The activity of enzymes regulating the processes providing functional activity of leukocytes was studied in the exudate leukocytes of healthy rabbits and animals with
alloxan
diabetes. Rabbits with diabetes displayed a reduction of
hexokinase
, phosphoglucomutase, glucose-6-phosphate dehydrogenase and adenylate kinase activity. The activity of UDPH-pyrophosphorylase, UDPH-glycogentranspherase, 6-phosphogluconate dehydrogenase and glutathion reductase showed no significant changes in the exudate leukocytes in diabetes. A reduction of
hexokinase
and glucose-6-phosphate dehydrogenase limiting glycolysis and the pentose-phosphate cycle, respectively, providing energy for leukocytes and important in protein metabolism of these cells, is of great significance in the reduction of functional activity of leukocytes in the inflammatory focus in diabetes.

The effects of sodium orthovanadate (0.6 mg/ml in drinking water) on
hexokinase
isozymes, pyruvate kinase and malic enzyme in liver and kidney of control and
alloxan
diabetic rats were studied and compared. Vanadate treatment of diabetic rats normalized hyperglycemia and almost completely restored the differentially altered enzyme profile of liver (a tissue that underutilizes glucose in diabetes) and kidney (a tissue that overutilizes glucose during diabetes). Vanadate therapy, however, could not restore the depressed plasma insulin level of diabetic rats. The study clearly indicates that vanadate can effectively normalize many metabolic abnormalities even at a low insulin level in both insulin-dependent and -independent tissues of diabetic rats.

Measurements were made of the levels of metabolic intermediates and activities of enzymes of the glycolytic route, pentose phosphate pathway, and polyol pathway in livers and kidneys of NOD mice. A 34% decrease in UDP-glucose, a 40% decrease in glucose-6-phosphate (G6P) and fructose-6-phosphate, and a 75% decrease in fructose-2,6-bisphosphate (F2,6P) were found in the livers of NOD mice. The fall in the level of F2,6P (the important regulator of glycolysis) is accompanied by a 20% reduction in the activity of phosphofructokinase. These changes are in agreement with previously reported liver depletion of glycogen and reduced synthesis of proteins and nucleic acids in the diabetic state. In the kidney, the increase in
hexokinase
activity is consistent with increased levels of G6P and glycogen content of kidney in diabetes. The decreased level of phosphoribosyl pyrophosphate was reported to be a regulator of kidney growth in the initial period of diabetes but can still be found in NOD mice 6 wk after development of hyperglycemia. The reported changes are similar to those seen in
alloxan
- or streptozocin-induced diabetic animals, but certain changes are more marked in NOD mice, especially those directed to increase nucleic acid and protein synthesis in the diabetic kidney.

activity in cytoplasmic fractions of transplantable radiation-induced rat islet cell tumours, ob/ob mouse pancreatic islets, rat liver and rat kidney. Half maximal inhibitory concentrations of
alloxan
were greater than those previously found for half maximal inhibition of pancreatic islet or liver glucokinase. D-glucose, preferentially the alpha-anomer, and D-mannose protected
hexokinase
activity against
alloxan
inhibition. 1,4-Dithiothreitol completely protected against and partially reversed the
alloxan
inhibition of
hexokinase
. The ability of various dithiols to reverse the inhibition of
hexokinase
by
alloxan
was dependent on the spacing between the SH (thiol) groups. Only dithiols with intermediate spacing between the SH groups were effective. Dithiols with two vicinal SH groups such as 1,2-dimercaptoethane and 2,3-dimercaptopropanol (BAL) and dithiols with more widely spaced SH groups such as 1,5-dimercaptopentane were ineffective. Thus a reaction of
alloxan
with two SH groups in the sugar binding site of the
hexokinase
with the formation of a disulfide bond may be involved in the reversible inhibition of the enzyme. Ninhydrin also inhibited
hexokinase
from all four tissues studied. The half maximal inhibitory concentrations of ninhydrin were lower than those of
alloxan
. Inhibition of
hexokinase
may be an important factor in the general cytotoxic action of ninhydrin. However, inhibition of pancreatic islet
hexokinase
is unlikely to be the initial event in the pancreatic B-cell toxic action of
alloxan
, even if inhibition of
hexokinase
by high concentrations of
alloxan

diabetes induced in white rats by intraperitoneal injection of

-monohydrate (15 mg/100 g body weight) was used to study changes in the glycogen phosphorylase a and b, phosphoprotein phosphatases and
hexokinase
activities under insulin deficiency conditions. Among the enzymes studied, an increase in muscle phosphorylase a activity as well as the a/b ratio have been obtained. In diabetic muscle phosphoprotein phosphatases and
hexokinase
activities were diminished. AMP increased the liver glycogen phosphorylase activity twice in diabetic rats whereas in normal animals the enzyme was less sensitive to this effector. The changes in liver

-induced hyperglycemic mice and gave no influence on insulin binding to isolated adipocytes. Aconitan A exerted no effect on the activities of hepatic
hexokinase
, glucokinase, glucose-6-phosphatase and glucose-6-phosphate dehydrogenase, whereas it significantly increased hepatic phosphofructokinase activity. Although the activity of hepatic glycogen synthetase showed a tendency to increase, the activity of liver phosphorylase and glycogen content were unchanged by aconitan A. The effect of cataractogenesis on the behavior of some enzymes involved in glucose metabolism was examined histochemically both in human lenses and in rat lenses from rats with
alloxan
-induced diabetes. Several modifications in the currently available techniques were made in order to localize glucose-6-phosphate dehydrogenase, aldose reductase, sorbitol dehydrogenase,
hexokinase
and ketohexokinase in ocular lens. Human cataractous lenses showed a precipitous drop in glucose-6-phosphate dehydrogenase activity, whereas the lenticular tissues of
alloxan
-treated rats showed a gradual decrease of this enzyme with the prolongation of diabetes. Aldose reductase activity increased in hypermature and senile diabetic cataracts, whereas sorbitol dehydrogenase activity decreased in these lenses. Similarly, in
alloxan
-diabetic rat lenses the activity of aldose reductase increased while that of sorbitol dehydrogenase decreased with the prolongation of diabetes. Attempts were made to localize

Characterization of glucokinase in pancreatic B-cells from ob/ob mice and from rat liver revealed identical characteristics. A narrow substrate specificity; high Km values for the two substrates, D-glucose and D-mannose, in the range of 10 and 20 mmol/l, respectively; higher Vmax values for D-glucose than for D-mannose; inhibition of glucokinase activities by D-mannoheptulose and by a specific glucokinase antibody. These characteristics distinguish glucokinase in soluble cytoplasmic fractions of pancreatic B-cells and liver from low Km hexokinases.
Alloxan
is a pancreatic B-cell cytotoxic agent, which has been widely used as a tool for the elucidation of the mechanisms of insulin secretion, because its inhibitory action on insulin secretion has been presumed to be intimately related to the mechanism of glucose-induced insulin secretion.
Alloxan
inhibited glucokinase but not
hexokinase
activity in cytoplasmic fractions of pancreatic B-cells and liver. The half maximal inhibitory concentration of
alloxan
was 5 mumol/l. Glucokinase activity was protected from
alloxan
toxicity only by D-glucose and D-mannose; the alpha anomer of D-glucose provided significantly greater protection than the beta anomer. The non-metabolizable sugar 3-O-methyl-D-glucose did not provide protection of glucokinase activity against inhibition by
alloxan
. Thus, inhibition of pancreatic B-cell glucokinase may contribute to the inhibition of glucose-induced insulin secretion by
alloxan
. These results support the contention that glucokinase regulates the metabolic flux rate through the glycolytic chain in the pancreatic B-cell and thereby generates the signal for glucose-induced insulin secretion.

Activity of glucose-6-phosphate dehydrogenase in haematopoietic cells of bone marrow of rabbits was not affected during insular deficiency induced by starvation or
alloxan
diabetes as well as after intramuscular hydrocortisone administration. It is suggested that slight decrease in the activity of
hexokinase
detected during starvation and hydrocortisone administration might be accounted for by the presence of mature leukocytes in the population of isolated myelokaryocytes.
