Tetrahymena pyriformis Wh 14 was grown in Erlenmeyer flasks under continuous stirring at 30 degrees C for three days . After the culture had produced dry matter of about 100 mg HCB was added in acetone at a dose level of 0, 0.001, 0.1 and 1.0 ppm to the culture and incubated for another 7 days. At a dose level of 0.001 ppm the activity of delta-aminolevulinate dehydratase, hexokinase, and pyruvate kinase remained unaffected but was increased for glutamic-oxaloacetic transaminase, glutamic dehydrogenase, isocitrate dehydrogenase, and malate dehydrogenase while 0.1 ppm HCB increased the activity of all enzymes studied, the only exception being glutamic-pyruvic transaminase, the activity of which was depressed by HCB exposure. A concentration of 1.0 ppm HCB depressed the activity of most of the enzymes below control values with the exception of the two mitochondrial enzymes, MDH and ICDH, studied here.

We studied simultaneously in serum (S) and CSF (L) the enzyme activities of GOT, GPT, LDH, ICDH, MDH, ALD, and CPK in 28 patients with head injuries divided into three groups according to the severity of the trauma. We found a correlation between severity of brain lesion and enzyme activity. The best correlation was found for SGOT, SCPK, LGOT, LLDH, LMDH and LCPK. We do not believe that enzyme activity is of prognostic value. We think that further studies should be made of the specific isoenzymes of the Central Nervous System.

A description is given of an outbreak of equine infectious anaemia (E.I.A.) in Campania [at Naples and Aversa (Caserta)]; it was diagnosed by clinical, pathological and serological examinations (Coggins test). Using the serum of 45 horses with E.I.A. and 11 healthy horses (controls), numerous investigations were carried out on: enzymes, intrinsic coagulation factors, lipids and other substances. The results obtained were very interesting and show that in this disease there are significant increases in many enzymes (LDH, LAP, gamma-GT, CPK, PK and ALD) and copper. Insignificant increases were found in other enzymes (SDH, GLDH, MDH, ICDH, AIP, lysozyme, cholinesterase, GOT and GPT) and also intrinsic coagulation factors, lipid substances (total cholesterol, esterified cholesterol, triglycerides) and glucose. LDH-1-isoenzyme remains unchanged, whilst AcP decreases slightly.

The study was carried out on rats of the Wistar strain with experiments hypertension by the method of Selye, modified by Kolarova. These authors examined the activity of the enzymes of protein metabolism (GOT, GPT, AP), of glycolysis (ALD, LDH, alpha-HBDH) and of the citric cycle (MDH) in the left and right kidney at various intervals after modeling the process (3th, 15th, 30th and 90th day). The activity of the enzymes of the glycolytic chain and the cycle of Krebs in the left kidney was lowered at all stages of the experiment. Transaminase activity diminished progressively. Such a dynamics revealed the activity of adenosine phosphate as well. The activity of all examined enzymes of the right kidney showed phase changes-after initial elevation on the third day there was a lowering at the later stages of the experiment. The altered activity off the left kidney was connected with the disturbed vascularization with subsequent metabolic changes. The changes in the enzymic activity in the right kidney were assumed as a consequence of the occurred adaptive hyperfunction.

Serum activity analyses of nine enzymes [CPK, HBDH, SDH, ALP, GPT, GOT, LDH, MDH, ALD] in patients with rheumaitc carditis revealed certain elevations of the mean values of LDH, GOT, HBDH, GPT, ALP and MDH. These changes have no particular diagnostic importance because similar findings were also obtained in patients with both rheumatic and nonrheumatic arthritis. The changes occurring during antirheumatic treatment are minimal and statistically not significant. Greater elevations of LDH, ALD, GOT, HBDH, GPT, MDH, and ALP are found in patients with decompensated rheumatic heart disease associated with active rheumatic carditis.

Serum GOT, GPT, LDH, CHE, MDH, SDH, PK, GLDM, LAP and amylase levels and serum ammonia were determined in 49 major burn cases with 2nd and 3rd degree lesions covering 25% to 80% of the body surface. GOT, GPT, LDH, CHE and gamma-GT have above normal in all cases. Limited and inconsistent movements were noted for the remaining enzymes. Blood ammonia was constantly increased and it is felt that the enzyme picture may be of considerable prognostic value in major burn cases.

Seven-day exposure to clinostatic and antiorthostatic hypokinesia led to a significant decrease of Krebs cycle enxymes (malate dehydrogenase--MDH and isocitrate dehydrogenase) and creatine phosphokinase with no significant changes in lactate dehydrogenase (LDH), aspartate aminotransferase, alanine aminotransferase and MDH and LDH isoenzymes. This hypofermentemia returned to normal during 7-day readaptation. It was not associated with hemodynamic changes or emotional stress. The hypofermentemia was adaptive and induced by diminished motor activity.

1. PK and LDH activities in the muscle of Periophthalmodon schlosseri and Boleophthalmus boddaerti were at least 100-fold higher than their respective activities in the liver. 2. The ratio of PK:PEPCK in liver of B. boddaerti was smaller than that of P. schlosseri. 3. PK:PEPCK ratios in both fishes were intermediate between those of aerobic and anaerobic organisms. 4. MDH activity was higher than other enzymes assayed in the liver of both fishes. 5. The ratios of LDH:MDH in the liver of both mudskippers were comparable to those of anaerobic organisms. 6. AST was at least eight times more active than ALT in the liver of both fishes. 7. In the muscle of these mudskippers, the aspartate content was significantly less than that of alanine. 8. Exposure of these fishes to various experimental conditions led to changes in specific activities of PEPCK, LDH, AST and ALT.

Biochemical changes in the placenta were studied using alloxan-induced diabetes mellitus in the female rat. In comparison with a control group (n = 13) the placentas of the diabetic animals (n = 12) had significantly higher glucose, glycogen and protein levels. It was, however, shown that this supply of substrate was inadequately utilised for energy, as ATP/ADP quotient was lower and the ADP content was significantly higher. Metabolism still appeared to take place under aerobic conditions, as evidenced by the unchanged lactate levels. In terms of the protein content of the placentas, the activity of the enzymes we investigated (GOT, GPT, LDH, G-6-PDH, MDH, ICDH) was lowered by 25-44%. These results support the idea of global placental insufficiency in diabetics.

Heparinized venous blood was stored under sterile conditions at different temperatures (4 C, 20 C, 37 C) for various intervals (0-7 days). After storage the granulocytes and lymphocytes were isolated with routine methods. Naphthol AS-D-chloroacetate esterase as a granulocyte marker and acid alpha-naphthyl acetate esterase as a T-lymphocyte marker were identified on smears of the washed cell suspension. Different enzymes were identified in the cell sediment with electrophoresis. Relatively pure lymphocyte suspensions were obtained within the first 24 h. After this time, however, the percentage of these mononuclear cells declined markedly. The percentage of isolated granulocytes varied slightly; there was a marked predominance of granulocytes (more than 70%) at all intervals investigated during the isolation. Cytochemical analysis of the granulocytes and lymphocytes indicated that the decrease in the percentage of enzyme-positive cells depends in each case on the duration of the storage interval. During the first 24 h, only PGM1 and GOTM could be identified in the lymphocyte suspension with horizontal starch gel electrophoresis. The enzymes PGM1, PGM3, PGI, MDH, GOTM, 6-PGD, ADA could always be identified in the granulocyte suspension; AK, FUCA, MEM could be occasionally identified; and GPT and GLO could never be identified.
