Analysis of Ag specificity of TRC-gamma delta+ T cells in humans has been hampered by the fact that cloned lines of these cells expanded in IL-2 generally display high NK-like cytotoxic activity. A TCR-gamma delta+ CTL clone, isolated in IL-4, strongly lysed a specific stimulator cell, the EBV-transformed cell line JY, but failed to lyse K562 and other target cells sensitive for NK cell activity. Subsequent culture of this clone (CD124) in IL-2 induced high cytotoxic activity against the NK sensitive target cells. K562 cells were unable to induce the secretion of N alpha-benzyloxycarbonyl-L-lysine thiobenzyl ester [(BLT)-serine esterase] or influx of Ca2+ ions in clone CD124 cultured in either IL-4 or IL-2. In contrast, JY cells induced high BLT-serine esterase secretion and an increase of cytosolic Ca2+ levels. By using a combination of a 51Cr-release assay and a BLT-serine esterase secretion assay, the reactivity of clone CD124 against a limited number of target cells was analyzed. CD124 which expresses HLA-A2 and -B7, recognized an Ag shared by JY (HLA-A2; B7; C blank; DR4,6) and one haplotype expressed by the cell line
SPS
(HLA-A1; B14; Cw6; DR4). The only specificity shared by
SPS
and JY was HLA-DR4. However, clone CD124 failed to lyse 5 other HLA-DR4+ target cells. The cytotoxic activity of clone CD124 was inhibited by the class I MHC specific mAb W6/32 and the anti-beta 2m mAb A88, but not, or only marginally, by the anti HLA-DQ mAb SPV-L3 or the anti-HLA-DR mAb 135. These data strongly suggest that clone CD124 recognizes a class I MHC Ag different from HLA-A, -B, or -C.

A sensitive and selective bioanalytical method for simultaneous determination of diclofenac and oxybuprocaine in human aqueous humor using reversed-phase HPLC and electrochemical detection is described. Chromatographic separation was achieved by using a Regis

100 RP-8 column (5 microns; 150 x 4.6 mm I.D.). This support is coated with a hydrophilic polyoxyethylenepolymer. It allows protein-containing samples to be injected directly onto the column. The electrochemical detector permit a detection limit of 500 pg diclofenac per ml (daily relative standard deviation 6.3%) and 50 ng oxybuprocaine per ml (daily R.S.D. 2.6%), respectively. Results of administered and measured drug-concentrations in time dependent decrease are presented.

Participants were 117 patients attending a specialized anxiety disorders unit for treatment. While all three measures were able to detect differences between social phobic patients and patients with panic disorder with or without agoraphobia, a logistic regression analysis showed that the SPAI, but not the
SPS
and SIAS, was a significant predictor of membership of the social phobia group. Receiver operating characteristic (ROC) analysis also showed that the SPAI was the better measure for discriminating between social phobia and panic disorder with and without agoraphobia. Analysis of the sensitivity, specificity and positive and negative predictive power of the measures at the optimum cutoff scores produced by the ROC analysis are presented.

We suggest the variation of charmonium production rate with Feynman x(F) in heavy ion collisions as a novel and sensitive probe for the properties of the created matter. Final state interactions with the comoving matter create a minimum at x(F) = 0, which is especially deep and narrow if a quark-gluon plasma is formed. While a strong effect is predicted at
SPS
, at the RHIC energy it overlaps with the expected peak formed by shadowing effects and needs comparison with pA data. We predict a steep dependence on centrality and suggest that this new probe is complementary to the dependence on transverse energy, and is more sensitive to a scenario of final state interactions.

The possible formation of a multienzyme complex between sucrose (Suc)-phosphate synthase (
SPS
) and Suc-phosphate phosphatase (SPP) was examined by measuring the rates of Suc-6-phosphate (Suc-6-P) synthesis and hydrolysis in mixing experiments with partially purified enzymes from spinach (Spinacia oleracea) and rice (Oryza sativa) leaves. The addition of SPP to
SPS
stimulated the rate of Suc-6-P synthesis.
SPS
inhibited the hydrolysis of exogenous Suc-6-P by SPP when added in the absence of its substrate (i.e. UDP-glucose) but stimulated SPP activity when the
SPS
substrates were present and used to generate Suc-6-P directly in the reaction. Results from isotope-dilution experiments suggest that Suc-6-P was channeled between
SPS
and SPP. A portion of the
SPS
activity comigrated with SPP during native polyacrylamide gel electrophoresis, providing physical evidence for an enzyme-enzyme interaction. Taken together, these results strongly suggest that
SPS
and SPP associate to form a multienzyme complex.

PMID:Physical and Kinetic Evidence for an Association between Sucrose-Phosphate Synthase and Sucrose-Phosphate Phosphatase. 1222 2
The effect of leaf-to-air vapour pressure deficit (VPD) was studied in well-watered, potted, 1-2-year-old plants of the leguminous tree P. juliflora grown outside in northern India. The long-term responses to VPD were analysed from diurnal and seasonal variations in gas exchange parameters measured in two cohorts of leaves produced in February and July, respectively. In general, inhibitory effects of high VPD were visible only when the VPD level exceeded a threshold of >3 kPa. There was a substantial decline in net photosynthesis rate and stomatal conductance at high VPD >4 kPa and transpiration showed a decrease in steady-state rate or feedforward response to VPD. The feedforward responses were visible in all seasons, although the plants were exposed to a wide range of VPD during the year and leaf relative water content was constant. The maximum quantum efficiency of PSII measured predawn was constant (around 0.8) in all seasons except summer. Short-term experiments showed that, although gas exchange was severely affected by high VPD in the leaves of both cohorts, the plant maintained a constant, water use efficiency in different seasons. High VPD also caused reductions in Rubisco activity, affecting carboxylation efficiency, and reductions in sucrose and starch content due to a decrease in the activity of
sucrose-phosphate synthase
. However, the relative quantum yield of PSII and electron transport rates measured at 1500 micromol m(-2) s(-1) were unaffected by increasing VPD, indicating the presence of a large alternative sink possibly, photorespiration. The overall results showed that P. juliflora can withstand high VPD by reducing metabolic activity and by effective adjustments in the partitioning of electron flow between assimilation and non-assimilation processes, which, in turn, imposed a strong limitation on the potential carbon gain.

The constitutive cytosolic expression of a yeast ( Saccharomyces cerevisiae ) invertase within potato ( Solanum tuberosum ) tubers has previously been documented to produce a dramatic metabolic phenotype in which glycolysis, respiration and amino acid synthesis are markedly enhanced at the cost of starch synthesis. These transgenic lines were further characterised by a massive cycle of sucrose degradation and resynthesis via
sucrose-phosphate synthase
. We have recently developed a B33 patatin driven alc gene construct allowing tight chemical control of gene expression following supply of acetaldehyde with minimal pleiotropic effects of the inducing agent on metabolism. This construct was used for chemical induction of the yeast invertase gene after 10-weeks growth to dissect the complex metabolic phenotype obtained after constitute expression. Inducible expression led to increased invertase activity within 24 h in well-defined areas within growing tubers. Although the sucrose levels were reduced, there was no effect on the levels of starch whilst levels of many amino acids decreased. Labelling experiments revealed that these lines exhibited increased rates of sucrose cycling, whereas rates of glycolysis and of starch synthesis were not substantially changed. From these results we conclude that sucrose cycling is stimulated in response to a short-term increase in the rate of sucrose mobilisation, providing evidence for a role of sucrose cycling as a buffering capacity that regulates the net rate of sucrose usage. In contrast, the dramatic increase in hexose-phosphate levels and the switch from starch synthesis to respiration seen on the constitutive expression of the invertase was not observed in the inducible lines, suggesting that this is the result of cumulative pleiotropic effects that occurred when the transgene was expressed throughout development.

As measured 7, 14, and 21 days after the application of 10(-2) M vanadyl sulfate solution to the foliage of 4.5-month-old sugar beet plants, significantly less growth of the leaves and an increase in the sucrose content of the storage root resulted. Accompanying these alterations were a higher rate of carbon dioxide fixation, a lower rate of respiration, and a decreased rate of nitrate reductase, glutamic-pyruvic transaminase, phosphatase, and invertase activity. The enzymes of sucrose synthesis, sucrose synthetase,
sucrose phosphate synthetase
and uridine diphosphate glucose-pyrophosphorylase were stimulated. The content of reducing sugar, nitrite N, amino acids and protein was less, and that of nitrate N was greater in the vanadium-treated plants. In the majority of cases the greatest magnitude of change occurred during the first 7 days following treatment. The changes in growth and chemical composition are believed to be closely related to the stimulation or inhibition of the various enzymes by vanadyl sulfate.

The effects of water stress and CO(2) enrichment on photosynthesis, assimilate export, and sucrose-P synthase activity were examined in field grown soybean plants. In general, leaves of plants grown in CO(2)-enriched atmospheres (300 microliters per liter above unenriched control, which was 349 +/- 12 microliters per liter between 0500 and 1900 hours EST over the entire season) had higher carbon exchange rates (CER) compared to plants grown at ambient CO(2), but similar rates of export and similar activities of sucrose-P synthase. On most sample dates, essentially all of the extra carbon fixed as a result of CO(2) enrichment was partitioned into starch. CO(2)-enriched plants had lower transpiration rates and therefore had a higher water use efficiency (milligrams CO(2) fixed per gram H(2)O transpired) per unit leaf area compared to nonenriched plants. Water stress reduced CER in nonenriched plants to a greater extent than in CO(2)-enriched plants. As CER declined, stomatal resistance increased, but this was not the primary cause of the decrease in assimilation because internal CO(2) concentration remained relatively constant. Export of assimilates was less affected by water stress than was CER. When CERs were low as a result of the imposed stress, export was supported by mobilization of reserves (mainly starch). Export rate and leaf sucrose concentration were related in a curvilinear manner. When sucrose concentration was above about 12 milligrams per square decimeter, obtained with nonstressed plants at high CO(2), there was no significant increase in export rate. Assimilate export rate was also correlated positively with
SPS
activity and the quantitative relationship varied with CER. Thus, export rate was a function of both CER and carbon partitioning.
