The objective of the paper was to assess the occurrence of congenital struma in kids in relation to the clinical and biochemical finding in their mothers. Observations involved 46 imported goats of Saanen and Alpine breeds in the course of kidding and their kids. Thyroid gland hypertrophy (39 goats) and somewhat worse or even bad state of nutrition were dominant clinical findings in pregnant goats and in goats after kidding. Abortions in the last month of pregnancy were recorded in 14 goats, and 14 goats delivered stillborn kids. Eighteen goats delivered 26 liveborn kids, but 18 out of them died within 12 to 24 hours after birth. Dead kids were hairless, they had skin edema, and very shortened thoracic as well as pelvic limbs. The thyroid gland was well visible and palpable. Surviving kids lagged behind in their growth and often suffered from bronchopneumonia as an additional disease. Iodine concentration in the blood serum of goats (5.58 +/- 2.14 mumol/l) was significantly lower (P < 0.01) in comparison with kids (133.4 +/- 15.61 mumol/l). This state was characterized by adequate T3 and T4 concentrations in the blood serum of goats (1.78 +/- 0.59 and 4.53 +/- 4.44 nmol/l, resp.) and of kids (4.66 +/- 2.26 and 182.93 +/- 2.59 nmol/l, resp.). Iodine content in the thyroid gland of the seven kids that died was 1.86 +/- 0.96 mg/kg fresh tissue. Examination of indicators of the internal environment in the blood serum showed alternate statistical differences (P < 0.01) between adult goats and their kids in erythrocyte counts, hemoglobin, hematocrit value, leucocyte counts, activities of aspartate aminotransferase, alanine aminotransferase, gamma-glutamyl transpeptidase, alkaline phosphatase, concentrations of total protein, albumin, total immunoglobulins, total lipids, cholesterol, phosphorus, copper, iron and zinc, while the explicit relation to disorders of iodine metabolism and thyroid hormones was not confirmed. The average content of iodine in the examined samples of soil (14.67 mg/kg) and alfalfa hay (0.1 mg/kg) demonstrated that primary deficiency of iodine in goats was the cause of congenital struma in kids.

The objective of this study was to determine the concentration of Zinc (Zn), Copper (Cu) and Manganese (Mn) in hepatic tissue from extrahepatic biliary atresia (EHBA). Liver biopsy samples were obtained at time of portoenterostomy from 49 infants ages 1.1 to 20.7 months (median 2.1) with EHBA. Samples were dry ashed and analyzed by flame (Zn) or flameless (Cu and Mn) atomic absorption spectrophotometry. Hepatic Cu concentrations are physiologically elevated at birth and decline rapidly during the first 2 month of life, therefore only samples from 29 infants, ages greater than 8 weeks were considered for Cu. Concentrations (mg/kg dry weight, mean and range) were: Zn 142 (70-507), Cu 204 (19-570), Mn 9.1 (2.8-21.8) vs. literature controls in the same age range: Zn 262 (82-543), Cu 92, Mn 4.3 (3.3-11.5). No correlations were found between serum alkaline phosphatase, AST or total bilirubin and hepatic trace element concentrations, between trace element concentrations and age, or between Cu and Mn. Decreased bile flow with intrahepatic cholestasis may result in hepatic accumulation of Mn as well as Cu. The low hepatic Zn concentrations indicate the need for further study of Zn metabolism in this population.

This study examined muscle swelling and changes in inflammatory markers in the blood following eccentric exercise-induced muscle damage. Subjects (N = 14) who had not been involved in a resistance training program performed 24 maximal eccentric actions of the elbow flexors. Muscle swelling was assessed by measures of the upper arm circumference (CIR), ultrasonography (USG), and magnetic resonance imaging (MRI). Plasma concentrations of interleukin-1 alpha, interleukin-1 beta, interleukin-2, interleukin-6, tumor necrosis factor-alpha, and plasma levels of C-reactive protein, cortisol, and zinc were analyzed. Established indicators of muscle damage (maximal isometric force, range of motion, muscle soreness, and plasma creatine kinase, aspartate aminotransferase, and lactate dehydrogenase activities) were also measured. All measures, including CIR and USG, except for MRI, were assessed immediately before and after and for 5 d post-exercise. MRI was taken at pre- and 1, 3, 6, 10, 23, 31, and 58 d post-exercise. All muscle damage indicators changed significantly after exercise. A large increase in CIR (> 20 mm) was found 4-5 d after exercise, and this coincided with USG, showing an increase in muscle thickness. The echointensity of USG increased with the enlargement of the elbow flexors. MRI displayed enlargement of the biceps brachii and brachialis cross-sectional area that started at 1 d, and lasted until 23 d, post-exercise. The most profound increase in the enlargement and signal intensity of the MRI was found 3 or 6 d after exercise. However, none of the plasma levels of inflammatory makers showed significant muscle swelling, which is indicative of muscle edema, but the inflammatory responses after exercise appear to be different from those accompanying infection or tissue injury.

Markers of lead intoxication have been developed based on their capacity to identify lead intoxication at the preclinical, ie biochemical stage of manifestation. However, little information on these markers is available under conditions of low lead exposure. This prompted us to conduct a community-based study to determine the usefulness of theta-aminolevulinic acid dehydratase (ALAD) and zinc protoporphyrin (ZnPP) in conditions of low lead environmental exposure by studying the relationships between low blood lead levels, ALAD and ZnPP in a large group of healthy dogs living in an Italian urban area. The study population consisted of 79 dogs. Each sample was tested for ALAD, lead and ZnPP and for complete blood count, hemoglobin, AST, ALT, and urea values. A weak inverse relationship between ALAD and ZnPP was found. An inverse relationship between ALAD and lead concentrations was found in the whole group (p < 0.0005). This relationship remained significant when selecting the values falling between 2 standard deviations of the mean blood lead concentrations of the population below the "concerned lead levels" (< 10 mg/dl; p = 0.0005). There was no relationship between whole blood ZnPP concentrations and whole blood lead levels. The sensitivity and specificity of ALAD measurements, calculated by using the 2 x 2 contingency table with respect to blood lead concentrations, were of poor predictive diagnostic value.

Experimental liver cirrhosis was produced by administration of thioacetamide. Cirrhotic animals were divided into two groups: one group was given zinc sulphate and the second kept as cirrhotic control. Zinc-treated animals showed a restoration of normal hepatic and plasma zinc and copper levels. Similarly, plasma levels of aspartate aminotransferase, alanine aminotransferase, gamma-glutamyl aminotransferase, and total bilirubin decreased significantly. Light microscopic studies showed that most of the hepatocytes appeared normal in zinc-treated as compared with untreated cirrhotic animals. The amount of fibrin, reticulin, and collagen, which was high in the cirrhotic livers, decreased following zinc treatment. Staining with periodic acid Schiff's reagent showed the ability of hepatocytes to store glycogen after zinc treatment. These results revealed that zinc may have some beneficial effect in the treatment of liver cirrhosis.

Eight Bonsmara bulls and eight Bonsmara heifers, having masses of between 210 and 266 kg when selected, were randomly allocated to four groups, each comprising two bulls and two heifers. Group 1 received 0.6 mg of copper (Cu)/kg of body mass per day (bm/d), group 2, 10 mg of Cu/kg of bm/d and group 3, 20 mg of Cu/kg of bm/d as a copper sulphate solution, given orally, 5 d a week over 745 d. Group 4 was the control group. One bull from group 3 was euthanased on day 679 of the trial, a heifer from group 3 and a bull from group 2, on day 695 of the trial, and a heifer from group 2, on day 731 of the trial, after they had shown clinical signs. During the course of the trial, clinical signs, serum gamma glutamyltransferase and aspartate aminotransferase activity, blood urea nitrogen, and plasma copper, zinc and iron concentrations were monitored. Live mass was recorded weekly to determine any effect on mass gain. The liver and kidney copper, zinc, iron and managanese concentrations at the time of death or slaughter are given. From the results it was concluded that subclinical damage to the liver and eventual copper toxicity can occur when cattle are continually exposed to oral doses > or = 12 mg of Cu/kg of bm/d. It was also concluded that cattle can probably tolerate oral doses of < or = 0.6 mg of Cu/kg of bm/d for an indefinite period, provided there are no other sources of copper, such as may occur with air-pollution, or provided no other adverse mineral interactions occur, such as may occur with molybdenum deficiency.

Nucleotide sequence analysis of the Helicoverpa zea S-type nucleopolyhedrovirus (HzSNPV) genomic interval between the polh and iel genes has revealed an open reading frame (HOAR ORF) that contains a complex A 1-T rich triplet repeat region (RAT-repeats). HOAR ORF is predicted to encode an acidic, arginine residue rich. 712 aa protein, with a C3HC4 (RING-finger) zinc binding motif. RAT-repeats, distributed over 450 bp. consist of GAT. AAT, and GTT codons, correspond to Asp, Asn and Val residues which display an extreme codon bias not seen with nine other genes of this virus. A survey of four other (field) isolates of Helicoverpa sp. NPVs confirms a high incidence of mutation in the RAT-repeat region. A 158-bp conserved block, homologous to the pe38-ien promoter of AcMNPV, was identified upstream of HOAR ORF. The sub-region of the genome in which HOAR ORF is located is susceptible to rearrangement.

Concentrations of 34 biochemical constituents of sera were determined on 998 randomly selected urban school children and adolescents aged 8-18 years from Zagreb, Croatia. Reference intervals were obtained by using non-parametric methods to estimate 2.5 and 97.5 percentiles of distribution as upper and lower normal reference intervals, according to the IFCC recommendations. These were compared to reference intervals in the healthy adult population, aged 20-30 years from the same geographical area. Serum glucose, potassium, sodium, chloride, magnesium, iron, zinc, total serum proteins and electrophoretic fractions, and amylase, did not show age or sex differences; total serum bilirubin, total calcium, phosphate, high density lipoprotein cholesterol, total iron binding capacity, unsaturated iron binding capacity, copper, aspartate aminotransferase, alkaline phosphatase, cholinesterase, creatine kinase, and lactate dehydrogenase had higher reference intervals than the adult population. Urea, creatinine, urate, alanine aminotransferase, gamma-glutamyltransferase, total cholesterol and low density lipoprotein-cholesterol, and triglycerides had lower reference intervals than the adult population.

Two groups of mouflons (Ovis ammon) were held in captivity to study the effects of repeated capture on physiological, haematological and biochemical parameters. The first one (Group I) was of 6 mouflons captured in the wild, while the second (Group II) was also of 6 mouflons, but which had been in captivity for 3 years. In Group I, body temperature, mature neutrophil count and lactate increased during activity, while red blood cells, haemoglobin, ALT, AST, total lipid, phospholipids, cholesterol, BUN, creatinine, phosphorus and zinc decreased at different times during the study period. In Group II, few statistical differences were observed. Most of these changes were related to stress and reflected a lack of adaptation to repeated handling.

In order to identify cytochemical traits relevant to understanding excitatory neurotransmission in brainstem auditory nuclei, we have analyzed in the dorsal cochlear nucleus the synaptic distribution of aspartate aminotransferase, glutamate, and vesicular zinc, three molecules probably involved in different steps of excitatory glutamatergic signaling. High levels of glutamate immunolabeling were found in three classes of synaptic endings in the dorsal cochlear nucleus, as determined by quantitation of immunogold labeling. The first type included auditory nerve endings, the second were granule cell endings in the molecular layer, and the third very large endings, better described as "mossy." This finding points to a neurotransmitter role for glutamate in at least three synaptic populations in the dorsal cochlear nucleus. The same three types of endings enriched in glutamate immunoreactivity also contained histochemically detectable levels of aspartate aminotransferase activity, suggesting that this enzyme may be involved in the synaptic handling of glutamate in excitatory endings in the dorsal cochlear nucleus. There was also extrasynaptic localization of the enzyme. Zinc ions were localized exclusively in granule cell endings, as determined by a Danscher-selenite method, suggesting that this ion is involved in the operation of granule cell synapses in the dorsal cochlear nucleus.
