We have cloned a delayed rectifier type K channel from rat heart (RH1). RH1 was identical to the rat brain K channel BK2 and differed from recently cloned rat cardiac K channel

by one amino acid residue. Endothelin receptors(ETRs)-mediated modulation of RH1 current (IRH1) was studied using Xenopus oocyte expression system. Activation of two different subtypes of ETRs by endothelin-1 equally suppressed the amplitude of IRH1. Stimulation of phosphatidylinositol turnover will probably be responsible for the suppression.

We have cloned a cDNA (designated
RAK
) coding for a delayed-rectifier K current (IRAK) from adult rat heart atrium and expressed it in Xenopus oocytes.
RAK
differs from the cloned rat brain K current, BK2 [McKinnon, D. (1989) J. Biol. Chem. 264, 8230-8236], by one amino acid at residue 411.
RAK
expressed in oocytes compares closely to the intrinsic adult rat atrial delayed-rectifier current measured by using whole-cell recording of single isolated cells. Northern blot analysis confirmed the presence of the channel in adult rat atrium, and to a lesser extent, in rat ventricle. IRAK activates with time constants ranging from 58 ms at -20 mV to 6 ms at +60 mV and does not show significant inactivation over 800 ms. It is blocked by 4-aminopyridine greater than barium much greater than tetraethylammonium chloride, which is similar to the relative potencies of these blockers on the native delayed rectifier current. We conclude that the main delayed rectifier K current in adult rat atria is virtually identical to a neuronal delayed rectifier, BK2.

Nasopharyngeal carcinoma (NPC) provides a unique opportunity to evaluate distinctive epidemiologic features and a possible etiologic relationship with Epstein-Barr virus (EBV) in human malignancy. The lack of a uniformly accepted pathologic classification for NPC has limited the application of this data, although the World Health Organization (WHO) developed a classification that may solve this problem. Monoclonal keratin antibodies were used for staining of NPC for evaluation of its assistance in diagnosis and classification. In the present immunohistochemical study, monoclonal keratin antibodies, designated AE1, AE2, and AE3, and a polyclonal keratin antibody (
RAK
) were used for study of the presence of keratin in 121 cases of NPC obtained from China and the United States. AE1 monoclonal antibody, which recognizes keratin protein classes 56.5K, 50K, and 40K, was shown to be the most sensitive and specific for NPC tumor cells among the keratin antibodies studied. In addition, some different keratin expression patterns could be identified between different kinds of epithelium and different tumor groups, with possible relevance to the histogenesis of the histologic subtypes of NPC.

We previously identified multiple proteins structurally related to 3 alpha-hydroxysteroid dehydrogenase in rat liver, lung, kidney, and testis ((1991) Arch. Biochem. Biophys. 291, 258-262). We further used these monoclonal antibodies to screen several lambda gt11 cDNA libraries derived from male rat liver, lung, and kidney. Five additional unique cDNA clones were isolated and sequenced; the proteins encoded by these cDNAs were found to exhibit 37-62% amino acid sequence homology to rat liver 3 alpha-hydroxysteroid dehydrogenase. Because these encoded proteins belong to the aldo-keto reductase superfamily, we named these proteins RAKa to RAKf.

represents rat aldo-keto reductase, and RAKa is the previously described rat liver 3 alpha-HSD. Northern blot analysis and reverse transcription-polymerase chain reactions were performed to examine their expression in various tissues. Only RAKe, which resembles human aldehyde reductase, was ubiquitously expressed in liver, kidney, lung, and other tissues, while the remaining mRNAs were found to have a more tissue- and sex-specific distribution. Genomic blot analysis showed complex, yet distinctive, restriction band patterns when different cDNAs were used as probes, suggesting that these cDNA clones are products of different genes and more related gene(s) may exist.

Neurotransmitter receptors alter membrane excitability and synaptic efficacy by generating intracellular signals that ultimately change the properties of ion channels. Through expression studies in Xenopus oocytes and mammalian cells, we found that the G protein-coupled m1 muscarinic acetylcholine receptor potently suppresses a cloned delayed rectifier K+ channel through a pathway involving phospholipase C activation and direct tyrosine phosphorylation of the K+ channel. Furthermore, analysis of neuroblastoma cells revealed that a similar tyrosine kinase-dependent pathway links endogenous G protein-coupled receptors to suppression of the native
RAK
channel. These results suggest a novel mechanism by which neurotransmitters and hormones may regulate a specific type of K+ channel that is widely expressed in the mammalian brain and heart.

A cDNA (CSMK1) encoding a delayed rectifier K+ channel of the Kv1.2 class was cloned from canine colonic circular smooth muscle and expressed in Xenopus oocytes. These channels appear to be uniquely expressed in gastrointestinal muscles and may participate in the electrical slow wave activity. Functional expression of CSMK1 in Xenopus oocytes demonstrated a K+ current that activated in a voltage-dependent manner upon depolarization. This current was highly sensitive to 4-aminopyridine (IC50, 74 microM). A low-conductance K+ channel was identified in inside-out patches from oocytes injected with CSMK1. This channel displayed a linear current-voltage relation with a slope conductance of 14 pS. The channels were blocked in a concentration-dependent manner by 4-aminopyridine. Northern blot analysis demonstrated that CSMK1 is expressed in a wide variety of gastrointestinal smooth muscles. Portal vein, renal artery, and uterus do not express CSMK1, suggesting that, among smooth muscles, expression of this K+ channel may be restricted to gastrointestinal smooth muscles. Breast and gynecological cancer-associated antigens

p120, p42 and p25 exhibit molecular, immunological and genetic homology to HIV-1 proteins. Normal tissues, including the majority of tissues adjacent to cancer, do not express these unique cancer markers. Antigens

are now detected in 100% of prostate cancer and in the majority of prostate benign hyperplasia (BPH) cases. Polymerase chain reaction (PCR) with HIV-1 gp41-derived primers revealed prostate cancer-associated DNA fragments of similar size (140 bp) as in HIV-1 genome. Ninety-five percent of BPH samples obtained from prostate cancer patients tested PCR-positive. For comparison, only 61.9% of BPH samples obtained from cancer-free patients tested PCR-positive. The DNA fragments amplified in prostate cancer and in BPH showed more than 90% homology to the HIV-1 gene for gp41. The obtained results strongly suggest that a retrovirus related to HIV-1 may be associated with cancers of the reproductive system.

antigens has been found to be inhibited by a peptide derived from variable loop V3 of HIV-1. Breast cancer DNAs of 40 patients were PCR amplified with HIV-1 gp41-derived primers, and all of the samples were found to be positive. The DNA fragments amplified in seven blindly selected breast cancer samples were sequenced. The breast cancer DNA sequences showed at least 90% homology to the HIV-1 gene for gp41. Antisense oligonucleotides complementary to the HIV-1-like sequences inhibited reverse transcriptase activity and inhibited the growth of breast cancer cells in vitro. Viral particles detected in breast cancer cell lines were strongly immunogold labeled with the anti-HIV-1 gp120 MAb. The results obtained strongly suggest that the long-postulated breast cancer virus may, in fact, be related to HIV-1.
