The performance of the Olympus AU 400 clinical chemistry analyzer was evaluated according to the guidelines of the European Committee for Clinical Laboratory Standards. The following analytes were tested: glucose, urea, creatinine, calcium, AST, ALT, CK, LDH, ALP and amylase. The Olympus AU 400 was compared with the Olympus AU 800. Coefficients of correlation showed high correlation between the compared analyzers. Other performances (intra- and inter-assay variation, carry-over and interferences) of the analyzer were satisfactory.

The toxicity of diet containing 10% of Citrullus colocynthis fruits or 10% of Nerium oleander leaves or their 1:1 mixture (5%+5%) for rats treated for 6 weeks was determined. Dullness, ruffled hair, decreased body weight gains and feed efficiency, and enterohepatonephropathy characterised treatment with C. colocynthis and N. oleander given alone. Diarrhoea was a prominent sign of C. colocynthis poisoning. Organ lesions were accompanied by leucopenia, anaemia and alterations in serum AST, ALT and ALP activities and concentrations of total protein, albumin, urea, bilirubin and other serum constituents. Feeding the mixture of C. colocynthis and N. oleander caused more marked effects and death of rats.

20 year old man 2 years treated for the seropositive rheumatoid arthritis was admitted for fever accompanied with jaundice, anemia and leukopenia. The underlying disease has been compensated already for long period of time, before his admission only Prednisone (in the dose of 5 mg daily) and Methotrexate (15 mg once a week) was given. His physical examination of admission was without any significant abnormalities, out of the routine laboratory examination the value of leukocytes count was 2.1 x 10(9)/L, erythrocytes 3.7 x 10(12)/L, hemoglobin 95 g/l, hematocrit 0.29, platelets 156 x 10(9)/L. Since admission to hospital the hepatic enzymes ALT, AST, GMT, ALP were about ten times elevated comparing to normal values, the coagulation examination has shown the decrease of Quick test to 55%. With respect to the permanent leukopenia the bone marrow aspiration was taken with the finding of the increase number the RES elements (18.4%) with the signs of hemophagocytosis. The phagocytic reticulum absorbs blood elements erythrocytes, normoblasts, granulocytes, platelets. According to the literature experience we started the combination of the immunosuppressive treatment consisting of corticosteroids and Cyclosporine. Already the day following the application of the high dose of corticosteroids the fever subsided, icterus went away gradually with the normalization of the liver tests. After 20 days of hospitalisation the patient was discharged in good shape. Now, after 4 months the is stabilized on the follow-up treatment of Prednisone a Cyclosporine.

The course of Trypanosoma congolense infections in African grey duiker (Sylvicapra grimmia) and sheep and goats were studied. Several parameters suggested that the grey duiker was much more resistant to trypanosomosis than sheep and goats. They showed increases in weight during infection, had a much longer pre-patent period, and their peak parasitaemia levels were about 100-fold lower than those of sheep and goats. Parasites were no longer detected in grey duiker blood 35 days after infection. Anaemia, measured as drops in packed cell volume (PCV), haemoglobin (Hb) concentration and erythrocyte (RBC) counts were not observed in the grey duiker. In contrast, sheep and goats suffered severe weight losses and had continuously high parasitaemia levels. Sheep and goats developed progressively severe normocytic normochromic anaemia and leucopenia from day 14 post-infection onwards. Serum levels of total protein, globulin and albumin of grey duiker did not change significantly throughout the course of infection, while the levels of total serum protein, globulin and gamma-globulin exhibited significant increases from day 21 post-infection onwards in sheep and goats, with peak values recorded on 28 and 35 days post-infection in sheep and goats, respectively. There were inconsistent variations in albumin levels in sheep and goats throughout the course of infection. There were no significant changes in erythrocyte activities of AST and ALT, while there were transient but significant elevations of ALP level on day 35, and GGT levels between 14 and 35 days post-infection in grey duiker. Conversely, the levels of all the enzymes were progressively depressed, especially from 14 to 49 days post-infection. In vitro erythrocyte peroxidation remained relatively unchanged throughout the period of the experiment in the grey duiker, except for slight but significant increase on day 42 post-infection. However, in vitro erythrocyte peroxidation increased significantly by between 100 and 300% of pre-infection levels from 14th to 42nd day p.i. both in sheep and goats, before returning to pre-infection levels after 14 days of treatment. Haematological values, serum and erythrocyte indices studied returned to near pre-infection levels 14 days after treatment with Berenil((R)). It is concluded that the grey duiker is inherently trypanotolerant. This is shown by its ability to control parasitaemia, suffer less severe anaemia, and to a relative degree resist pathobiochemical derangements of some serum and erythrocyte metabolites and enzymes, as well as reduction of infection-induced erythrocyte lipid peroxidase damage than sheep and goats.

Body weight loss, inefficiency of feed utilization, diarrhea, ruffled hair and enterohepatonephrotoxicity were effects on male Wistar rats fed diet containing 10% Cassia senna or 10% Citrullus colocynthis ripe fruits for 6w. Rats fed a 1:1 mixture (5% + 5%) of fruits from these plants were more adversely affected and had deaths than rats fed the individual plants. The changes associated with the macrocytic hypochromic anemia and leukopenia were increased serum AST, ALT and ALP activities, increased urea, and decreased total protein, albumin and calcium. Serum bilirubin concentration did not change.

PMID:Combined toxicity of Cassia senna and Citrullus colocynthis in rats. 1130 22

Serum samples were collected from 42 harpooned minke whales (Balaenoptera acutorostrata) during commercial whaling off the coast of northern Norway (1997 and 1998) and analyzed for serum chemistry parameters in order to find clinical reference values for the northeastern Atlantic stock of this species. Mean and median values, as well as standard deviation and 90% central range, are presented for 28 different serum chemistry parameters. Lipemia is a common finding in marine mammals such as the minke whale, and chemical analysis of lipemic serum samples may produce artifacts. We found statistically significant elevated values of total protein, globulin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), sodium and chloride in strongly-lipemic compared to non-lipemic samples, all which may be artifacts due to interference of lipids with the methods used for analysis. In addition, we found significantly elevated levels of creatin kinase, lactate dehydrogenase (LDH), urea, uric acid and triglycerides, as well as a decrease in creatinine in the strongly lipemic samples. Reanalyzing serum samples after twelve mo storage at -20 C (n = 13) revealed reduction in the serum concentration of the enzymes ALT (42%), alkaline phosphatase (ALP; 10%), LDH (19%), gamma glutamyl transferase (17%) and amylase (11%), as well as for triglycerides (9%) and non-esterified fatty acids (16%). It is crucial that serum chemistry analysis is performed without delay after sampling. Possible changes in the values of some parameters due to the presence of high amounts of lipids or long term storage of samples must be considered when interpreting results from serum chemistry analysis in these animals.

This study was undertaken to assess the biochemical changes induced in chronic schistosomiasis and/or chronic HCV, as well as to pinpoint the most significant parameters which could be used as dependable indices for the differentiation of single and coupled infections with or without liver cirrhosis. The selected patients were allocated into 2 broad groups: GrII (Schistosomiasis) which was subdivided into 3 subgroups: GrII(a) schistosomal patients with hepatosplenomegaly; GrII(b) hepatosplenic schistosomal patients with decompensated liver cirrhosis; GrII(c) schistosomal patients with no organomegaly. GrIII (Combined) comprised 2 subgroups: GrIII(a) schistosomal-HCV infection with decompensated liver cirrhosis; GrIII(b) schistosomal-HCV infection without liver cirrhosis. For statistical comparison normal healthy subjects were taken as a reference group (Gr I). Results showed that schistosomal patients without organomegaly manifested non significant changes in all studied parameters compared to normal controls. Highly significant elevations in serum ALT, AST, ALP and GGT activities were recorded in all other subgroups but the highest levels are reported in GrIIb. AST/ALT and direct/indirect bilirubin ratios were highest in GrIIIa (1.17+/-0.26, 1.54 +/- 0.37, respectively). Serum total protein and albumin levels showed the highest reduction (33 and 59%) concomitantly with the highest increase in gamma-globulin level (75%) in GrIII(a). Blood total iron was significantly reduced in GrII(a,b) (15.6 and 12%) (8.8%) bilirubin, GGT and AST in this order are good discriminators between the different subgroups in GrII. On the other hand, ALT, AST, albumin, ALP, GGT, protein and direct bilirubin are the most significant indices to differentiate chronic schistosomiasis and the combined group with/or without liver cirrhosis.

This study evaluated the influence of Westernised and traditional African diets on biochemical and haematological profiles in vervet monkeys (Cercopithecus aethiops). Twelve adult male vervet monkeys bred at the Medical Research Council, all over 4 years of age and weighing more than 5 kg each, were divided into two groups of six individuals. These monkeys were raised on a standard in-house diet post-weaning, before they were fed for 8 weeks on diets containing milk solids (17.2%) or maize + legume (17.4%), as sources of high crude protein (+/- 3.5 g/kg). High protein diets had no significant effect on serum biochemical indices such as aspartate aminotransferase (AST) and gamma glutamyl transferase (GGT) concentrations (P > 0.10). However, alanine aminotransferase (ALT) concentrations were significantly higher during week 8 (P < 0.05) for the maize + legume protein group. Alkaline phosphatase (ALP; P < 0.07), total protein (P < 0.0001), albumin (P < 0.02), and bilirubin (P < 0.003) were elevated in the milk solids group, while glucose levels were also significantly higher for the milk solids group (P < 0.05) between weeks 2 and 6. Elevated protein intake had no significant effect on haematological parameters such as red blood cells (RBC), platelet and white blood cell (WBC) counts, haemoglobin levels and monocyte and neutrophil concentrations (P > 0.10). In contrast, serum lymphocyte levels were significantly raised in the maize + legume protein group (P = 0.03), whereas values for the haematocrit (P < 0.002), mean cell volume (MCV; P < 0.03) and mean corpuscular haemoglobin concentration (MCHC; P < 0.0001) were higher in the monkeys that were fed the milk solids. This investigation showed that the type of dietary protein that is consumed may well affect certain biochemical and haematological indices in vervet monkeys. Compared to the group that were given the traditional African food regime, the animals on the Western-type milk solids diet showed significant elevations in a number of important biological indicators. However, longer-term studies should be completed in this area if we are to make firmer conclusions regarding the link between the nature of dietary proteins that are consumed and its effect on metabolism.

Reference values are usually based on blood samples from healthy men or non-pregnant women. Blood samples from pregnant women may be compared with these reference values. Correct references for pregnancy can be extremely important for clinical decisions such as ablatio placentae, appendicitis, premature rupture of membranes and preeclampsia. Previous studies of normal variations during third-trimester pregnancy are incomplete. Blood samples during pregnancy weeks 33, 36 and 39 as well as 1-3 h postpartum were collected from pregnant women with dietary iron supplement and at least one previous pregancy without a history of hypertension or preeclampsia. When the sampled values were compared with the present reference values from men and non-pregnant women, the following differences were found during normal pregnancy: Haemoglobin and ferritin were reduced, CRP was slightly elevated, WBC (white blood cell count) and HNL (human neutrophilic lipocalin) were elevated during pregnancy and significantly increased postpartum. Albumin was reduced. ALT and AST were slightly elevated and GGT was unchanged during pregnancy. ALP, D-dimer and fibrinogen were elevated. Uric acid increased during the third trimester and thrombocyte count decreased. Separate reference values for pregnant women are essential for correct diagnostic decisions during third-trimester pregnancy. Elevated levels of D-dimer do not necessarily indicate ablatio placentae. A diagnosis of progressive preeclampsia cannot be based on increasing uric acid levels and reduced platelet count in a stable clinical condition. HNL signals activation of neutrophilic granulocytes and can thereby offer a helpful tool for diagnosing infection during pregnancy and postpartum.

Biomphalaria alexandrina were fed on either Allium cepa or A. sativum to study their effects on some biochemical parameters such as total proteins, free amino acids and liver enzymes (ALT, ALP and AST) on egg laying activity of the snails. The results revealed that ALP was highly significantly reduced in haemolymph of snails that fed on either Allium cepa or A. sativum. Also, ALT and AST were highly significantly reduced in haemolymph of snails that were fed on A. cepa while those fed on A. sativum showed no change in ALT activity and a high significant increase in AST activity. Total proteins were significantly decreased in haemolymph of all treated snails whereas variations in free amino acids contents were also observed. The reproductive activity of snails fed on either Allium cepa or A. sativum was highly affected. In addition, growth rate of newly hatched snails fed on either A. cepa or A. sativum was affected. Exposure of snails to water containing either A. cepa or A. sativum caused snail toxicity which may result from alterations in the snails' habitat.
