A refractile body antigen Etp28 Gene of Eimeria tenella (Guangdong Strain) sporozoites was cloned by PCR from the synthesized first strand cDNA. It has a high homology with the previously reported Etp28 gene of Merck Strain LS18. The gene was expressed through standard procedures in the modified Autographa californica nuclear polyhedrosis virus (AcMNPV-OCC(-)) expression system and large amount of heterologous fusion protein (

-6xHis-Etp28) was obtained. The expression product was about 21.3% of the total cellular protein of an infected cell and corresponding to approximately 0.42 mg of recombinant protein per 10(6) cells. The apicomplexan protozoon Eimeria coecicola is an infectious agent of intestinal
coccidiosis
in rabbits, causing severe injuries in the appendix as the final target, but also in the liver though not being a target. Here, we investigated the effect of E. coecicola on the spleen of the rabbit Oryctolagus cuniculus with respect to structure and gene expression using 2-color Agilent whole rabbit genome oligo-microarray technology in combination with quantitative PCR. At maximal fecal output of E. coecicola oocyts on day 7p.i., the spleen did not contain any parasites, but displayed moderate inflammatory changes evidenced as fused white pulp areas, diffuse appearance of the marginal zones, and increased number of macrophages in the red pulp, eventually resulting in an increased histological score. The infections induced 36 genes to be up-regulated and 156 genes to be down-regulated, among which were 139 genes encoding diverse regions of antibodies. The highest upregulated genes were those encoding granzyme H (10-fold), lim1 (10-fold), xanthine dehydrogenase (9-fold), whereas the downregulated genes could be majorly assigned to the immune response, as e.g. the genes encoding the macrophage cationic peptide-2 (5-fold). Quantitative PCR of genes encoding GZMH, XDH, HSD17B1, SULT3A1, SAA, BPI, MCP-2, and

exhibited the same transcriptional level as that detected by microarray analysis. The E. coecicola-induced changes in gene expression of the spleen were totally different to those found previously in appendix and liver. Only the granzyme H gene became upregulated in all three organs. Our data indicate that the spleen, though not a final target of E. coecicola, responds to E. coecicola infections, suggesting that the spleen may be part of an orchestrated host defense against E. coecicola critically involving GZMH-expressing NK-cells.

Coccidiosis
is caused by multiple species of the apicomplexan protozoa Eimeria. Among them, Eimeria tenella is frequently considered to be the most pathogenic. Zinc finger proteins (ZnFPs) are a type of protein containing zinc finger domains. In the present study, a putative Eimeria tenella AN1-like ZnFP (E. tenella AN1-like zinc finger domain-containing protein, putative partial mRNA, EtAN1-ZnFP) was cloned and characterized, and its immune protective effects were evaluated. The 798-bp ORF sequence of EtAN1-ZnFP that encoded a protein of approximately 27.0 kDa was obtained. The recombinant EtAN1-ZnFP protein (rEtAN1-ZnFP) was expressed in Escherichia coli. Western blot analysis showed that the recombinant protein was recognized by the anti-

monoclonal antibody and anti-sporozoite protein rabbit serum. qPCR analysis revealed that EtAN1-ZnFP was highly expressed in unsporulated oocysts and sporozoites. Immunostaining with an anti-rEtAN1-ZnFP antibody indicated that EtAN1-ZnFP was uniformly distributed in the cytoplasm of sporozoites, except for the refractive body; furthermore, this protein was evenly distributed in the cytoplasm of immature schizonts but seldom distributed in mature schizonts. The results of the in vitro invasion inhibition assay indicated that the antibodies against rEtAN1-ZnFP efficiently reduced the ability of E. tenella sporozoites to invade host cells. Animal challenge experiments demonstrated that the chickens immunized with rEtAN1-ZnFP protein significantly decreased mean lesion scores and fecal oocyst output compared with challenged control group. The results suggest that EtAN1-ZnFP can induce partial immune protection against infection with E. tenella and could be an effective candidate for the development of new vaccines.

spp., which goes through a complex life cycle in the intestinal mucosa of infected hosts. Specific immunoglobulins (IgY) could be produced in egg yolk by immunizing hens with specific antigens. In the present study, we cloned the
E. maxima gam56
gene, expressed the
GST
-GAM56 fusion protein and raised IgY to
GST
-GAM56 in hens. The anti-
GST
-GAM56 IgY antibody was isolated and used to treat chickens infected with
E. maxima
oocysts. Intramuscular injection of the antibodies provided minimal protection against parasite infection. However, oral dosing of the IgY 3 or 5 d after oocyst inoculation significantly improved body weight gain, reduced oocyst output and intestinal lesion score were reduced at 3 or 5 d after oocyst challenging, compared to the untreated control group. Our findings suggest that the IgY to gam56 could be an effective prophylactic or therapeutic agent against
E. maxima
infection in chickens and should have a practical application value.
