Preliminary studies of 13 enzymes subserving various metabolic pathways were undertaken in tumor-free liver biopsy samples from cancer patients and control subjects. The observations indicate that as a result of nonhepatic neoplasms, with (7 cases) or without (6 cases) hepatic involvement, the biochemical composition of the liver becomes partially undifferentiated. Quantification of appropriate enzymes in histologically normal liver samples could thus distinguish clearly between cancer hosts and controls. The best discriminators include two hepatic enzymes whose concentrations were decreased to less than 30% of normal (soluble glutamate dehydrogenase and the cold stable pyrroline-5-carboxylate reductase) and three for which it was elevated two to four-fold (hexokinase, peptidyl proline hydroxylase and thymidine kinase) in response to distant neoplasms. The same alterations in hepatic enzyme pattern were not seen in any cancer-free patients with or without morphologic liver damage.

We examined effects of temperature acclimation on ultrastructural characteristics of cardiac myocytes and maximal activities of metabolic enzymes in cardiac tissue of striped bass (Morone saxatilis). Ventricular mass and ventricular mass divided by body weight were significantly increased (29% and 40%, respectively) in animals acclimated to cold (5 degrees C) vs. warm temperatures (25 degrees C). Mean myocyte diameter was increased at cold temperature (3.47 +/- 0.14 vs. 2.98 +/- 0.08 microns), which is sufficient to explain the increase in ventricular mass. Ventricular enlargement did not alter volume densities of mitochondria, myofibrils, protein concentration, or citrate synthase activity. Thus total volume of mitochondria and myofibrils increased proportionately with cardiac mass in cold animals. Activities of hexokinase (34%) and carnitine palmitoyltransferase (42%) increased in cold animals, suggesting positive compensation and increased aerobic capacity for utilization of glucose and fatty acids for energy production. Enlargement of the ventricle and an increased capacity for ATP production in striped bass may help compensate for kinetic constraints at cold temperatures and maintain circulatory support to oxidative axial musculature for swimming activity.

UDP-glucose pyrophosphorylase (UGPase) is a key enzyme producing UDP-glucose, which is involved in an array of metabolic pathways concerned with, among other functions, the synthesis of sucrose and cellulose. An Arabidopsis thaliana UGPase-encoding gene, Ugp, was profoundly up-regulated by feeding sucrose to the excised leaves and by an exposure of plants to low temperature (5 degrees C). The UGPase activity and its protein content also increased under conditions of sucrose feeding and exposure to cold. The sucrose effect on Ugp was apparently specific and was mimicked by exposure of dark-adapted leaves to light. Drought and O2 deficiency had some down-regulating effects on expression of Ugp. The sugar-signalling pathway for Ugp regulation was independent of hexokinase, as was found by using transgenic plants with increased and decreased expression of the corresponding gene. Subjecting mutants deficient in abscisic acid (ABA) to cold stress conditions had no effect on Ugp expression profiles. Okadaic acid was a powerful inhibitor of Ugp expression, whereas it up-regulated the gene encoding sucrose synthase (Sus1), indicating distinct transduction pathways in transmitting the sugar signal for the two genes in A. thaliana. We suggest that Ugp gene expression is mediated via a hexokinase-independent and ABA-insensitive pathway that involves an okadaic acid-responsive protein phosphatase. The data point towards Ugp as a possible regulatory entity that is closely involved in the homoeostatic readjustment of plant responses to environmental signals.

Hexokinase from larvae of the freeze-avoiding goldenrod gall moth, Epiblema scudderiana, was purified 20-fold using chromatography on DE52 Sephadex, phosphocellulose, and blue dextran. Final specific activity was 75.8 U/mg and SDS-PAGE gave a molecular weight of 94,000 for the monomer. Arrhenius plot showed a break at 16 degrees or 12 degrees C in the absence vs. presence of 10% v/v glycerol, indicating a conformational change in the enzyme at lower temperatures but suggesting a stabilizing effect of glycerol. Comparison of hexokinase kinetic properties at 22 degrees and 4 degrees C showed higher affinity for both glucose and ATP (Km values were 45-50% lower), as well as for the cofactor Mg(2+), at the lower temperature. Furthermore, product inhibition by glucose-6-phosphate and ADP was reduced at 4 degrees C. Glucose levels rise in E. scudderiana as an apparent by-product of high rates of glycogenolysis during glycerol synthesis. The temperature-dependent properties of hexokinase would facilitate the recycling of this glucose back into the pathway of glycerol synthesis and could help to achieve the near stoichiometric conversion of glycogen to glycerol that is seen during cold hardening. Some key enzymes of EMP pathway and TCA cycle in a psychrophilic yeast Y18 were studied in this paper compared with those of Saccharomyces cerevisiae. The results indicated that fructose, 1,6-bisphosphate aldolase, succinate dehydrogenase, and hexokinase in Y18 were very thermolabile and have high activity at low temperature. These enzymes belong to cold-active enzymes. Alpha-ketoglutarate dehydrogenase existed possibly in isoenzyme which had different temperature characteristics. Citrayl synthetase was very similar in temperature characteristics to that of mesophiles. The Km value of succinate dehydrogenase both from Y18 and S. cerevisiae were studied and Some features of enzyme in psychrophiles were also discussed in this paper.

We tested the hypothesis that the physiological strategy for acclimating to low body temperature is similar among closely related fish. Largemouth bass (Micropterus salmoides), green sunfish (Lepomis cyanellus), bluegill sunfish (Lepomis macrochirus), black crappie (Pomonix nigromaculatus), and white crappie (Pomonix annularis), all members of the family Centrarchidae, were acclimated to 5 degrees and 25 degrees C. Morphometric variables (total mass, total length, organ masses) and enzyme activities (hexokinase; lactate dehydrogenase; and cytochrome oxidase in heart, liver, and muscle) were measured in 5 degrees C- and 25 degrees C-acclimated fish at 5 degrees and 25 degrees C assay temperatures. Each species displayed a distinct physiological response to cold acclimation that differed among tissues. These data suggest that the response to cold acclimation is highly variable within families. Our findings are consistent with other studies suggesting that acclimation responses are labile and may evolve independently even among closely related species.

Seasonal collections of the subtidal horse mussel, Modiolus modiolus, from a depth of 10 m were made at the Isles of Shoals, New Hampshire to assess changes in overall energetic demand, measured as respiration, the maximal activities of rate-limiting enzymes of intermediate metabolism, level of oxidative stress, and the expression of heat shock proteins (HSP). Weighted respiration rates of mussels from winter collections were significantly lower than summer rates but decreased by less than 20%. Specific activities of several rate-limiting enzymes were measured in mussels from the summer and winter collections at the temperature of collection and the reciprocal seasonal temperature (15 and 5 degrees C). Comparisons of these enzyme activities and the protein concentrations of hexokinase and citrate synthase show that a quantitative strategy is used to acclimatize to winter temperatures by these rate-limiting enzymes of intermediate metabolism. The activities and protein concentrations of the antioxidant enzyme, Cu/Zn superoxide dismutase (SOD) is seasonally indistinguishable while the concentration of HSP 70 was greater in winter than in summer samples. These results show that mussels seasonally compensate for decreases in temperature by increasing the concentration of rate-limiting metabolic enzymes while maintaining the same level of antioxidant protection in summer and winter consistent with high aerobic metabolism in both winter and summer. Lastly, the significantly greater concentrations of HSP70 in winter samples suggests that protein chaperone functions must be maintained while other seasonal adjustments to cold temperatures are occurring.

The present study was carried out to investigate mechanism of adaptogenic activity of seabuckthorn dry leaves aqueous lyophilized extract, administered in rats at a dose of 100 mg/kg body weight prior to cold (5 degrees C)-hypoxia (428 mmHg)-restraint (C-H-R) exposure up to fall of T(rec) 23 degrees C and recovery (T(rec) 37 degrees C) from C-H-R induced hypothermia. The effect of extract treatment was studied on key metabolic regulatory enzymes in blood, liver and muscle and tissue glycogen in rats on attaining T(rec) 23 degrees C and post stress recovery of T(rec) 37 degrees C. In control rats during C-H-R exposure on attaining T(rec) 23 degrees C there was significant decrease in enzyme activities of blood hexokinase (HK), citrate synthase (CS) and glucose-6-phosphate dehydrogenase (G-6-PD); liver CS; and in muscle glycogen, and CS and G-6-PD activities. In control rats on recovery of T(rec) 37 degrees C there was also a significant decrease in liver and muscle glycogen levels along with decreased enzyme activities of blood G-6-PD; liver CS; and liver and muscle G-6-PD. This suggested that during severe stressful exposure to C-H-R and post stress recovery the aerobic metabolism as well as hexose monophosphate (HMP) pathway is suppressed. The single and five doses extract treatment restricted the decrease or better maintained tissue glycogen and enzyme activities, viz. HK, phosphofructokinase (PFK), CS and G-6-PD, in blood, liver and muscle, during C-H-R exposure (T(rec) 23 degrees C) and recovery of T(rec) 37 degrees C. The results suggest that seabuckthorn extract treatment caused a trend for shifting anaerobic metabolism to aerobic during C-H-R exposure and post stress recovery.

Skeletal muscle hexokinase (HK) from Richardson's ground squirrels was analyzed to determine how the enzyme is regulated during hibernation, a state of cold torpor. The HK II isozyme dominated in muscle and ~15% of total HK was bound to the insoluble fraction. HK maximum activity was 33% lower in hibernator muscle and the enzyme showed a significantly higher K ( m ) ATP (by 80%) and a lower K ( i ) for glucose-6-P (by 40%) than euthermic HK (assayed at 22 degrees C). However, 5 degrees C assay significantly reduced K ( m ) glucose of hibernator HK. Stimulation of AMP-dependent protein kinase (AMPK) in hibernator extracts elevated the HK activity and reduced K ( m ) ATP, but did not affect euthermic HK. Stimulation of protein phosphatases significantly lowered the HK activity in both situations. AMPK-dependent phosphorylation was confirmed by immunopreciptiation of (32)P-labeled HK. DEAE-Sephadex ion exchange chromatography revealed two peaks of HK in hibernator muscle extracts (low and high phosphate forms), whereas only a single peak of phospho-HK was present in euthermic muscle. We conclude that differential control of muscle HK in euthermic versus hibernating states is derived from two main regulatory influences, reversible protein phosphorylation and temperature effects on kinetic properties.

Mechanism of rhodiola root extract adaptogenic activity was studied in rats. The extract was orally administered in rats (100mg/kg body weight), 30 min prior to cold (5 degrees C)-hypoxia (428 mmHg)-restraint (C-H-R) exposure up to fall of T(rec)23 degrees C and recovery (T(rec)37 degrees C) from hypothermia. In untreated control rats serum lactate and non-esterified fatty acids (NEFA) increased on attaining T(rec)23 degrees C with decreased blood enzyme activities hexokinase (HK), phosphofructokinase (PFK), citrate synthase (CS) and glucose-6-phosphate dehydrogenase (G-6-PD), on attaining T(rec)23 degrees C and T(rec)37 degrees C. Decreases were also observed in liver and muscle tissues HK and G-6-PD enzyme activities and liver glycogen and CS on attaining T(rec)23 degrees C and recovery; muscle PFK during recovery; muscle CS on attaining T(rec)23 degrees C. Single and five doses of extract administration restricted increase in serum lactate values of rats on attaining T(rec)23 degrees C and maintained blood NEFA in single dose extract treated animals, indicating improved utilization of NEFA as energy fuel. The single and five doses extract treatment decreased or better maintained tissue glycogen and enzyme activities, viz. HK, PFK, CS and G-6-PD, in blood, liver and muscle, on attaining T(rec)23 degrees C and recovery. The results suggest that rhodiola extract treatment in rats shifted anaerobic metabolism to aerobic, during C-H-R exposure and post stress recovery.
