Dietary fibers may tend to enhance or inhibit chemically induced experimental colon cancer, depending on the particular fiber consumed. This study examined the relationship between colonic thymidine kinase enzyme activity and mucin histochemistry and the reported effects of various dietary fibers on chemically induced colon carcinogenesis. Fiber-supplemented diets containing fibers reported to inhibit (wheat bran) or enhance (guar gum, carrageenan) chemically induced colon carcinogenesis in the rat were selected. Four groups of male Fischer 344 rats consumed 10% wheat bran, 5% guar gum, 5% carrageenan, or fiber-free diets ad libitum for 4 weeks. At the completion of the treatment period, the distal 12 cm of colonic mucosa was scraped off and homogenized for determination of thymidine kinase activity, and a 0.5-cm section of midcolon was processed by the high-iron diamine/Alcian blue method for mucin histochemistry. Final animal weights did not differ significantly among groups. Thymidine kinase enzyme specific activity (mumole thymidine phosphate formed x 10(6)/min/mg protein, means +/- SEMs) was not significantly different in the fiber-free, wheat bran, and guar gum groups (10.98 +/- 1.50, 7.41 +/- 1.09, and 9.11 +/- 2.04, respectively) but was markedly elevated at 41.84 +/- 4.65 in the carrageenan group (alpha less than 0.001). Mucin histochemistry failed to reveal any significant differences among dietary groups.

Oncofetal markers for colon carcinomas are CSAp, a nonsulfated mucin, a second trimester fetal antigen, an altered thymidine kinase, a monosialoganglioside, and glycolipid antigens. For gastric carcinoma, they are basic fetoprotein, a sulfoglycoprotein, and for pancreatic carcinomas--POA, an oncofetal pancreatic antigen, and designated as CAPI, an oncofetal antigen. Tumor-associated markers for colon carcinomas are: UDP-galactosyltransferase and zinc glycinate marker; for gastric carcinomas, sulfated glycoprotein and for pancreatic carcinomas, pancreas carcinoma-associated antigen, a polycytidylic acid-specific ribonuclease, and galactosyltransferase. Suggested as tumor-specific markers for colon carcinomas are an altered mucoprotein, basic antigen, beta 2-microglobulin-associated antigen, and a specific adenosine deaminase; for gastric carcinomas, a specific protein, an antigen with 3-oxyanthranilic acid, and an antigen of unknown origin in gastric secretions; for pancreatic carcinomas, an antigen with molecular weight of 380,000 daltons and an antigen suggested by tumor immunity.

Because the MUC1 mucin is highly expressed in breast and other carcinomas, interest is focused on the MUC1 promoter, particularly in the context of the delivery of genes to carcinomas. Earlier in vitro studies showed that the region between -152 and -66 of the MUC1 promoter is required for transcriptional activity in MUC1-expressing cells. Experiments reported here showed that sequences -119/-62 within this region are able to modulate transcription of the heterologous constitutively active herpes simplex virus thymidine kinase promoter in a pattern consistent with MUC1 expression. Band-shift experiments showed that although several factors (including Sp1 and Sp3) bind to these sequences, the element important in directing this MUC1 pattern of expression was an Sp1 GC box at -97. The data also show that the positioning or phase of the GC box was crucial for directing expression. The importance of the Sp1 transcription factor was confirmed by demonstrating that overexpression of Sp1 in MUC1-nonexpressing cells increased, not only the expression of a reporter gene driven by the 1.4-kb MUC1 promoter, but also the expression of MUC1 from the endogenous gene. Together, these data define an important role for Sp1 in the cell type-specific transcription of MUC1.
