1,2-Diaminobenzene and its derivatives react with selenous acid in acidic solution to form the piazselenols, which can be extracted into toluene. Microgram amounts of selenium can be determined spectrophotometrically by measuring the absorbance of these piazselenols extracted into toluene. A more sensitive method, in which the piazselenols extracted into toluene are detected by electron-capture gas chromatography, has been developed. In order to find a more sensitive reagent, 13 piazselenols were synthesized. The retention behaviour and sensitivity in electron-capture detection gas chromatography and the distribution ratios between aqueous solution and toluene were studied for each piazselenol extracted into toluene. Of these piazselenols, 4,6-dibromopiazselenol, formed by the reaction of 1,2-diamino-3,5-dibromobenzene with selenous acid, was found to be best as regards sensitivity and distribution ratio. Under the optimal conditions for the formation and the extraction of the piazselenol, the practical detection limit was 1 ng. Selenium(VI) and total selenium in NBS Bovine Liver, SRM 1577, were determined successfully.

The halogenated pyrimidine analogue 5-bromo-2-deoxycytidine (BCdR) was infused into BALB/C mice bearing EMT-6 tumors via either the intravenous or intra-arterial route. Hepatic dehalogenation of the drug was blocked by 5-diazouracil (DAZU) in order to ascertain its relative importance in the degradation of intravenously administered analogues. Increased radiosensitization was noted with higher intravenous pyrimidine concentrations, but DAZU blockage of dehalogenation had little effect. These studies show that following intravenous infusion, enough BCdR apparently bypasses the hepatic vessels to permit tumor radiosensitization despite dilution of the drug by the systemic circulation.

The Mackay-Marg, pneumatonograph, TonAir, and EMT-20 tonometers were evaluated in the normal dog eye. Tonometric scale readings were compared to the manometric recordings from the anterior chamber. The Mackay-Marg tonometer evaluated in open and closed systems was the most reliable (goodness of fit [r2] = 0.96) with intraocular pressure up to 100 mm. Hg. The pneumatonograph also exhibited high reliability (r2 = 0.92) but tended to underestimate intraocular pressures above 40 to 50 mm. Hg. The TonAir and EMT-20 tonometers were the least satisfactory for the dog, with r2 of 0.77 and 0.66 to 0.81, respectively. The Mackay-Marg is the most satisfactory tonometer for the dog.

The centriole regions of prophase PTK2 cells were irradiated with a laser microbeam. Cells continued through mitosis normally. Ultrastructural analysis revealed either an absence of centrioles or severely damaged centrioles at the irradiated poles. Microtubules appeared to focus into pericentriolar cloud material.

To judge standard practice for managing arrhythmias, what an EMT should be able to do in the field, drugs of choice, the success of EMT training, and the quality of EMT performance, clinical algorithms were developed. Branching logic, forcing yes/no decisions and delineating actions for all contingencies helped formalize and systematize EMT management of urgent and emergency cases. The algorithm set was sent to 19 consultants for review of content, sequence, drug dosage, and drug usage. Blind defibrillation, defibrillation of an unconscious, pulseless adult without electrocardiographic verification of arrhythmia, allows early definitive treatment of cardiac arrest victims. Basic EMT-As have the ability to perform blind defibrillation in a prehospital setting, and place an esophageal obturator airway. When basic EMT-As are performing blind defibrillation, there should be a standard operating procedure involving diagnosis, defibrillation, CPR and re-evaluation.

Undergraduate and postgraduate training in sexually transmitted diseases and genitourinary medicine in the UK is described. The course for overseas postgraduate students is briefly described. It is noted that once funds are available it has been agreed to establish a professorial chair of venereology at the Middlesex Hospital Medical School. This will be a major advance and is the most desirable goal to aim at in the encouragement of education in STD for the next few years.

Electron microscopic visualization of molecular hybrids formed in situ is feasible at the present time. It can be accomplished by two alternative approaches. In one, the in situ hybridization is carried out on ultrathin sections of target embedded in glycol methacrylate. In the other, whole cells are used for hybridization and they are subsequently prepared for electron microscopy. The choice of the method to be adopted depends on the type of target tissue. When there is a choice, the second approach seems preferable. Some of the important technical steps in the hybridization procedure, such as DNA denaturation in ultrathin sections, have been discussed and attention has been drawn to practical problems that may arise during the preparatory steps. Our light microscope experiments demonstrate that preparations made after glutaraldehyde fixation have a lower hybridization efficiency than those fixed with 3 : 1 methanol-acetic acid. Attempts are therefore being made to explore the possibility of using methanol-acetic acid for electron microscope in situ hybridization. First results of straight-forward fixation show that the preservation of nuclear structure may be fairly satisfactory for the purpose. However, the cumultative effects of subsequent treatments in the procedure still remain to be examined. For electron microscope autoradiograph (EM ARG) of hybridized preparations, the most suitable emulsion at present appears to be Ilford L4. Various factors conductive to optimum resolution consistent with maximum efficiency in this emulsion have been pointed out. Practical problems that may arise in autoradiographs of hybridized preparations such as background and variation of grain density in adjacent sections have also been considered.

Ethidium bromide (10 mug/ml) and bromodeoxyuridine (25 mug/ml) were used to sensitize selective cell organelles to visible wavelengths of an argon ion laser (488 and 514 nanometers). Ethidium bromide was shown to be selective in sensitizing nucleoli, chromosomes, and the centriolar region of PTK2 cells to the laser microbeam. Similarly, BrDU sensitized chromosomes to the microbeam irradiation. The lesions produced on the chromosomes when either agent was used appeared as a phase paling of the irradiated segment. Nucleolar lesions also appeared as a phase paling, and the centriolar region alteration appeared either as a phase paling or a phase darkening.
