Thymidine kinase
-deficient OTT6050 mouse teratocarcinoma cells were fused with
hypoxanthine phosphoribosyltransferase
-deficient Fu5AH rat hepatoma cells by means of inactivated Sendai virus. The resulting hybrid cells, which were selected in hypoxanthine/aminopterin/thymidine medium, retained almost all of the mouse chromosomes and various numbers of rat chromosomes, and showed many chromosomal rearrangements. The hybrid cells, as well as both parental lines, formed tumors after subcutaneous injection into athymic nude mice. Single rat--mouse hybrid cells from a clonally established subline were transplanted into C57BL6/J mouse blastocysts carrying many genetic markers suitable for the detection of hybrid cell-derived tissue contributions. From 144 blastocysts, each of which was injected with a hybrid cell and then surgically transferred to the uterus of a pseudopregnant foster mother, 62 adult mice developed without any visible coat mosaicism. However, three of these mice showed internal hybrid-cell participation in their livers and a limited number of organs of endomesodermal origin. A tumor classifiable as hemangio endothelioma was found in the liver, the only mosaic tissue, of one of the chimeric mice. Nine different rat-specific enzyme variants were detected in the mosaic organs. A considerable number of variations concerning the presence and quantitative activity of the foreign gene products probably resulted from chromosomal segregation, tissue-specific gene activity, or dosage compensation during differentiation in vivo. Our results demonstrate that cultured malignant rat--mouse hybrid cells differentiate normally and become functionally integrated during development. The appearacne in vivo of certain rat-specific gene products that are not found in the hybrid cells under conditions in vitro indicates differential gene expression of the introduced xenogeneic chromosomes.

Recombinant vectors containing the mouse metallothionein-I gene (MT-I) and the Escherichia coli xanthine-
guanine phosphoribosyltransferase
gene (gpt) were used to transfect human hgprt- HeLa cells. Transfected MT-I genes are transcriptionally regulated by cadmium but not by glucocorticoids. S1 mapping indicates that the transcripts from transfected MT-I genes begin at the correct transcription initiation site. We also transfected mouse tk- L cells with a vector containing the mouse MT- I gene and the herpes simplex virus-I thymidine kinase gene. MT-I gene transcription is regulated by cadmium but not by glucocorticoids in this homologous system as well. Finally, we fused the MT-I gene promoter/regulatory region to the thymidine kinase structural gene.
Thymidine kinase
activity is regulated by cadmium when this fusion gene is transfected into mouse tk- L cells. Deletion mapping experiments indicate that the DNA sequences necessary for regulation of the MT-I gene by cadmium lie within 148 bp of its transcription start site.

The purpose of this study was to investigate the cytotoxic and genotoxic potential of low intensity laser irradiation (660 nm, 12 mW, 5 kHz) on mammalian cells.
Thymidine kinase
(TK)-positive and TK-deficient Friend erythroleukaemia (FEL) cells, clone 707 and subclone 707BUF respectively, were used in this investigation. Following irradiation of exponentially growing cells in suspension at doses of 2 and 20 J/cm2 a number of sensitive bioassays were used to facilitate the detection of laser-induced mutations, DNA damage and cell killing. Mutations were assessed by the examination of chromosome spreads, the determination of micronucleus frequency and by the determination of the mutant frequency at the
hypoxanthine-guanine phosphoribosyltransferase
(hgprt) locus. DNA damage was quantified using a sensitive ELISA. The cytotoxic effect of laser irradiation was assessed using a cloning assay. The results of this investigation did not show any significant increase in mutation frequency, DNA damage or cell survival in the laser-irradiated cells, compared to sham-irradiated controls. The lack of any demonstrable cytotoxic and genotoxic effects of low intensity laser irradiation on mammalian cells in culture would support it as being a safe modality for clinical use.
