This study compared the function of reduced grafts prepared in situ or ex vivo and transplanted immediately or after 4 hr of cold storage. Measurements of acid/base balance, plasma electrolytes, albumin, and

showed no differences between groups. There was no difference between the increase and decline of plasma

in recipients of grafts transplanted immediately after either ex vivo or in situ reduction; the increase in plasma
AST
of recipients of stored grafts was up to 10-fold and persisted until the end of the study at 7 days, with some decline. Plasma fibrinogen decreased intraoperatively but levels were restored within 24 hr in all groups; plasma prothrombin and partial thromboplastin times were not significantly disturbed. The patterns of decline and return of tissue adenine nucleotides were similar in all groups. While the regenerative response measured by tissue thymidine kinase and mitotic figures was not different between the groups, comparison with results from a group of partially hepatectomized animals showed a 3-4-fold depression in response in reduced liver grafts. The contributions of the effects of ischemia, flushing, and preservation to the depressed regenerative response of reduced liver grafts need to be determined. The present studies suggest however, that with regard to functional assessment, results are not affected either by ex vivo or in situ reduction of the graft, or by cold storage for 4 hr.

, Na, K and Cl. The results were interpreted according to their healthy condition, their age as well as their sex. The study had revealed an extreme metabolic acidosis, haemoconcentration, hypoglycaemia and hypofunction in the kidney and liver. Furthermore, the calves with diarrhoea had showed hyponatraemia, hypochloraemia, and hyperkalaemia. Important correlations between clinical and some blood parameters were found. Metabolic acidosis was more severe in male calves than females. These pathophysiological changes should be put in consideration during the therapy of newborn calves suffering from diarrhoea.

On the other hand, serum total proteins, uric acid, cholesterol, calcium and potassium were significantly decreased 4 h after administration of the 3 doses. These changes in clinical chemistry parameters are most probably related to the toxic effect of the venom on the target organs.

(2.33-2.37 mmol.l-1) and bilirubin (3.49-5.15 mmol.l-1) did not show any larger changes in dependence on the time elapsing from parturition. Cholesterol concentrations were gradually increasing from 2.58 mmol.l-1 on the day of parturition to 4.99 mmol.l-1 on day 45 after parturition. The five cysteines, at positions 82, 191, 192, 270, and 401, of Escherichia coli
aspartate aminotransferase
(AATase) were, individually and in some combinations, converted to alanine by site-directed mutagenesis (C82A, C191A, C192A, C270A, C401A). Cys-191, which is conserved in all AATase isozymes, was mutated to serine as well (C191S). A quintuple mutant, with all cysteines converted to alanines (Quint), was also constructed. The effects of these single and multiple mutations were examined by steady-state kinetics and
urea
denaturation. The thermal stabilities of Quint and of the wild-type enzyme (WT) were determined by differential scanning calorimetry. The mutants had kcat values up to 50% greater than that of WT and KMAsp and KM alpha-KG values up to 1.5- and 3.3-fold higher than that of WT. The mutants C82A and C191A exhibit nearly the same CM in
urea
denaturation experiments as WT, while the other single mutants and Quint are less stable, with CM differences of up to 0.7 M
urea
. Quint is also less thermostable than WT, with a delta TM of 3.3-4.4 degrees C. Thus the five cysteine replacements yield small, but significant, changes in catalytic and denaturation parameters, but none of the cysteines was found to be essential. The changes manifested in the mutation of the conserved Cys-191 to alanine are no greater than those observed with the four nonconserved cysteines. We consider the evolutionary implications of these findings.
