, we propose that Tsc21 may play a role during mammalian male spermatogenesis. Our study should be a basis for function characterization of the Tsc21 gene, leading to the elucidation of the molecular events underlying mammalian male reproduction.

Half-smooth tongue sole (Cynoglossus semilaevis) is a rare marine flatfish whose mature ovary and testis greatly differ in volume and weight. The length and weight of mature females are over twice greater than those of mature males. To obtain sufficient information on gonad differentiation and the relationship between gonad development and growth in the fish, we compared gene expression between the ovary and testis using suppression subtractive hybridization (SSH).

cDNAs are subtracted from ovary cDNAs and are used to establish an ovary testis-subtracted cDNA library. A total of 41 nonredundant clones are identified, including 20 known cDNAs, 9 uncharacterized cDNAs (EST clones), and 12 novel sequences. -polymerase chain reaction (RT-PCR). The results confirm that these genes are only expressed in the ovary and not in the testis, or at higher levels in the ovary than in the testis. At the same time, expressions of certain genes such as ZPC, survivin, aquaporin, CPEB, and O15 are demonstrated to possess sexual dimorphism in the kidney or muscle, and spleen. The results suggest that these genes could play key roles not only in the ovary but in other female tissues as well.

, cauda epididymis, body epididymis, caput epididymis, seminal vesicles, prostate gland, Cowper's gland, heart, liver, spleen, lung, kidney, stomach, hypothalamus, pituitary gland, cerebrum, cerebellum, and hippocampus of adult Meishan boar and sow tissues were examined. The expression pattern in the testis of 2-, 30-, 60-, 90-, and 150-day old Meishan boars were analyzed using real-time PCR. We constructed a eukaryotic expression vector for the Stra8 gene and used it to transfect NIH-3T3 cells and third generation pig spermatogonial stem cells (SSCs) cultured in vitro. Testes weight and sperm count in the cauda epididymis were evaluated at various time points. The results showed that the length of the pig Stra8 gene cDNA was 1444 bp encoding 366 amino acids with one typical helix-loop-helix (HLH) domain. It is testes-specific expression. Expression was first detected in boar testis starting at day 2, and its expression significantly (p<0.05) increased with age and body weight. When NIH-3T3 cells and pig SSCs were transfected with the eukaryotic expression vector EGFP (enhanced green fluorescent protein)-N1-pStra8, it was expressed in the cytoplasm of NIH-3T3 cells. However, in SSCs, Stra8 was expressed predominantly in cytoplasm and few in nucleus. Our data suggest that perhaps Stra8 acts as a transcription factor to initiate meiosis in young boar.
