The isoenzyme
hexokinase
(HK) spectrum from normal rat large intestinal mucosa consisted of 3 isoenzymes. In tumours of this localization induced by
1,2-dimethylhydrazine
there proved to be a lack or marked decrease in the most rapid anodic isoenzyme. Only one HK isoenzyme was found in the metastases. Km (glucose) for tumour HK was 2--3 times lower than for normal intestinal HK; the HK activity was detected in the serum from the 1st month of the carcinogenic administration, and by the 5th month it was found in 80% of the tumour-bearing animals. No serum HK activity was ever found in control rats.

The activity of 19 enzymes (
hexokinase
, glucoso-6-phosphatisomerase, alpha-glycerophosphate-, lactate-, succinate-, isocitrate-, malate-, glucoso-6-phosphate-, 6-phosphogluconate-, glutamate-, alcohol-, inosine-5'-phosphate-, guanosine-5'-monophosphate-dehydrogenase, cytochromoxidase NAD.N2- and NADP.N2-diaphorase, monoaminoxidase, alkaline and acid phosphatase) was studied comparatively in the mucosa of control rats and in tumors of the small intestine (27), and large intestine (176), induced in 41 rats percutaneously by
1,2-dimethylhydrazine
. A decreased level of the enzymes of tissue respiration and Krebs cycle was found with a simultaneous increase in the activity of the enzymes of glycolysis and pentoso-monophosphate shunt. These data evidence variations in tumor metabolism consisting in oxidizing phosphorylation, being replaced by aerobic glycosis, and also reflecting an intensive proliferation of tumor cells.

Glucokinase,

, fructose 1,6-bisphosphatase and phosphoenolpyruvate carboxykinase specific activities were monitored in liver cytosol from rats that had been made cancerous with
1,2-dimethylhydrazine
and then treated with hydrazine sulfate. The presence of intestinal cancer, specifically, was confirmed by laparotomy and by histological analysis. Sustained changes in
hexokinase
and glucokinase specific activities were first evident during the latter weeks that the carcinogen was being administered. Upon subsequent treatment with hydrazine sulfate, glucokinase activity further decreased, and liver cytosolic phosphoenolpyruvate carboxykinase activity increased. Liver cytosolic
hexokinase
and fructose 1,6-bisphosphatase specific activities were not appreciably affected by the hydrazine sulfate treatment. These results indicate that hydrazine sulfate may influence carbohydrate metabolism at the level of selected liver enzymes not only with respect to gluconeogenesis, but also in terms of glucose uptake.

Mouse renal cell tumors (RCTs) were induced in male CBA mice by 5 subcutaneous injections of 8 mg
1,2-dimethylhydrazine
(DMH)/kg body weight once a week. After a lag period of 2 yr kidneys were removed, and serial cryostat sections of the kidneys were histochemically analyzed for the following parameters: glycogen content, basophilia, and the activities of glycogen synthase (SYN), glycogen phosphorylase (PHO), glucose-6-phosphatase (G6Pase), glucose-6-phosphate dehydrogenase (G6PDH),
hexokinase
(HK), pyruvate kinase (PK), lactate dehydrogenase (LDH), malic enzyme (ME), succinate dehydrogenase (SDH), alkaline phosphatase (ALPase) and gamma-glutamyltranspeptidase (GGT). RCTs displayed the same histochemical profile irrespective of their size and growth pattern. In comparison with the normal kidney epithelium, the neoplastic cells exhibited elevated activities of enzymes for glycolysis (HK, PK, LDH) and the pentose phosphate pathway (G6PDH), while negative G6Pase and low SDH activity were observed in these cells. The majority of RCTs showed high PHO activity and weak staining for SYN. Activities of ALPase and GGT were negative in most of the RCTs. Markedly enlarged cells with atypical nuclei were detected in some advanced RCTs. Higher activities of glycolytic and mitochondrial enzymes and G6PDH were found in these enlarged cells than in other tumor cells. Tubular preneoplastic lesions were similar to neoplastic lesions in morphological and histochemical characteristics. The present study revealed that a markedly elevated capacity for glycolysis and the pentose phosphate pathway occurred in RCTs in mice. A similar histochemical pattern in the few preneoplastic tubular lesions observed suggests that these metabolic aberrations emerge early during carcinogenesis, but additional studies on early stages of renal carcinogenesis are needed to substantiate this assumption.
