Erythrocyte properties of patients with congenital hypoplastic anemia were compared to those of patients with transient erythroblastopenia of childhood. The MCV was less than 85 in all nine
TEC
patients studied and greater than 90 in all 11 CHA patients. Hemoglobin F concentration was elevated beyond the normal level for age in eight CHA patients and almost always normal in
TEC
. The i antigen score was more likely to be elevated in CHA than in
TEC
. The activities of transaminase, aldolase, phosphofructokinase, and glutathione peroxidase were higher in CHA than in
TEC
(p less than 0.001). Some abnormal properties (namely, MCV and hemoglobi n F concentration) of CHA erythrocytes, present during remission but accentuated during relapse, seemed to vary with changes in serum erythropoietin. Early differentiation of
TEC
and CHA appears feasible, allowing prompt provision of a favorable prognosis and the avoidance of unnecessary corticosteroid therapy in
TEC
.

A new semiautomated apparatus for hemofiltration has been described. It is a postdilutional hemofiltration system that utilizes the RP-610 dialyzer and is made of standard lines used for hemodialysis, a blood pump and two precise volumetric pumps, Logeais-
TEC
model, for the ultrafiltrate and for the substitution fluid. The ultrafiltrate compartment of the hemofilter is connected to a manometer for detecting positive or negative pressure. The transmembrane pressure TmP is given only by the pressure on the blood side that is regulated by the screw clamp in the venous line. Once started the whole system may be regulated by this screw clamp in order to maintain zero pressure in the ultrafiltrate compartment, provided that ultrafiltration rate does not exceed 65 ml/min. This system is sufficiently accurate to replace bed scales or any other gravimetric device.

Three fistula sheep with average weights of 52.2 kgs were given 37.9 g of 15N and 14C labelled acetamide (= 1.09 mg 15N' and 0,95 mCi14C) which were administered directly through the fistula. The half-life period of 15N retention in the ruminal fluid (TCE soluble portion) was found to be 4 hrs. 18 hrs after 15N administration increasing amounts of 15N were carried back to the rumen by way of the rumino-hepatic circulation. The 15N concentration in the blood (TCE soluble portion) rapidly increased up to a peak value and, from 3 hrs after isotope administration, the 15N concentration was found to decline continuously, with a slight discontinuation at about the 10th hr of experiment. The rate of 15N incorporation into the protein fraction (
TEC
soluble portion) of the blood was delayed by 4 hrs, relative to the rate of 15N incorporation into ruminal proteins. An average of 43.1% of the administered amount of 15N was excreted in the urine within 7 days. Up to the 4th day of experiment the half-life period of urinary 15N excretion was 19 hrs. An average of 15% of the administered total amount of 15N was excreted in the faeces. In this process, the peak values in both TCE fractions were observed to occur on the 2nd day of experiment. The proportion of isotope in the TCE soluble fraction was found to increase continuously compared with the total amount of the isotope excreted in the faeces. Isotope concentrations between 0.03 and 0.13 atom% of surplus 15N were found in organ and muscle tissues of a sheep that had been slaughtered 7 days after administration of the isotope. The results obtained are discussed on the basis of comparisons made with the analogous behaviour of 14C activity.

T-cells have been implicated in autoimmune renal injury. To examine the role of T-cells in lupus nephritis we propagated T-cell clones from the cortical interstitium of MRL/lpr mice. All isolated kidney-infiltrating (KI) T-cell clones [6] express surface markers identical to the T-cells regulated by the lpr gene (Thy 1.2+, TCR alpha/beta +, Lyt-2-, L3T4-, B220+). Although KI T-cell clones have the same surface markers as lymph node-infiltrating (LNI) T-cells, they differ functionally. KI T-cells, but not LNI T-cells, are autoreactive and kidney-specific, exclusively proliferating to renal tubular epithelial (
TEC
) and mesangial cells. In addition, unlike LNI T-cell supernatants (SN), KI T-cell clones SN induce class II and ICAM-1 on cultured
TEC
. When KI T-cell clones are injected under the renal capsule, class II is increased on
TEC
. All clones transcribe mRNA for cytokines capable of inducing class II and ICAM-1 (IL-4, TNF-alpha, IFN-gamma). Anti-IFN-gamma mAb prevents the induction of class II and ICAM-1 on cultured
TEC
. Since class II and ICAM-1 expression on
TEC
precedes renal injury, the ability to propagate autoreactive, kidney-specific T-cell clones that induce these molecules provides evidence for their role in initiating renal injury in MRL/lpr mice.

Mouse P19 embryonal carcinoma (EC) cells express on their surfaces a Thy-1 glycoprotein. The expression of Thy-1 at the mRNA and protein levels is down-regulated during differentiation induced by retinoic acid (RA). Thy-1 is also expressed in human NTERA-2 EC cells, but its expression is not down-regulated during RA-induced differentiation. As a first step towards understanding differential regulation of the mouse and human Thy-1 gene in EC cells, we have introduced genomic DNA fragments encompassing the mouse or human Thy-1 gene into NTERA-2 and P19-derived cells and analyzed surface properties of the transfectants. In the transient transfection assay, both mouse and human Thy-1 genes were expressed on cell surfaces at comparable levels. P19-derived stable transfectants exhibited great clonal variations in the expressions of the transfected Thy-1 gene products, which in part reflected copy numbers. There was no simple correlation between the expression of the transfected Thy-1 gene and two stem cell surface markers,
TEC
-1 and
TEC
-4. In the course of differentiation induced by RA several clones with a surface phenotype of EC cells exhibited a significant decrease in the expression of the transfected mouse Thy-1, whereas expression of the human Thy-1 was less efficiently down-regulated. The results suggest the presence of multiple cis- and trans-acting elements controlling expression of the mouse and human Thy-1 genes in P19 EC cells and their differentiated derivatives.

Mouse eggs and pre-implantation stage embryos express on their surfaces a carbohydrate epitope,
TEC
-2, defined by an IgM monoclonal antibody,
TEC
-02. The
TEC
-2 epitope involves the oligosaccharide sequence GalNAc beta 1----4Gal beta 1----4 that is expressed on the plasma membrane and zona pellucida of mouse eggs and on a very limited number of other cell types. In this study we addressed the question whether or not the binding of
TEC
-02 antibody to the mouse eggs would interfere with their fertilization. Our data showed that the
TEC
-2 epitope is carried by two zona pellucida glycoproteins, ZP2 and ZP3. Binding of
TEC
-02 antibody to mouse eggs inhibited specifically and in a dose-dependent manner their fertilization in vitro. The inhibitory effect of
TEC
-02 antibody was dependent on the presence of an intact zona pellucida. Direct radioantibody binding assays indicated that the
TEC
-02 antibody completely inhibited fertilization at a concentration at which one quarter of all available
TEC
-2 binding sites was occupied. Binding of
TEC
-02 antibody to an egg did not interfere with initial attachment of the sperm to the egg but inhibited maintenance of sperm binding to the zona pellucida, the secondary binding. The combined data indicate that
TEC
-2, which is a well-defined zona pellucida specific carbohydrate epitope, might be a part of the secondary sperm receptor.

In this work we report our 24 months experience in the placement of 119 peritoneal catheters in 105 patients (59 males, 46 females, mean age 60.1 years; range 24-90) using the peritoneoscopic insertion technique. After catheter implantation a dialysis solution leak is encountered in 7.5% of cases, tunnel infection in 4 cases (3.3%) and exit site infection in 12.5% of the catheters. Placement by endoscopic control is considered as a technique able to avoid catheter migration; in our series we reported this complication in 15 cases (12.5%). The easy access to the peritoneal cavity and the atraumatic insertion of the catheter obtained with the Y-
TEC
procedure reduced the hospitalization period from 25 to 5 days on average. In our experience actuarial survival of catheters is 82.5% at 24 months.

Serological diagnosis was examined for deep mycotic infections, especially candidiasis. Candida antigen and antibody researches were made by means of CAND-
TEC
(RAMCO Co., Ltd) and passive hemagglutination test (PHA) (Roche Co., Ltd), respectively, and simultaneous determinations were made of D-arabinitol, a fungal metabolite and (1----3)-beta-D-glucan, fungal parietal component. 1) For normal subjects, 1 of 173 cases (0.6%), 5 of 200 cases (2.5%) and 7 of 157 cases (4.5%) showed greater than or equal to 1:4, greater than or equal to 320 x and greater than or equal to 11 mumol/l for CAND-
TEC
, Candida antibody and D-arabinitol, respectively, and values above these were judged positive. 2) Of the 171 patients with pyrexia refractory to general antibiotic agents, who were treated at our Department and related institutions from November 1988 to March 1990, 41 with obvious Candida infections were CAND-
TEC
, Candida antibody, D-arabinitol and (1----3)-beta-D-glucan positive in 33 (80.5%), 21 (51.2%), 29 (70.7%) and 13 cases (56.6%), respectively. At least one item-positive patient was as high as 97.4%. On the other hand, in the group having no Candida infection at all, 1 of 57 cases on CAND-
TEC
(1.7%) and 10 of 55 cases on Candida antibody (18.2%) were positive, indicating significant differences from the surely Candida-infectious group. 3) Comparative examination of CAND-
TEC
and other testing methods revealed a correlation of CAND-
TEC
with D-arabinitol in cases showing 1:4 and 1:8 less than or equal to, but not other significant difference.(ABSTRACT TRUNCATED AT 250 WORDS)

Candida spp. is frequently detected from perioperative patients, especially from those requiring central venous catheter (CVC) management. CAND-
TEC
could detect the blood fungus infection easily and quickly. We analyzed 438 specimens from 57 patients requiring perioperative CVC management for more than three weeks, and investigated usefulness of CAND-
TEC
comparing with other serological methods. The dilution rate of CAND-
TEC
showed a positive correlation with amount of beta-D-glucan which was measured by Toxicolor test and Endotoxin-specific assay. It also showed a positive correlation with amount of D-arabinitol. When blood samples diluted more than four times were positive with CAND-
TEC
, the patients were considered to be infected, and accumulative infection rate was up to 84.2% three weeks after introduction of CVC management. CAND-
TEC
would be useful in detecting Candida infection compared with conventional methods, and the fungus infection was much more frequent incidence than ever expected. It is important to detect fungus infection early and respond immediately to avoid nosocomial infection.

3,157 PTCA were performed in Austria in 1991, this is 27% more compared to 1990. Prognosis for 1992 is another 27 to 30% more. Frequency of PTCA exceeded bypass surgery in 1990 for the first time in Austria. WHO-guidelines of 1,000 interventions per year and 1 million inhabitants will be achieved in Austria by 50% in 1992. In this context it is worth while to state, that installation of further cathlabs is recommended preferably within established centers including established heart surgery. The following NON-POBA-PTCA-techniques ("New devices") were used in Austria for the first time: Atherectomy, stent, lasers,
TEC
, ROTABLATOR, ROTACS, magnum wire and angioscopy accounted for 11.3% of all PTCA cases in Austria recently. A total of 447 valvuloplasties were performed in Austria until now, frequency of pulmonary and mitral valvuloplasties is increasing, that of aortic valve interventions decreasing. High frequency, Radiofrequency and Direct current Ablations of supraventricular- and ventricular- or WPW-arrhythmias were performed in 184 patients in Austria until now. In 247 patients with rare conditions, interventions like atrioseptostomy, implantation of umbrellas, etc. were performed. State of interventional cardiology in Austria is appropriate, compared to other European countries.
