Immortalized hybrid cells were generated by the somatic fusion of the cells from the forebrain of embryonic mouse with N18TG2 neuroblastoma cells. Three monoclonal hybrid cell lines, designated NF26, NF81, and NF83 (neuroblastoma forebrain hybrid cells), expressing an alpha isoform of Ca2+/calmodulin-dependent protein kinase II (CaM kinase II) were isolated, and their expression was demonstrated by immunoblotting and immunocytochemistry using a monoclonal antibody specific to the a isoform of the enzyme. The kinase activity of the hybrid cells was 2- to 3-fold higher than that of the parent neuroblastoma line N18TG2 cells. The neuronal origin of these lines was shown by their immunoreactivity to neurofilament protein, a neuron specific marker. Lines NF26, NF81, and NF83 are the first cell lines to express the gene of the alpha isoform of CaM kinase II in the brain.

Diisopropyl phosphorofluoridate (DFP) is a type I organophosphorus compound and produces delayed neurotoxicity (OPIDN) in adult hens. A single dose of DFP (1.7 mg/kg, s.c.) produces mild ataxia in hens in 7-14 days, which develops into severe ataxia or paralysis as the disease progresses. We have previously shown altered expression of several proteins (e.g. Ca2+/calmodulin-dependent protein kinase II (CaM kinase II) alpha-subunit, tau, tubulin, neurofilament protein (NF), vimentin, GFAP) and an immediate early gene (e.g. c-fos) in DFP-treated hens. Here we show an increase in protein kinase A (PKA) protein level and activity in the spinal cord at 1-day and 5-days time periods after DFP administration. We also determined the protein levels of protein kinase C (PKC), CaM kinase II and several phosphatases (i.e. phosphatase 1 (PP1), phosphatase 2A (PP2A), phosphatase 2B (PP2B) in the spinal cord of DFP-treated hens after 1, 5, 10, and 20 days). There was increase in CaM kinase II alpha subunit level after 10 and 20 days of treatment, and decrease in PKC level at 1-day and 20-days time periods in spinal cord mitochondria. In contrast, the cerebrum, which is resistant to DFP-induced axonal degeneration, did not show change in PKA and CaM Kinase II levels at any time period DFP post-administration. No alteration was found in the protein levels of PP1, PP2A, and PP2B at any time period. An early induction in PKA, which is an important protein kinase in signal transduction, followed by that of CaM kinase might be contributing towards the development of OPIDN in DFP-treated hens.

Cultured cerebellar granule neurons were used to examine the hypothesis that NMDA or KCl-dependent Ca2+ influx activates a signaling cascade mediating transcription, translation and assembly of neurofilaments. The data indicate that 25 mM KCl and 140 microM NMDA induce neurofilament protein, mRNA, and assembly in neurites, and these require Ca2+ entry through voltage-sensitive Ca2+ channels or NMDA receptors, respectively. The CaM kinase and transcriptional inhibitors, KN-62 and actinomycin D, respectively, attenuate the stimulatory effect of Ca2+. Granular neurons are a valuable model to explore the molecular basis for transcriptional regulation of neurofilament gene expression by neuronal activity.
