The SRT,
DRT
, and MT of older men (OA) who have experienced a life style of chronic physical activity were compared to those of nonactive men of similar age (ONA), and also to active (YA) and nonactive young men (YNA). Although activity level and age were significant factors, most of the activity level-by-age interaction in all but
DRT
was attributed to the slower performance of the ONAs. At least in this study, a life style of physical activity appeared to play a more dominant role in determining SRT,
DRT
, and MT than age. The hypothesis that most of the slowing of responses in the aged is attributable to CNS processing rather than MT decrements is repudiated, since MT results paralleled those of SRT and
DRT
.

It has been reported that 6-aminomethyl-3-methyl-4H,1,2,4-benzothiadiazine-1, 1-dioxide (AMBD, TAG) is a specific blocker of taurine and beta-alanine responses in the central nervous system. We have re-examined the effect of AMBD on amino acid and synaptically evoked responses recorded from isolated hemisected frog spinal cords by means of the sucrose gap technique. When indirect responses were blocked by adding tetrodotoxin (0.2 microM) or manganese chloride (2 mM) to the normal Ringer solution, AMBD (0.01-0.5 mM) selectively antagonized taurine, beta-alanine, hypotaurine and kojic amine evoked depolarizations of primary afferents at their intramedullary part (dorsal root terminals,
DRT
) and on dorsal root ganglia (DRG), without significantly affecting responses to glutamate (on
DRT
), glycine (on
DRT
) or GABA (on
DRT
and DRG). Depolarizing responses to taurine and beta-alanine (1 mM) were depressed by up to 50% with 0.1 mM AMBD and often completely antagonized with 0.25 mM AMBD. In normal Ringer solution, AMBD selectively antagonized the dorsal root potential evoked by ventral root stimulation (VR-DRP, threshold at 0.02 mM AMBD, 90% block with 0.25 mM); other synaptic potentials increased in duration and/or amplitude, demonstrating a strong convulsant effect of AMBD. Thus, the depolarizing responses of taurine, beta-alanine and hypotaurine on primary afferents are pharmacologically indistinguishable from the VR-DRP. These results are in agreement with the proposal that taurine or a taurine-like substance (possibly beta-alanine or hypotaurine) is the mediator of VR-DRP in amphibian spinal cord.

One-hundred-eleven cases of histopathologically atypical or malignant lipomatous lesions in the somatic soft tissue and retroperitoneum were studied. These consisted of 48 differentiated fatty neoplasms of the somatic soft tissues (DFT-S), 21 fatty neoplasms of the retroperitoneum (DFT-R), 33 myxoid liposarcomas from various sites and nine pleomorphic liposarcomas. DFT-S were defined as lipomatous lesions composed of mature fat and containing atypical stromal cells or lipoblasts. In the somatic soft tissues, this group included lesions that would be classified using published criteria as "atypical lipoma", "pleomorphic lipoma", "well-differentiated lipoma-like liposarcoma", and "sclerosing liposarcoma". All of the DFT-R met previously published criteria for "well differentiated liposarcoma" or "sclerosing liposarcoma". We found no consistent histologic differences between the DFT-S and
DRT
-R. No pure "round cell" liposarcomas were encountered although many myxoid liposarcomas had "round cell" areas. Follow-up data were available in 80 cases (72%) with a mean follow-up period of over 7 years. Among the DFT-S there were no uncontrollable recurrences, distant metastases, or tumor-related deaths. The depth of the neoplasm correlated with the tendency for local recurrence; no neoplasms primary in the subcutis recurred; 29% of the tumors recurred when they originated in the deep soft tissues or within the muscle. None of the recurrent tumors demonstrated "dedifferentiation." DFT-R had a recurrence rate of 67% and, although there were no distant metastases, nine patients (43%) died of tumor. Five retroperitoneal tumors dedifferentiated but did not metastasize. In light of this experience, we believe that the term "atypical lipoma" is warranted for the DFT-S and "well differentiated liposarcoma" is an appropriate label for the DFT-R. The overall mean survival for the 52 cases of liposarcoma (excluding DFT-S) was 13.6 years. The mean survival in "well differentiated liposarcoma" (11.25 years) was between that for myxoid liposarcoma (16.25 years) and that for pleomorphic liposarcoma (7 years). Six patients (29%) with myxoid liposarcoma developed local recurrences and 6 patients (29%) developed distant metastases and died. Metastasis was always associated with a round cell (or pleomorphic) component with increased numbers of mitotic figures in either the primary tumor or a local recurrence.

Two experiments were carried out measuring
DRT
. Two stimuli, horizontal-vertical and oblique ones were used, and four subjects were required to discriminate them in two categories as quickly as possible. In Experiment 1, the stimulus was presented in peripheral retina, every subjects showed a marked oblique effect at the outset. This effect decreased with proceeding sessions. After six sessions (600 trials), the results of these subjects were compared with those of naive control subjects in direct vision, in Experiment 2. The Experimental Group showed only a little oblique effect as contrasted with the Control Group. Such practice effect and the transfer effect couldn't be attributed to the functional anisotropy of the orientation analyzers in the visual cortex, and consequently, a cognitive approach was suggested.

This study analyzed auditory reaction time (RT) data from 1,265 community-dwelling volunteers (833 males and 432 females) who ranged in age from 17 to 96. Cross-sectional analyses revealed slowing of simple (SRT) and relatively greater slowing of disjunctive (
DRT
; aka "go-no-go") reaction time across decades for both males and females. Repeated testing within participants (longitudinal analyses) over eight years showed consistent slowing and increased variability with age. Males were faster than females cross age groups, RT tasks, and visits. Beginning at about age 20, RTs increased at a rate of approximately 0.5 msec/yr for SRT and 1.6 msec/yr for
DRT
. Errors also increased, making unlikely a tradeoff of accuracy for faster responses. The findings are consistent with the hypotheses that slowing of behavior is: (a) a continuous process over the adult life span; (b) characterized by age-associated increases in within-participant variability; (c) a direct function of task complexity and, presumably, the degree of mediation by higher regions in the central nervous system; and (d) greater in women than men.

An interest to the study of superoxide dismutase (SOD) is growing as it has become known that this enzyme may be used as a medical preparation and in the biochemical research. The aim of the work presented is to investigate the influence of temperature and UV radiation on the structural-functional properties of SOD. Copper- and zinc-containing SOD from bovine erythrocytes with the light of a
DRT
-400 lamp (dose of irradiation 1.5 x 10(3) J/m2) through an UFS-1 light filter (240-390 nm). The influence of temperature (20-85 degrees C) on the functional properties of native and UV modified SOD has been studied. The process of thermodenaturation of the enzyme proceeds at least two consecutive stages. SOD is stable within the temperature interval 20-70 degrees C. Kinetics of thermodenaturation of protein has been studied. It was established that UV light increases enzyme thermostability. Changes found in the kinetic characteristics of the UV-irradiated SOD are apparently due to its conformational rearrangements.

By screening a human fetal brain cDNA expression library using a monoclonal antiphosphotyrosine antibody and by 5' RACE procedures, we have isolated overlapping cDNAs encoding a receptor-type tyrosine kinase belonging to the
EPH
family,
DRT
(Developmentally Regulated
EPH
-related Tyrosine kinase gene). The
DRT
gene is expressed in three different size transcripts (i.e. 4, 5 and 11 kb).
DRT
transcripts are expressed in human brain and several other tissues, including heart, lung, kidney, placenta, pancreas, liver and skeletal muscle, but the 11 kb
DRT
transcript is preferentially expressed in fetal brain. Steady-state levels of
DRT
mRNA in several tissues, including brain, heart, lung and kidney, are greater in the midterm fetus than those in the adult.
DRT
transcripts are detectable at low levels in a human teratocarcinoma cell line (NTera-2), but its expression is greatly increased after the NTera-2 cells are induced to become postmitotic neurons (NTera-2N) by retinoic acid treatment. These data suggest that
DRT
plays a part in human neurogenesis. A large number of tumor cell lines derived from neuroectoderm express
DRT
transcripts, including 12 neuroblastomas, two medulloblastomas, one primitive neuroectodermal tumor and six small cell lung carcinomas (SCLC). Interestingly, several neuroblastoma cell lines with 1p deletion and one SCLC cell line express
DRT
transcripts of aberrant size (i.e. 3, 6 and 8 kb) in addition to those found in normal tissues. We mapped the
DRT
gene to human chromosome 1p35-1p36.1 by PCR screening of human-rodent somatic cell hybrid panels and by fluorescence in situ hybridization. As the distal end of chromosome 1p is often deleted in neuroblastomas and altered in some cases in SCLCs, these chromosomal abnormalities may have resulted in the generation of aberrant size transcripts. Thus, the
DRT
gene may play a part in neuroblastoma and SCLC tumorigenesis.

The excitation parameters of a multiband excitation model for speech production consist of a pitch period and a series of voiced/unvoiced (v/uv) decisions for a number of fixed frequency bands around the harmonics of the fundamental. In this paper, it is shown that, at most, four v/uv variable-length nonoverlapping frequency bands spanning over the telephone bandwidth are adequate to represent the excitation spectrum that provides an improved modeling of mixed voicing and noisy speech. A quad-band excitation (QBE) method is presented which generates excitation source for a linear prediction vocoder for low bit rate transmission of speech signal and the
DRT
scores validate the potential of the method.

What was known in the past as "obsessive psychoneurosis" was considered to be rare, chronic and difficult to treat. New definitions proposed by APA diagnostic systems (DSM III, DSM III-R, DSM IV) and the WHO (ICD-10) have opened up the possibility of epidemiologic studies in the population as a whole and among patients consulting psychiatrists and general practitioners. All these studies reveal a much higher prevalence, the incidence increasing from 0.05% (former estimate) to about 2.5% of the general population (American ECA survey). This implies new clinical and etiopathogenic concepts of this pathology. Experience of so-called antidepressant drugs, with an essentially serotoninergic action, bringing about a notable improvement in obsessive disorders, has greatly contributed to these changes. COD and COS are very often interlinked with other psychiatric symptomatology, this frequent co-morbidity raising the question of the autonomy of certain syndromes, as well as that of the borders between the normal and pathological. The TOC of clinicians are not the same as the TOC of epidemiologists. Another problem is that of the role played by these obsessive disorders in what has been referred to as "general neurotic syndrome". What is the relationship between mood disorders and anxious pathology as a whole, insofar as the same drugs are indicated in these various disease patterns. We report here the results of the
DRT
survey involving patients presenting for the first time to psychiatrists in office practice and concerning 731 COD and COS, i.e. 17 % of 4 364 adult out-patients (6.5 % definite COD, 2.7 % probable COD and 7.5 % COS).

Eph-related receptor tyrosine kinases have been implicated in the control of axonal navigation and fasciculation. To investigate the biochemical mechanisms underlying such functions, we have expressed the
EphB2
receptor (formerly
Nuk
/
Cek5
/
Sek3
) in neuronal NG108-15 cells, and have observed the tyrosine phosphorylation of multiple cellular proteins upon activation of
EphB2
by its ligand, ephrin-B1 (formerly Elk-L/Lerk2). The activated
EphB2
receptor induced the tyrosine phosphorylation of a 62-64 kDa protein (p62[dok]), which in turn formed a complex with the Ras GTPase-activating protein (RasGAP) and SH2/SH3 domain adaptor protein Nck. RasGAP also bound through its SH2 domains to tyrosine-phosphorylated
EphB2
in vitro, and complexed with activated
EphB2
in vivo. We have localized an in vitro RasGAP-binding site to conserved tyrosine residues Y604 and Y610 in the juxtamembrane region of
EphB2
, and demonstrated that substitution of these amino acids abolishes ephrin-B1-induced signalling events in
EphB2
-expressing NG108-15 cells. These tyrosine residues are followed by proline at the + 3 position, consistent with the binding specificity of RasGAP SH2 domains determined using a degenerate phosphopeptide library. These results identify an
EphB2
-activated signalling cascade involving proteins that potentially play a role in axonal guidance and control of cytoskeletal architecture.
