The effects of two sequential therapies using mestranol as an estrogen and chlormadinone acetate and norethisterone acetate, respectively, as a gestagen on the aminotransferase (GOT and GPT), alkaline phosphatase and alpha-amylase activities, the cholesterol, total bilirubin and total protein contents and the indocyanine green (Ujoviridin, VEB Chemiekombinat Bitterfeld) half-life were studied in two groups, each comprising 12 women of fertile age. The serum protein was separated by paper electrophoresis and both the thymol turbidity and zinc sulphate tests were performed during the first treatment cycle. An estrogen-induced rise in alanine aminotransferase (GPT) activity from 6.31 U/liter to 12.14 U/liter was observed during application of the mestranol/chlormadinone acetate therapy. The value dropped to 5.34 U/liter when chlormadinone acetate was administered as a gestagen in addition to the estrogen. No significant changes were noted in the other biochemical parameters. Sequential therapy involving application of mestranol and norethisterone acetate produced only statistically secured rises in the alpha 1-globulin content and the GPT activity. All other changes in the different parameters remained within the normal range and were statistically insignificant.

The influence of acute poisoning with Dursban (O.P.I.) and D.D.T. (O.cl.I.) on serum enzymes and histopathological examination of the liver, kidney and testes was investigated in albino rats. Two repeated i.p. injections of Dursban in a dose of half the LD 50 resulted in a significant increase in serum GOT, GPT and alkaline phosphatase activity and a decrease of cholinesterase. In case of DDT, two doses of 150 mg/kg orally resulted in a significant increase in the activity of serum GPT only, while three doses increased serum GOT and GPT. No significant change was observed in serum alkaline phosphatase and cholinesterase activity. Regarding the pathological examination it was found that in animals treated with Dursban there was liver necrosis of mid-zonal type and fatty change at the periphery. In case of DDT the liver cells lost their radial arrangements and showed fatty change. There was cellular infiltration in the centre, mostly mononucleolar cells. In both insecticides there was necrosis of some of the seminiferous tubules of the testes and cloudy swelling of the convoluted tubules of the kidney. Histochemical study of the liver in animals treated with Dursban showed that glycogen was deposited at one side of the cell. However, there was depletion of glycogen around the central vein. In liver treated with DDT there were large globules of fat inside the liver cells, indicating increased fat content compared to control liver, where there were tiny minute droplets of fat.

The determination of enzyme activity in serum for the diagnosis of chronic hepatitis has become increasingly popular. According to the author's experience serum aminotransferase is raised in about 100% of cases of chronic active hepatitis and also in active cirrhosis, but in only about 70--80% of persisting hepatitis or in moderately active chronic hepatitis. They are frequently normal in inactive cirrhosis. After aminotransferases the alkaline phosphatase is of great importance for the differential diagnosis of icterus. If aspartate aminotransferase, alanine aminotransferase and alkaline phosphatase are determined at the same time, every cholestatic icterus can be diagnosed with certainty.

Liver enzymes alkaline phosphatase and transaminases (GPT & GOT) were studied in cases of protein-depleted rats. Alkaline phosphatase activities were determined with and without Mg addition to the incumedia, since it is the essential metal for this enzyme. The liver transaminases were also determined before and after pyridoxine injection, which is the coenzyme for this group. Both liver alkaline phosphatase and transaminases activities were increased on protein depletion. The study indicates that the increased activities of liver alkaline phosphatase in protein-depleted animals is suggestive of increased enzyme protein synthesis. On the contrary, high activities of liver transaminases are suggestive to be a result of some regulation mechanisms between the enzyme protein and its coenzyme.

The following blood indices were determined in the blood of 34 pregnant sows of the Large White breed under standard feeding conditions: haemoglobin, haematocrit, leucocytes, and--in the blood serum, --total protein, glucose, urea, bilirubin, cholesterol, enzyme activity (AP, GOT, GPT, GGTP) and mineral concentrations (Ca, P--inorg., Mg, Na, K, Fe, Cu, Zn, Mn). The blood was sampled in the first and third pregnancy at an average live weight of 165.12 and 197.36 kg, at an average age of 318 and 630 days and at an about the same average length of pregnancy in the time of both samplings (59 days). In younger, still growing gilts (first pregnancy) a significantly (P less than 0.05) lower content of total protein, magnesium, iron and copper was revealed, as compared with adult sows. The content of glucose, calcium, potassium and manganese in the blood serum of the gilts was significantly (P less than 0.05) higher than in sows in their third pregnancy. The adult sows showed a significantly (P less than 0.05) lowered activity of alkaline phosphatase and gamma-glutamyl transpeptidase, as distinct from gilts.

Normal values for a number of blood components of grivet monkeys are reported. Haematological data and values for glucose, cholesterol and urea are similar to those of rhesus monkeys. Activities of alkaline phosphatase (1526 U/l), glutamine oxaloacetate transaminase (30.9 U/l), glutamine pyruvate transaminase (13.7 U/l), lactate dehydrogenase (629 U/l), alpha-hydroxybutyrate dehydrogenase (175 U/l), creatine phosphokinase (227 U/l), gamma-glutamyl transpeptidase (38.7 U/l) and sorbitol dehydrogenase (14.2 U/l), and levels of lysozyme (178 mg/dl), zinc (162 microgram/dl), copper (81.3 microgram/dl) and iron (296.5 microgram/dl) have not previously been reported for this animal. Values for serum amino acids, proteins, electrolytes, triglycerides and creatinine are compared with those of other primates.

Twenty-five intravenous cholangio-cystograms were carried out in children aged 2 to 15 years (10 by injection, 15 by infusion). Total bilirubin, GOT, GPT, GLDH and alkaline phosphatase were determined before and after injection of the contrast medium. The contrast media used were "Biligram for infusion" (17%) and "biligram for injection" (35%). Contrast dose per kilo body weight depends on the age of the patient: a) For infusion: infants 1.6 ml/kg/KG, small children 1.2 ml/kgKG, older children 0.8 ml/kg/KG. b) for injection: infants 0.8 ml/kg/KG, small children 0.6 ml/kg/KG, older children 0.4 ml/kg/KG. Both methods, in the above doses, provided good demonstration of the biliary tree and gall bladder. Films were taken at 30 minutes, 60 minutes and 90 minutes after the end of the injection, and 40 minutes after a fatty meal. No allergic reactions were observed, nor any effect on the liver enzymes.

The effect of the 3-monthly injectable contraceptive depot medroxyporgesterone acetate (DMPA) on liver function and lipids was assessed in Thai women both with and without liver fluke (Ophisthorchis viverrini) infestation. DMPA administration was started in the immediate postpartum period and women who accepted immediate postpartum IUD insertion of sterilization were recruited as a control group. Complete 18-month followup results were obtained for 108 DMPA and 106 control fluke-positive subjects and for 89 DMPA and 74 fluke-negative subjects. No woman in any of the groups developed signs or symptoms of hepatic disease and the DMPA users had fewer health-related complaints during followup than the control subjects. Over 80% of both groups of users were amenorrheic 18 months postpartum, compared with about 15% of those in the control group. A large majority of subjects in each group continued to breastfeed for the entire study period without complaint. Weight change was small and similar in both the DMPA and control groups. Total bilirubin, aspartate aminotransferase, alanine aminotransferase, dehydrogenase, and alkaline phosphatase levels at 6, 12, and 18 months in the DMPA groups were generally equivalent to or lower than those in the corresponding control groups. Cholesterol levels were significantly decreased in the fluke-positive DMPA subjects while serum triglycerides were significantly decreased in both DMPA groups compared with their controls throughout the followup period. We conclude that during 18 months of use, DMPA did not cause any deleterious effects on health or on the metabolic factors studied in women with and without liver fluke infestation.

The present study investigated the activities of lysosomal enzymes of the liver after administration of Furosemid. 10 weeks and 1-year old male albino rats were treated with 40 mg Furosemid for 4 subsequent days. According to the method devised by de Duve a sediment rich in lysosomes was produced by fractionated centrifugation and subsequently the enzyme activity of beta-glucuronidase, beta-acetylglucosaminidase, cathepsin D and a collagenolytic enzyme was measured in the sediment as well as in the corresponding lysosomal supernatant. The protein content served as a reference for the enzyme activities. In addition, we investigated the activities of cytoplasmic enzymes such as GOT, GPT, gamma-GT and the alkaline phosphatase. The enzyme activity changes were age-dependent. With Furosemid treatment the activities of beta-glucuronidase and cathepsin D increased in the lysosomal supernatant and the lysosomal sediment of the 1-year old rats, whereas the activities of the collagenolytic enzyme increased in the lysosomal sediment of the same group. In the lysosomal sediment of the 10-weeks old rats a decrease of beta-glucuronidase, beta-acetylglucosaminidase and cathepsin D was observed. These results are discussed in the light of reports from the literature.

5 serum protein polymorphic systems (haptoglobin, alkaline phosphatase, group-specific (Gc) proteins, beta2-glycoprotein 1 and leucine aminopeptidase) and 6 red-cell polymorphisms (adenosine deaminase, adenylate kinase, phosphoglucomutase, glutamic-pyruvic transaminase, phosphogluconate dehydrogenase and acid phosphatase) have been investigated in 54 subjects with tuberous sclerosis. The frequencies of all systems were compared with those of a control sample drawn from a similar mentally retarded population and abnormal distributions were detected in the haptoglobin and Gc system. Quantitative estimation of the serum levels of the Gc protein failed to detect any inter-group differences. Data on the deviations from the Hardy-Weinberg equlibrium, Haldane's Log ratio test between groups, and gene frequencies of both test and control groups are given. It is suggested that selection by mortality is the possible causation for the abnormal distribution of the Gc phenotypes, but the haptoglobin phenotype distribution requires further investigation with care being taken in the selection of control subjects.
