The potential therapeutic effects of differentiating agents on leukemic and solid tumor cells are being evaluated worldwide. These effects can be followed by morphologic as well as biochemical parameters. The enzymatic profile of four enzymes and the level of carcinoembryonic antigen were studied in 24 human colorectal carcinoma specimens and their adjacent uninvolved mucosa. The enzymes studied were
thymidine kinase
and
6-phosphogluconate dehydrogenase
as markers of proliferation, and alkaline phosphatase and gamma-glutamyl transpeptidase as markers of differentiation. A consistent finding was a marked increase in the activities of
thymidine kinase
and
6-phosphogluconate dehydrogenase
in the tumor cells as compared with the adjacent normal mucosa. The activity of gamma-glutamyl transpeptidase was not significantly different between tumor and uninvolved colon tissue. Alkaline phosphatase activity was markedly reduced in the tumor specimens. A relationship between the degree of differentiation and the degree of penetration and CEA expression was demonstrated in the tumor specimens as well as in their surrounding uninvolved mucosa.

PEG-mediated fusion between mouse Cl1d cells and primary Chinese hamster spleen cells produced interspecific hybrids which slowly and nonrandomly segregated Chinese hamster chromosomes. Cytogenetic and isozyme analysis (31 loci) of HAT and BrdU selected hybrid clones and subclones and of members of a hybrid clone panel retaining different combinations of Chinese hamster chromosomes enabled provisional assignment of the following enzyme loci on Chinese hamster chromosomes:

6-phosphogluconate dehydrogenase
, and enolase 1 to chromosome 2 were confirmed, and a chromosome 2 deletion (q23-q33) enabled the provisional assignment of PGM1 to that region. The assignments provide markers for the study of the genetic consequences of chromosomal rearrangements in Chinese hamster cell lines and support the concept of conservation of mammalian autosomal linkage groups.

We selected the common shrew (Sorex araneus) to generate the first insectivore gene map. Shrew-Chinese hamster and shrew- mouse somatic cell hybrid cells were constructed. When the 119 shrew-rodent clones were characterized, only shrew chromosomes were found to have segregated. A panel of hybrid clones was selected for gene assignment. The genes for hypoxanthine phosphoribosyl transferase (HPRT), glucose-6- phosphate dehydrogenase (G6PD), and malate dehydrogenase 1 (MDH1) were assigned to shrew Chromosome (Chr) de [which is the product of a tandem fusion between the 'original' mammalian X Chromosome (Chr) and an autosome], the gene for adenosine deaminase (ADA) and
6-phosphogluconate dehydrogenase
