The deficiency of serine protease inhibitor, alpha-1-antitrypsin (AATD), is genetically determined defect that increases the risk of lung and liver disease development. The results of recent epidemiological studies indicate the overwhelming majority of individuals with alpha-1-antitrypsin deficiency still remain undiagnosed. The complete laboratory diagnosis of AATD is based on combination of quantitative and qualitative methods. The measurement of plasma/serum AAT concentration is always the initial test performed in clinically suspected individuals. Nevertheless, only the AAT phenotype or genotype identification allows the full medical verification of the diagnosis. Among the various techniques of either AAT variant phenotyping or genotyping accepted by reference medical centers worldwide, the isoelectric focusing, real-time-PCR and restriction fragment-length polymorphism PCR (RFLP-PCR) are "considered the most effective" performed the most commonly. The AAT diagnostics in Poland still awaits for introduction into clinical routine. The aim of present review is to outline the major methods of AATD diagnosis and discuss with the special issuing of their potential benefits and disadvantages.

A study was undertaken to determine the effect of a synthetic immunomodulator, i.e. methisoprinol applied in ovo, upon the hatchability of turkey poults under conditions of a standard hatchery as well as on their health status evaluated based on analyses of selected biochemical indices in their blood serum. Experiments were conducted on 5 groups of BUT 9 turkeys at the age of 5 days (35 birds in each group) hatched from eggs to which methisoprinol (VetAgro, Lublin, Poland) was applied in ovo at a dose of 5 mg (group I), 10 mg (group II) or 20 mg per egg (group III) on the 26th day of incubation. Turkeys hatched from eggs to which a physiological solution of NaCl was applied on the same day at a dose of 0.1 ml per egg (group IV) as well as those hatched from eggs without in ovo injection (group V) served as controls. Five hundreds eggs were used in each group. Hatchability was evaluated based on the number of hatched poults in respect of the number of eggs with live embryos transferred from the setting compartment to the hatching compartment, that were subjected to in ovo administration of the preparations according to the experimental design. Blood serum of the 5-day-old turkey poults was analyzed for activities of AST, ALP, LDH-L, CK, lysozyme and ceruloplasmine as well as for total protein and albumin contents. Analyses were also conducted for the immune system organ index - percentage contribution of organs of the immune system (spleen, thymus and the bursa of Fabricius) in the body weight of turkeys. The study demonstrated that methisoprinol administered to turkey embryos in ovo on day 26 of incubation at doses of 5, 10 or 20 mg per embryo did not induce any disturbances in the hatching process or affect its final result. In addition, it was shown not to exert any negative effect on the health status of the reared turkey poults.

Liver disease has become a global concern worldwide. In absence of reliable liver protective drugs in modem medicine, a large number of medicinal preparations are recommended for the treatment of liver disorders as they are believed to be harmless based on their natural origin. The aim of the present study was to determine the hepatoprotective activity of Phyllanthus amarus plant extract against carbon tetrachloride (CCl4-induced liver damage in female mice. Carbon tetrachloride administration caused a significant increase in liver and serum alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP) and acid phosphatase (ACP), while total protein content significantly decreased as compared to vehicle control. The effect was dose-dependent. Oral administration of aqueous extract of Phyllanthus amarus along with carbon tetrachloride caused significant mitigation of CCl4-induced changes.

Benign acute childhood myositis (BACM) is characterised by sudden calf pain and inability to walk. We analyzed the characteristics of seven boys and two girls with BACM treated in the Pediatric Department from April 2005 to March 2009. The mean age at onset of symptoms was 7 +/- 2 years. Two boys were hospitalized twice for BACM. All cases occurred in winter or spring. 7 out of all admissions were clustered together in one week long periods. Patients demonstrated prodromal symptoms of flu-like illness followed by the sudden onset of difficulty in walking. One girl additionally complained of a painful right hip. Four patients received inosine pranobex for prodromal viral infection before the clinical onset of myositis. In all cases, creatine phosphokinase (CPK; the highest value at 8988 U/l) and aspartate aminotransferase (AST; the highest value at 329 U/l) values were elevated. The serum concentration of myoglobin was elevated in five out of six tested patients (the highest value at 2172 microg/l). The following haematological abnormalities were detected: leucocytopenia (the lowest WBC 1.35 x 10(3)/microl), neutropenia, and trombocytopenia. All patients made a rapid recovery within 1 to 5 days. Pediatricians and emergency medicine specialists must be aware that BACM is a self-limiting disorder with the acute onset of inability to walk, elevated CPK and AST levels, and transient haematological abnormalities. There is no sufficient data from clinical reports on immunostimulant use before the onset of BACM.

The aim of this study was to compare some blood biochemical indicators in cows with displacement of abomasum (DA) which recovered or died after treatment. Examinations were performed on 60 multiparous cows with left (L) or right (R) displacement and on 15 healthy herdmates. Diagnosis was made by experienced practitioners on the basis of clinical examination. Surgical treatment was undertaken during the first 24 hours after diagnosis. Almost all animals (55 = 91.5%) became sick in the post parturient period (21 days p.p. on average) with the exception of 5 (8.3%) that became sick later. Blood samples were taken from each cow immediately before surgical procedure. Serum nonesterified fatty acid (NEFA), glucose (Glu), cholesterol (Chol), aspartate aminotransferase (AST), total bilirubin (Bil) and blood urea nitrogen (BUN) were measured. Sick animals were characterized by low mean values of Chol (< or = 2 mmol/l) and normal level of BUN (12-15 mg/dl), higher levels of NEFA (> 600 micromol/l) and Bil (> 22 micromol/l), higher activity of AST (> 100 U/l). Seven cows (11.67%) died after surgical correction and all others recovered. No significant differences in NEFA, Chol, AST, Bil and BUN levels were observed as dependent on the efficacy of treatment (survival, deaths). It was found that cows which died after surgical treatment were characterized by significant higher levels of glucose (5.05 mmol/l) compared to surviving cows (2.93 mmol/l).

An increasing number of new biomarkers of alcohol abuse appear in the literature. The most commonly used biomarkers (5-hydroxytryptophol, fatty acid ethyl esters, ethyl glucuronide, phosphatidyl ethanol, ethyl sulphate, mitochondrial aspartate aminotransferase, carbohydrate deficient transferrin, acetaldehyde adducts, beta-hexosaminidase, and sialic acid) were described. Then other known and less known biomarkers associated with alcohol abuse were described in brief (e.g. acetaldehyde, acetate, methanol, alpha-amino-n-butyric acid, dolichol, proteomics). Their sensitivity and specificity is generally higher than that of traditional biomarkers. The time of detection in biological fluids occur from one day to few months after alcohol consumption. Hence, their usefulness in clinical practice as well as in experimental studies is increasing.

The increasing interest in enzymatic growth promoters prompted this investigation of the effect of an enzymatic stimulating complex on selected hematological and serum biochemical parameters in boars. The enzymatic complex [five proteases (proteinases-endopeptidases) and two peptidases (exopeptidases) obtained by fermentation from Streptomyces fradiae] was added to diets for three months, at initial doses of 90 (group E1) and 120 (group E2) g/ton feed for the first seven days followed by 40 and 60 g/ton, respectively, in the subsequent weeks. The evaluation was based on selected hematological (RBC, WBC, HCT (Ht), HGB (Hb), MCV, MCH, and MCHC) and biochemical (AST, ALT, AP, Ca, Mg, urea, cholesterol, creatinine, inorganic phosphorus, and oxyglucose) parameters determined before and after the experimental period. The reported values were within physiological norms. Statistical differences were found between experimental groups with respect to WBC, HCT (Ht), MCV, MCH, AP, Ca, Mg, creatinine, and inorganic phosphorus. The study results indicate that the complex of proteolytic enzymes administered to boars caused no negative changes in their metabolic profiles. The statistically different hematological and serum biochemical parameters, while within normal limits, point to an early stage of microcytic anemia and heightened agitation that could be the result of intensified nitrogen metabolism.

The objective of the study was to determine the effect of replacing triticale (high rumen degradable starch) with maize grain (low rumen degradable starch) during the transition period and the first 120 days of lactation on metabolic and hormonal profile indices, milk production and fertility performance in cows. Forty-eight Holstein-Friesian dairy cows were divided into 4 groups: TT (2.5 kg triticale grain/cow per day supplemented from 14 days prepartum to day 120 postpartum), TM (2.5 kg triticale grain/cow per day supplemented from day 14 before parturition to calving, and then 2.5 kg maize grain to 120 days of lactation), MT (2.5 kg maize grain/cow per day supplemented from day 14 before parturition to calving, and then 2.5 kg triticale grain to 120 days of lactation), MM (2.5 kg maize grain/cow per day supplemented from 14 days prepartum to day 120 postpartum). Blood samples were collected 3 weeks and 1 week before calving and on days 14, 56 and 70 of lactation, and they were analyzed in terms of concentrations of glucose, insulin, leptin, insulin-like growth factor I, nonesterified fatty acids, triglycerides, cholesterol, blood urea nitrogen and activities of aspartate aminotransferase and gamma glutamyl transpeptidase. Milk samples were collected twice a day at weekly intervals and analyzed for fat, protein and lactose. Milk yield and individual dry mater intake were recorded at weekly intervals. Body condition was estimated 3 weeks before calving, on parturition day and on days 14, 56 and 120 of lactation. Replacing triticale grain with maize grain in the transition period and during lactation positively affected fertility of lactating cows. An increased first service conception rate and shortening of the days open period was observed in MM and TM groups in comparison to those found in group MT (P < or = 0.05). The lowest number of services per conception was recorded in groups MM and TM (P < or = 0.05). Although the impact of milk production and the most of the blood indices were not significantly affected by this treatment, the results of the study suggest that maize grain in the transition period and lactation might be a more effective energy source for dairy cows than triticale grain.

In recent years there have been remarkable developments in the prevention of diseases, especially with regards to the role of free radicals and antioxidants. Ethanol-induced oxidative stress appears to be one mechanism by which ethanol causes liver injury. The protective effect of aqueous plant extract of Aframomum melegueta on ethanol-induced toxicity was investigated in male Wistar rats. The rats were treated with 45 % ethanol (4.8 g/kg b.w.t.) for 16 days to induce alcoholic diseases in the liver. The activities of alanine aminotransferase, aspartate aminotransferase and triglyceride were monitored and the histological changes in liver examined in order to evaluate the protective effects of the plant extract. Hepatic malondialdehyde and reduced glutathione, as well as superoxide dismutase and glutathione-S-transferase activities were determined for the antioxidant status. Chronic ethanol administration resulted in a statistically significant elevation of serum alanine aminotransferases and triglyceride levels, as well as a decrease in reduced glutathione and superoxide dismutase which was dramatically attenuated by the co-administration of the plant extract. Histological changes were related to these indices. Co-administration of the plant extract suppressed the elevation of lipid peroxidation, restored the reduced glutathion, and enhanced the superoxide dismutase activity. These results highlight the ability of Aframomum melegueta to ameliorate oxidative damage in the liver and the observed effects are associated with its antioxidant activities.

The aim of this study was to verify the hypothesis that the health and growth of turkey poults may be improved by supplementing diets fed to parent flocks with available selenium. Experimental poults originated from parent flocks fed with diets containing 0.3 mg/kg inorganic selenium (control group Se(M)) and organic selenium (experimental group Se(O)). Egg yolk selenium content was comparable in both flocks (0.72 and 0.70 mg/kg d.m., respectively). Eggs from the Se(O) flock had a significantly lower content of thiobarbituric acid reactive substances - TBARS (31.13 vs. 53.10 nmol/g, p > 0.001). Se(O) group poults were characterized by higher activity of glutathione peroxidase (7.54 vs. 5.92 U/mL, P = 0.001) and superoxide dismutase (89.30 vs. 79.23 U/mL, P = 0.026). The thigh muscles of Se(O) group birds had significantly higher selenium concentrations (0.74 vs. 0.57, p = 0.045) and a significantly lower TBARS content (38.42 vs. 65.01, p = 0.001). No differences were found between the groups with respect to the content of total protein, albumins and uric acid, and the activites of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (DLH) in day-old poults. On day 28, groups Se(O) and Se(M) differed in the activity of ALT (20.50 vs. 26.33, p = 0.05) and SOD (87.29 vs. 100.02 U/mL, p = 0.035). There were no differences between the groups regarding the percentages of T lymphocyte subpopulations CD4+, CD8+, CD4+CD8+ and B lymphocyte subpopulations (IgM+) at 1 and 28 days of age. Over the experimental period, mortality rates were similar in both groups (7.32 and 8.87%), and so were the final body weights of birds (1108 vs. 1135 g). The results of the study show that the dietary supplementation of organic selenium in turkey parent flocks reduces the rate of oxidation processes in the egg and in the tissues of newly-hatched poults, yet it has no effect on the analyzed parameters of cell-mediated immunity and the growth performance of birds during the first five weeks of their life.
