The value of rose bengal plate test (RBPT) in diagnosing brucellosis in cattle was determined by statistical comparison of its results with the results of the tests used in Poland, i.e. SAT, CFT, AGT and
MET
. RBPT was made in 2 variants. In routine, highly specific test, equal parts of antigen and serum--0.03 cm3, were used whereas in the experimental one the sensitivity was increased using half the amount of antigen--0.015 cm3 (RBPT0.015). Two batches of cattle serum were examined. In group I 249 cattle serums from the herds infected with brucellosis were examined. In group 2 there were 1269 cattle serums from the herds free of brucellosis, positive in SAT but negative in CFT. The reactions in SAT were considered as not specific if the reaction in the additional examination in CFT, AGT and
MET
was negative. On the basis of the results in group I, mainly the sensitivity of RBPT was determined compared with the total evaluation of the results of SAT/CFT. In RBPT0.015 the consistency of the results was 99.4% but in RBPT0.03 only 87.9%. Detectability of reactions, i.e. the percentage of positive results in infected herds was calculated. The results were as follows: AGT--89.6%, RBPT0.015--74.3%, SAT/CFT--66.3%, CFT--65.9%, RBPT0.03--59.8%, SAT--55.4%. In the group 2 mainly specificity of RBPT in relation to CFT, AGT and
MET
was determined. In RBPT0.03 it achieved 97.1% whereas in RBPT0.015--83.1%. The application of RBPT0.03 in the group 2 eliminated 95.8% of the not specific reactions in SAT and that of RBPT0.015--77.9%, respectively. The author suggests to use RBPT0.03 as a screening method instead of SAT and CFT to diagnose cattle brucellosis in the areas free from the disease. On the other hand, RBPT0.015, as more sensitive test is suggested to be used in the herds suspected of brucellosis to identify quickly infected animals.

Serological activity of swine IgM and IgG against Brucella abortus in RBPT was determined in relation to four other reactions used in Poland for diagnosing brucellosis standard agglutination test, complement fixation test, antiglobulin test, 2-mercaptoethanol test). Isolation of IgG was performed by the method of filtration on Sephadex gel G-200 of swine sera raised against Brucella abortus S19 by double immunization with suspension of killed bacteria. The presence of a certain Ig class in the fractions thus obtained was confirmed by immunoelectrophoresis and immunodiffusion tests. RBPT revealed the reaction of antibodies of IgM and IgG class which proves usability of this reaction diagnosis both early (IgM) and chronic (IgG) infection with brucellosis. Both classes of antibodies mentioned above were active also in SAT and CTT. Also the results obtained in AGT and
MET
were found interesting. In one of the sera, the absence of incomplete antibodies was observed, whereas positive reaction in antiglobulin test was found in its fractions containing IgG. This phenomenon was determined as concealment of incomplete agglutinins through higher level of complete antibodies in normal serum. In swine (the results were different from those obtained for cattle), apart from incomplete antibodies in IgG class, the presence of these agglutinins in IgM class was noted. On the other hand, the results obtained in
MET
proved that IgM antibodies of swine were not totally reduced when affected by 2-mercaptoethanol.

Intrapartal cardiotocographic disturbances of fetal heart rate patterns, the way of delivery and the neonate state have been analyzed in the group of 61 women in labor with pregnancy complicated by
EPH
gestosis. The results have been compared with pre-labor values of amniotic fluid index (AFI) in that group. In the group of 30 women in labor with AFI < or = 5 cm the analysis revealed the higher prevalence of non stress test reactivity losses, severe variable decelerations, late decelerations, cesarean sections and lower birth weight in comparison with the group of 31 women in labor with AFI > 5 cm. The study revealed that pre-labor AFI value estimation is usefull in predicting possibilities of fetal compromise during labor in women with pregnancies complicated by
EPH
gestosis.

The aim of our study was to assess the efficacy of labor induction among patients affected by
EPH
gestosis after intracervical extra-amniotic insertion of 15 mg PGF2 alpha in methyl-cellulose gel. We concluded that PGF2 alpha when given intracervically in methyl-cellulose gel is a highly effective inducer of cervical ripening process as well as subsequent uterine contractions, too. It should be stressed that in no case did we find blood pressure increase after such therapy.

It is suggested that ventricular arrhythmias may be the reason of sudden death in stable patients with COPD. Strenuous effort might provoke danger ventricular arrhythmias in those patients. The impact of maximal exercise test on cardiac ventricular arrhythmias was studied in 24 patients with advanced chronic obstructive pulmonary disease (COPD). The treadmill, self limited exercise test according to Bruce protocol and accompanied by pulse oximetry was performed during 24 hour Holter monitoring. We found ventricular extrasystoles in all patients, in an average 492 +/- 770, complex ventricular extrasystoles in 17 patients and nonsustained ventricular tachycardia in 8 patients. 81% of all ventricular extrasystoles, 80% complex ventricular extrasystoles and 70% nonsustained ventricular tachycardia occurred during the test and up to 2 hour after the test. Patients with complex ventricular arrhythmias were younger and mostly achieved more
MET
during exercise, but they did not differ in advancing of COPD as measured by spirometry, gas measurements and echocardiography. Although maximal exercise test enhances ventricular arrhythmias it produced no sustained, symptomatic ventricular arrhythmias in patients with advanced and stable COPD.

Selected immunological parameters of peripheral blood leukocytes in 30 tb contracts and control group consisting of 30 healthy blood donors in similar age were examined. All contacts showed the exudate type Mantoux reaction (over 20 mm in diameter) and all revealed normal chest X-ray. No differences between both groups in total T cells, CD4, CD8 counts, CD4/CD8 ratio and in proliferative response to mitogens (PHA and Con A) were found. Tb contacts had the similar serum IgM and elevated IgG and IgA concentrations as compared with the controls. Tb contacts showed depressed granulocyte metabolic activity as measured by CL response to chemostatic peptide N-FORMYL-
MET
-LEU-PHE (FMLP) in comparison with control subjects.

The complement levels were determined in 54 parturient women with
EPH
gestosis, in 32 normal parturient women and in 24 normal non pregnant women. Complement components: C4, C3c, factor B /Bf/ and C1 inactivator /C1 INA/ were measured by radial immunodiffusion. Peripheral and retroplacental blood were examined. Concentrations of C4, C3c, Bf in peripheral blood of both groups of parturient women were significantly higher than in normal non pregnant women. Peripheral blood of parturient women with
EPH
gestosis contained significantly higher concentrations of C3c than in normal parturient women. Complement Bf levels of normal parturient women were lower in the retroplacental blood than in the peripheral. The levels of C4 were significantly decreased and C3c increased in the retroplacental blood of parturient women with
EPH
gestosis when results were compared with retroplacental blood of normal parturient women. Results of present study suggest that complement activation could contribute to the pathogenesis of
EPH
gestosis.

Basic mechanisms of transcription initiation are conserved from yeast to man. However, in contrast to genes transcribed by RNA polymerases II and III, ribosomal gene transcription by RNA polymerase I (
Pol
I) is species-specific. Promoter selectivity is mediated by SL1/
TIF
-IB, a multiprotein complex containing the TATA-binding protein (TBP) and TBP-associated factors (TAFs) which bind to DNA and nucleate the assembly of initiation complexes. Using a human cell line that expresses epitope-tagged yeast TBP, we have isolated a chimeric complex consisting of yeast TBP and human TAFs which faithfully promotes human rDNA transcription in vitro. This result argues that specific interactions between TBP and
Pol
I-specific TAFs have been evolutionarily conserved between distant species. In addition, this finding also underscores the importance of TAFs in determining promoter selectivity of

I.

We have characterized a transcription factor from Ehrlich ascites cells that is required for ribosomal gene transcription by RNA polymerase I (
Pol
I). This factor, termed
TIF
-IC, has a native molecular mass of 65 kDa, associates with
Pol
I, and is required both for the assembly of Sarkosyl-resistant initiation complexes and for the formation of the first internucleotide bonds. In addition to its function in transcription initiation,
TIF
-IC also plays a role in elongation of nascent RNA chains. At suboptimal levels of
TIF
-IC, transcripts with heterogeneous 3' ends are formed which are chased into full-length transcripts by the addition of more
TIF
-IC. Moreover, on a tailed template, which allows initiation in the absence of auxiliary factors,
TIF
-IC was found to stimulate the overall rate of transcription elongation and suppress pausing of
Pol
I. Thus
TIF
-IC appears to serve a function similar to the
Pol
II-specific factor TFIIF which is required for
Pol
II transcription initiation and elongation.

The aim of this study was to evaluate the presence of the mRNA for the hematopoietic STK-1 receptor in various human tumor cell lines using RT-PCR directed analysis of gene expression. We have found
STK
-1R mRNA to be expressed in non-hematopoietic malignant cells of ectodermal, endodermal and mesodermal origin confirms a hypothesis that
STK
-1R is similar to other receptors with intrinsic tyrosine kinase activity, is not for specific hematopoiesis and is also expressed in non-hematopoietic cells. The fact that
STK
-1R is detectable in different tumor cell lines should also inspire further studies to evaluate its role in the transformation and growth of malignant cells.
