A rapid and reliable analytical method for the determination of trace elements in human bone by atomic absorption spectrometry is reported. Calcium was determined to estimate the homogeneity of samples. Human bone from the iliac crest was obtained at autopsy of adult subjects. Before analysis samples were decomposed by microwave digestion and acid digestion in a Parr bomb. Zinc, rubidium, strontium, calcium and iron were determined by flame atomic absorption spectrometry (FAAS) and aluminium,

and lead by electrothermal atomic absorption spectrometry (ETAAS) at optimum measurement conditions. The results for the two digestion procedures agreed for zinc, rubidium and calcium within +/-5%, for
copper
within +/-7% and for strontium, iron, aluminium and lead within +/-10%. The repeatability of measurement (R.S.D.) for determination of calcium and trace elements after microwave digestion and acid digestion in a Parr bomb was tested in one representative autopsy bone sample by six parallel determinations. It was found to be better than +/-5% either for microwave digested samples or samples digested in a Parr bomb, for all elements determined by FAAS and ETAAS techniques. A continuous-flow extraction system was developed to speed up, facilitate, and improve the accuracy of the chemical fractionation of metals in solid materials. A three-step sequential extraction scheme was used to evaluate the novel system by analyzing calcium (Ca), iron (Fe), manganese (Mn),
copper
(Cu), and zinc (Zn) in a soil certified reference material (National Institute of Standards and Technology [NIST]
SRM
2710). In the proposed system, extraction occurred in a closed chamber through which extractants were passed sequentially. The extracts were collected in a number of subfractions for subsequent name atomic absorption analysis. Apart from the advantages of simplicity, speed, and less risk of the contamination that flow analysis systems usually possess, the continuous-flow system can improve the accuracy of chemical fractionation of metals by sequential extraction. The system ensures that extraction is performed at designated pH values without any need of adjustment. Variation of sample weight to chamber volume ratios from 1:12 to 1:40 had no effect on the extractability of the metals studied. In the extraction of the acid soluble fraction, concentrations of acetic acid in the range 0.11 to 0.5 mol L(-1) had no significant effect on the amounts of metals extracted, except Fe. Increasing the concentration of hydroxylamine in the reducible fraction step from 0.04 to 0.5 mol L(-1) affected the extraction efficiency for Fe, Mn, and Zn. The extraction profile, rather than a single value of extracted concentration, of each element offers additional information about the kinetics of leaching processes and chemical associations between elements in the solid materials.

The biodistributions of carborane-containing
copper
porphyrins, CuTCP and CuTCPH, have been studied previously in mice bearing subcutaneously implanted mammary carcinomas. We now report biodistributions of those porphyrins in Fischer 344 rats bearing intracranial and/or multiple subcutaneous isogeneic 9L gliosarcomas (9LGS). The porphyrin was given either by i.v. infusion or by multiple i.p. injections. When 190 mg CuTCPH/kg body weight was given to the rats by i.v. infusion, median tissue boron concentrations (microg/g) 3 days after the end of infusion were: 64 in subcutaneous tumor, 13 in intracranial tumor, 1 in blood and 3 in brain. When 450 mg CuTCPH/kg body weight was given to the rats by serial i.p. injections, the median concentrations (microg B/g) 4 days after the last injection were: 117 in subcutaneous tumor, 50 in intracranial tumor, 4 in blood, and 4 in brain. CuTCPH biodistribution was also studied in xenografts of the human malignant gliomas U87 and U373, and of the murine

-6 mammary carcinoma and the rat 9LGS, each grown subcutaneously in mice with severe combined immunodeficiency (SCIDs). In SCIDs, median boron concentrations (microg/g) 2 days after the last s.c. injection of a total of 190 mg CuTCPH/kg body weight were: 251 in U373, 33 in U87, <0.6 in blood and <0.5 in brain. Because there were such high boron levels in the U373, and because xenografted U373 is similar to spontaneous intracerebral human glioblastoma multiforme (GBM) microscopically, CuTCPH could prove useful as a boron carrier for boron neutron-capture therapy (BNCT) of GBM and of other human malignant gliomas.

2711 by ICP-AES was carried out with high accuracy and precision (RSD<3.7%). The certified concentrations of the SRMs were obtained for arsenic, cadmium, lead, and silver by using an ultrasound-assisted extraction method with a digestion solution of (1+1)-diluted aqua regia. The determination of
copper
in SRMs by the ultrasound-assisted extraction method and analysis by ICP-AES failed to obtain the certified concentrations at the 95% level of confidence using (+/-2 s) as confidence limits of the mean. However, the same results were observed with the use of the microwave digestion method and reflux, which is the ISO 11466 standard method. The analysis of the SRMs showed that the ultrasound-assisted extraction method is highly comparable with the other methods used for such purposes. The major advantages of the ultrasound-assisted extraction method compared to the microwave and reflux methods are the high treatment rate (50 samples simultaneously in nine minutes) and low reagent usage, the main benefit of which are the low chloride and nitrate concentrations in the extracts.

containing IUD. Pretreatment and 1, 3, and 6 month treatment blood samples were obtained and assayed for serum cholesterol, triglycerides, and high density lipoprotein (HDL) cholesterol. In the subjects using EE+LYN, the HDL cholesterol/cholesterol ratio was significantly (P0.01) elevated after 6 months treatment. Also, triglycerides increased significantly (P0.01). In the
LYN
group, no significant alterations in the lipid parameters occurred. In the IUD group, cholesterol levels decreased significantly (P0.05) and a significant (P0.01) rise in HDL-cholesterol/cholesterol ratio was seen. Triglyceride levels did not change. The results obtained suggest that the low-dose EE+LYN and
LYN
OCs do not induce such changes in serum lipids that could be related to the risk of development of atherosclerosis. The decrease in cholesterol levels during IUD use remains to be confirmed by other further studies.

Inductively coupled plasma quadrupole mass spectrometry (ICP-QMS) was used for the accurate determination of
copper
in coal fly ash samples in the presence of excess titanium, using the reaction of Cu(+) ions with NH(3) in the cell. The method eliminated the effect of polyatomic isobaric interferences at m/z 63 and 65 caused by the formation of (47)Ti(16)O(+), (49)Ti(16)O(+) and (47)Ti(18)O(+) on (63)Cu(+) and (65)Cu(+) by detecting Cu(+) as the product cluster ion Cu(NH(3))(2)(+). As the signal of (63)Cu(NH(3))(2)(+) overlapped with that of (97)Mo(+) which existed in the samples, (65)Cu(NH(3))(2)(+) was detected at m/z 99. The effect of the operating conditions of DRC system was studied in order to obtain the best signal to noise ratio for Cu(NH(3))(2)(+) at m/z 99. The formation of Cu(NH(3))(2)(+) was through the clustering reaction Cu(+)+2NH(3)-->Cu(NH(3))(2)(+) which resulted in the separation of analyte from the interfering oxide. The detection limit for Cu(NH(3))(2)(+) was 0.015 ng mL(-1) as Cu. The method was applied to the determination of

in NIST
SRM
1633a and 1633b coal fly ash reference materials. The precision between sample replicates was better than 2.0% and the analysis results were in good agreement with the certified values.

A previously developed microwave heated vapor-phase digestion method for biological samples was modified to enable digestion of difficult to digest organic samples. Organic samples containing ca. 100 mg of organic carbon were digested using volume calibrated quartz inserts inside second generation type medium pressure microwave vessels. As digestion reagents, 98% sulfuric acid, 70% nitric acid and 30% hydrogen peroxide were used. The accuracy of the method was tested with six certified reference materials. Elements Ca, Fe, K, Na, Mg, P and Zn were determined from NIST-

8433 corn bran. Elements Al, Fe, Cd, Cu, and Zn were determined from NRCC DOLT-2 dogfish liver. The element Cd was determined from IRMM-VDA Cd in polyethylene No. 001-004 reference materials. These elements were determined from digested samples by ICP-OES. The results were close or within certified limits. The modified method could digest nearly all the materials tested, including the above mentioned reference materials, 2-nitrobenzoic acid (2-NBA), 4-NBA and

copper
(II)diacetyl-bis(N(4)-methylthiosemicarbazone) ((64)Cu-ATSM). (64)Cu-ATSM localizes to hypoxic regions of tumors and has been used for administering a high local dose of radiation therapy after uptake by cells. 2-DG, a glucose analog, selectively accumulates in cancer cells and interferes with energy metabolism, resulting in cancer cell death. 2-DG has been shown to potentiate the cytotoxic effect of ionizing radiation and certain chemotherapeutic agents. We have tested the effect of 2-DG on tumor response when combined with (64)Cu-ATSM in a mouse breast tumor model using the highly aggressive mouse mammary carcinoma cell line

(II)pyruvaldehyde-bis(N(4)-methylthiosemicarbazone. When 2-DG was combined with a single dose of (64)Cu-ATSM (2 mCi), tumor growth was inhibited approximately 60% compared with untreated mice, and animals survived approximately 50% longer than untreated mice or animals treated with each agent alone (32 versus 20 days). The maximum effect on tumor growth and survival was observed when 2-DG was administered daily for the lifetime of the mouse. Our results indicate that 2-DG potentiates the effect of (64)Cu-ATSM on tumoricidal activity and animal survival. We hypothesize that 2-DG alters the metabolic state of the cell, leading to increased uptake of (64)Cu-ATSM by the tumor. This would result in a higher local dose of radiotherapy. The continued presence of 2-DG would then prevent the repair of damaged cells, leading to inhibition of tumor growth. Our data indicate that the strategy of combining tumor-specific cytotoxic agents that function by differing mechanisms can result in an effective, selective, tumor-specific cell death with minimal effect on the host.
