1. The changes with the time of the activities of some energy-supplying enzymes and of the hydrolytic enzyme, acid phosphatase, were studied over 2 weeks of complete ischaemia, produced in the rat soleus muscle by section of the abdominal aorta and terminal devascularization, leaving nerve and tendon intact. 2. Activities of glycolytic enzymes, oxidative enzymes,
hexokinase
and acid phosphatase are affected in a different manner. Activities of the glycolytic enzymes, lactate dehydrogenase, triosephosphate dehydrogenase and glycerolphosphate dehydrogenase, are lowest on the 1st day and increase thereafter. The first two reach the control values again on the 4th and 14th day, respectively, while glycerolphosphate dehydrogenase reaches about 50% of the control value on the 14th day. The maximum decrease in activity of the oxidative enzymes, citrate synthase, beta-hydroxyacyl-CoA-dehydrogenase and malate dehydrogenase occurs later (4th day); thereafter their activity returns slowly to control values, but does not reach them even on the 14th day. Hexokinase activity is slightly decreased on the 1st day; then it increased and reached on the 7th day twice the control value. Thus on the 1st day the activity of the enzymes of aerobic metabolism prevail, and on the 4th day those of anaerobic carbohydrate (glucose) metabolism; the recovery of enzyme activity of aerobic oxidation occurs later. 3.
Acid phosphatase
activity increased from the 2nd day onwards, reaching up to 3 times the control value on the 4th day and still twice that value on the 14th day. This agrees well with the histochemical picture of acid phosphatase. 4. Histochemical changes of alkaline phosphatase activity reveal destruction of capillary endothelial cells during the first few days after operation and their later proliferation from the periphery, correlating with the loss and recovery of oxidative enzyme activity.

Several enzymes of amino acid and carbohydrate metabolism were studied in primary cultures of fetal rat hepatocytes. One day after plating, activity of both esterase and gamma-glutamyl transpeptidase decreased to half of the freshly isolated hepatocytes, and then remained constant.
Acid phosphatase
revealed lower activity after plating but recovered to the same levels of isolated cells on day-8. Leucine aminopeptidase and glutathione S-transferase showed a peak of activity respectively on day-6 and day-8. On the other hand, activities of glucose-6-phosphate dehydrogenase,
hexokinase
and pyruvate kinase increased linearly from Day-1, but only pyruvate kinase reached a plateau on Day-2. These results suggest that different patterns of enzyme activities in the culture might be a reflection, both of a release from homeostatis and adaptation to a new environment.

Alterations in the activities of some enzymes in a freshwater catfish, Heteropneustes fossilis, have been examined in liver, kidney, intestine, ovary, gills, and muscles after exposure to 0.26 mg/liter of cadmium for 15, 30, and 60 days. The fish were hyperglycemic and hyperlactemic after 15 and 30 days of exposure. The liver and muscle glycogen content was depleted in the first two periods of exposure. In contrast, 60 days of cadmium treatment increased the glycogen content of the two tissues. Liver lactic acid level was elevated after 15 days. Muscle lactic acid content fell significantly after 15 and 60 days of exposure, but it was elevated after 30 days.
Acid phosphatase
activity was inhibited in liver, ovary, and gills but the enzyme activity increased in kidney and intestine. The activity of alkaline phosphatase decreased in liver, kidney, and intestine but elevation was recorded in ovary and muscles. In all three exposure periods,
hexokinase
activity of kidney and ovary was inhibited but the enzyme activity increased in intestine. Hexokinase showed elevation in liver, gills, and muscle after 15 and 30 days of exposure and inhibition after 60 days of exposure. The activity of xanthine oxidase decreased in liver and muscles and elevated in the rest of the tissues. Glutamate dehydrogenase fell significantly in intestine, ovary, and gills. In liver, kidney, and muscles the enzyme activity was elevated. Liver, intestine, gills, and muscles showed elevation in aminoacid oxidase activity. However, the enzyme activity was inhibited in kidney and in ovary.
