The effects of dietary thiamin, riboflavin and pyridoxine deficiencies on dimethylnitrosamine-induced lethality and hepatotoxicity were investigated in the rat. Development of deficiencies was monitored by growth rate, food intake, ratio of liver weight to body weight and the biochemical parameters (thiamin diphosphate effects for thiamin deficiency, glutathione reductase activity coefficient for riboflavin deficiency and erythrocyte glutamate-oxaloacetate transaminase activity for pyridoxine deficiency). Thiamin deficiency slightly increased the acute toxicity of dimethylnitrosamine as observed by the lowering of the LD50 dose and the greater increase in the serum glutamate-oxaloacetate transaminase and serum glutamate-pyruvate transaminase levels. Riboflavin deficiency, on the other hand, slightly increased the LD50 dose of dimethylnitrosamine and resulted in less dimethylnitrosamine-induced damage to the liver. Pyridoxine deficiency did not affect the lethal dose nor significantly alter the transaminases levels.

31 healthy Thai males, 22 Thai male regular drinkers not suffering from any clinical signs or symptoms of alcoholism, and 52 patients from a neurological hospital in Bangkok suffering from the effects of chronic alcohol consumption were investigated. Alcohol consumption in asymptomatic drinkers ranged from 7 to 134 (median 44) g/d ethanol, and for the patients 22 to 517 (median 197) g/d ethanol, as assessed by questionnaires. The symptomatic alcohol drinkers had consumed alcohol for 2 to 35 years and the hospitalized patients for 5 to 40 years. Only the median levels of serum triglycerides and serum glutamyl transferase (gamma-G) were significantly increased and vitamin B1 deficiency was found with higher frequency in the group of alcohol drinkers without clinical signs compared with the healthy non-alcohol drinkers. Statistically significant correlations were demonstrated in the group of asymptomatic alcohol drinkers only, between alcohol consumption and the Quetelet's index, gamma-G, and alkaline phosphatase levels. Alkaline phosphatase also correlated significantly with gamma-G. In the group of hospitalized patients, compared with healthy males statistically significantly higher median values of systolic and diastolic blood pressure, serum triglyceride, gamma-G, aspartate aminotransferase (GOT), alanine aminotransferase (GPT), alkaline phosphatase, haemoglobin, hematocrit, folate and total protein were found. The median levels of cholesterol, bilirubin, vitamin B2, B6 and B12 in the hospitalized group were lower than, but not significantly different from the other two groups.

RNA, DNA, total protein, transketolase (TK), alanine aminotransferase (AlAT), aspartate aminotransferase (AsAT), methionyl-tRNA (met-tRNA)- and cysteinyl-tRNA (cys-tRNA)-synthetases in the thymus and spleen of 46 intact rats exposed to hypokinesia and vitamin B1 deficiency were measured. It was found that 15-day hypokinesia induced a significant decrease of the thymus and spleen weight and an increase of AlAT in the thymus. Vitamin B1 deficiency (hydroxy thiamine in drinking water at a dose of 2 mg/kg body weight for 15 days) led to a substantial decrease of TK in blood, AlAT and AsAT in the spleen; it diminished AsAT and increased AlAT in the thymus. Combined exposure to hypokinesia and vitamin B1 deficiency caused a more marked decrease of the weight of lymph organs, a significant loss of body weight, an increase of DNA in the thymus and spleen, and an increase of TK, met-tRNA-synthetase and AlAT in the thymus. These results suggest that vitamin B1 deficiency aggravates disorders in protein and nucleic acid metabolism in the lymph organs of hypokinetic animals.
