The morphological, histochemical and biochemical changes in the liver of rats challenged with a single intraperitoneal injection of the toxin of lupinosis were investigated. The resulting injury is biphasic. The initial non-specific phase lasts for a few days, affects mainly the centrilobular region and is characterised by mitochondrial swelling, lipid accummulation and autophagosome formation. In addition, the hepatic concentrations of OCT, LDH, SDH, and AST fall while that for ALP increase. This is accompanied by a rise in the serum concentration of many of these enzymes. The second phase lasts longer and is characterised by phenomena indicative of abnormalities in hepatocytic division. Normal and abnormal metaphase as well as multinucleated hepatocytes falls. The possibility that the toxin of lupinosis interferes with microtubular units is considered.

The possible aggravation of liver injury by impaired cellular antioxidant function was investigated. A vitamin E-deficient diet (0.5 mg/kg alpha-tocopherol; control 100 mg/kg) significantly reduced rat liver alpha-tocopherol concentrations after 4 weeks (1.8 +/- 1.7 micrograms/g; control 34.4 +/- 2.4 micrograms/g, p < 0.001). The effects of copper loading (Cu, 3 g/kg diet); galactosamine (GalN, 0.85 g/kg i.p.); or carbon tetrachloride (CCl4, 10 mmol/kg i.p.) were examined. Serum aspartate transaminase activity was elevated slightly by vitamin E deficiency but not by hepatic copper accumulation. In vitamin E-replete (E+) and vitamin E-deficient (E-) rats, GalN or CCl4 caused a large and comparable elevation in serum AST and OCT activity. This effect on AST was markedly reduced by copper loading in vitamin E replete (E+) rats, but in E(-) rats copper had significantly less protective effect. Copper also diminished the OCT response to GalN in E+, though not E-, rats. A significant rise in total hepatic alpha-tocopherol content followed administration of GalN or CCl4 in both normocupric and copper-laden E(-) rats. Thus alpha-tocopherol deficiency (a) was not hepatotoxic per se; (b) failed to potentiate the toxicity of copper, GalN or CCL4; but (c) partially abolished the protection by copper against toxin-induced liver injury. Retention of hepatic alpha-tocopherol after liver damage may partly explain low serum vitamin E levels seen in clinical liver disease.

Although copper is believed to be hepatotoxic in Wilson's disease and Indian Childhood Cirrhosis (ICC), the rat shows only minimal hepatic damage on copper-loading. To investigate the possibility that copper deposition may potentiate the effects of a superimposed hepatitis, D-galactosamine (GalN) was given to copper-loaded and control rats. In the non-copper-dosed rats, GalN 0.85 g/kg i.p. produced elevated serum AST (3731 +/- 545 IU/l; normal 64.8 +/- 2.1), ALT (2090 +/- 190 IU/l; normal 18.0 +/- 0.7), and OCT (16.7 +/- 2.6 mmol/min/ml; normal 0.12 +/- 0), and liver cell necrosis with portal infiltration. In rats whose liver copper was elevated to 1298 +/- 169 micrograms/g (control 18.7 +/- 1.7) by oral copper supplementation, GalN produced much smaller increases in AST (825 +/- 122 IU/l), ALT (103 +/- 15 IU/l) and OCT (0.27 +/- 0.02 mmol/min/ml) and minimal histological damage. Viable bacterial cell counts from faecal homogenates showed that the anaerobically cultured bacteria were reduced on copper-dosing of rats. Therefore the protective effect of copper may be due to a decrease in gut-derived endotoxin acting on the liver, or to an impaired prostaglandin synthesis or perhaps to synthesis of acute phase reactants.

Health conditions were evaluated in 80 electrical workers exposed for many years to polychlorinated biphenyl (PCB) mixtures with a 42% mean chlorine content, who had blood PCB concentrations from 41 to 1319 micrograms/kg. The clinical study was based on personal history data, physical examination, and laboratory tests (red cell and leukocyte count; determination of haemoglobin, packed cell volume, bilirubin, serum protein electrophoretic fractions, pseudocholinesterase, AST, ALT, GGT, and OCT). Fifteen workers were found to have skin diseases--chloracne (4), folliculitis (4), oil dermatitis (1), juvenile acne (1), and dermatitis due to irritative or allergic agents (5). Sixteen workers showed more or less pronounced hepatic involvement, consisting most often of hepatomegaly with an increase in serum GGT, AST, ALT, and OCT values. In two workers bleeding cavernous haemangiomas were discovered, in one case associated with chronic myelocytic leukaemia. All the workers with chloracne were employed on electric capacitor impregnation with PCBs, and no definite association was found between chloracne and blood PCB concentrations. Conversely, a significant positive association was found between the abnormal liver findings and blood PCB concentrations, particularly trichlorobiphenyl blood concentrations. The abnormal hepatic findings observed are similar to those reported in experimental animals given PCBs, and in some workers such findings should probably be considered as clinical signs of hepatic microsomal enzyme induction.

Recent studies of Reye syndrome (RS) patients have suggested aspirin treatment as a possible factor in the etiology of this often fatal childhood disorder. the relationship of aspirin treatment to other factors that have been strongly implicated (influenza, ammonia toxicity) cannot be examined directly in patients because aspirin treatment is usually initiated by family members in the prodromal period before RS is diagnosed. In this report we describe the use of an animal model for RS in examining the interactions of these several potential etiological factors. Hyperammonemia and coma were produced in young male ferrets by a brief feeding of an arginine-deficient diet. The effects of influenza infection or aspirin treatment (or both) of control and hyperammonemic ferrets on their serum levels of ammonia, glutamic-oxaloacetic transaminase (GOT;L-aspartate:2-oxoglutarate aminotransferase, EC 2.6.1.1), ornithine carbamoyltransferase (OCT; carbamoylphosphate:L-ornithine carbamoyltransferase, EC 2.1.3.3), bilirubin, and salicylate were studied. Liver levels of lipids, proteins, and several urea-cycle enzymes were also determined in the comatose ferrets and compared with those of untreated controls and of controls treated with influenza or aspirin, or both. Synergism of these three factors (hyperammonemia, influenza infection, and aspirin treatment) in causing RS-like alterations in these parameters was observed.

Outbreaks of ovine white liver disease (WLD) on 7 farms in eastern Victoria were investigated. Most occurred in late spring and mainly affected lambs 3 to 6 months old, with a morbidity of 20 to 100% and mortality of 8 to 15%. Clinically affected lambs showed illthrift, emaciation and bilateral, serous, ocular discharge. Clinical pathology showed mild anaemia, elevated serum liver enzymes (GGT, OCT, AST) and low levels of serum vitamin B12. Grossly, the livers were pale, fatty and friable; microscopically there was parenchymal fatty change, bile duct proliferation and ceroid pigmentation. Liver cobalt values were consistently low (mean 0.4 +/- 0.4 mumol/kg D.W.). Levels of cobalt in pasture from 2 properties were very low (0.34 mumol/kg D.W.) The diagnosis of white liver disease was made on the basis of clinical features, specific liver pathology and low cobalt status. Treatment trials established that cobalt injections or oral bullet administration resulted in clinical improvement, significant weight gains, and improved serum vitamin B12 levels. WLD did not recur in previously affected sheep using these treatments. However, when blocks containing cobalt were available continuously, WLD recurred 2 years after the initial outbreak.

Thirty wine growers have been examined in a preliminary study to define the morbidity related to the use of pesticides. The health status was defined (history, physical examination, liver and kidney function study, cholinesterase, EMG) before any exposure and some parameters were reassessed during exposure. Several results vary significantly during treatment with pesticides (ALT, AST, OCT, ACHE, BCHE). The initial health status is good, showing no systematic pathology. Several episodes of acute intoxication are mentioned in the patient's histories.

Nine populations of Oncomelania, field-collected from Anhui, Shanghai, Jiangsu, Zhejiang, Jiangxi, Hunan, Hubei, Sichuan and Yunnan were studied by horizontal starch gel electrophoretic method with 24 enzyme systems (AAT, AcPH, AK, AO, APH, CK, EST, GDH, GPI, G6PD, HBD, ISDH, LAP, LDH, ME, MDH, MPI, NADD, OCT, PGM, 6PGD, SDH, SOD, XDH) analyzed. 40 loci and 117 alleles were detected in the Oncomelania. Both of GPI and PGM-I, with 7 alleles, were the most variable loci. 22 loci had more than 3 alleles each. Of 40 loci examined in the 24 isozyme systems, 14 were found to be polymorphic, the proportion of multilocus enzymes being 58.3%. Our results showed that the genetic polymorphism existing in the populations of Oncomelania in the mainland of China. PGM and MDH, were found in both the populations of Oncomelania and strains of Schistosoma japonicum in the mainland of China. The results provided a new idea for studying snails and Schistosoma. Also, we found that there might be some correlation between the polymorphic locus and the feature of the shell of Oncomelania snail.

Thirteen enzymes encoded by 16 loci of six population of Oncomelania hupensis in Zhejiang, China, were investigated by means of starch gel electrophoresis. Ten loci (AO, 6PGD, ME, AKP, OCT-1, HBDH-1, HBDH-2, XDH, MDH and MPI) were monomorphic and 6 loci (OCT-2, PGI, AAT, PGM-1, PGM-2 and ACP) were polymorphic. Three enzymes (OCT, HBDH and PGM) were encoded by 2 loci. The results indicated that there were allozyme variations in two subspecies, O.h. hupensis and O.h. fausti in Zhejiang, China. Nei's multilocus genetic distances (D) between subspecies ranged from 0.167 to 0.265. Minor genetic distances were detected between populations of the same subspecies. The results indicated that the enzyme acid phosphatase (ACP) is a possible marker to measure the degree of susceptibility of O. hupensis to S. Japonicum.

Blood of 24 healthy goats of different breeds in the age between two and six years as well as their 36 lambs in the peripartal period was taken at regular intervals from the day 10 ante partum up to the day 30 post partum. The content of plasma--nonspecific, clinical essential enzymes as well as the selenium content--has been examined. 1. Normal parturition shows an influence on the enzyme activity in plasma. Abundant alterations in concentration (p < or = 0.05-0.001) are detectable in AST, LDH, alpha-HBDH, GGT, GLDH and CK. GGT responds indistinctly to birth. GLDH-concentration is significant higher in the lactation period (p < or = 0.01). 2. The registration of OCT in blood plasma and serum of goats is possible but the activity of the enzyme in plasma and serum is slight. 3. In the blood of new born lambs a high plasma activity of CK and GGT can be found. Up to day 5 post natum the activity of CK descends to post partum adult level and up to day 30 post natum lower enzyme values than those of adult goats are found. GGT activity at day of birth after colostrum uptake is more than 5 times higher than at day 5 post natum. Up to day 30 post natum the activity remains above the mean of adult goats.
