Four enzyme activities related to glucose metabolism, i.e. those of glucose-6-phosphate dehydrogenase (G6PDH; EC 1.1.1.49), lactic dehydrogenase (

; EC 1.1.1.27), pyruvate dehydrogenase complex (PDC) and
citrate synthase
(CS; EC 4.1.3.7) were estimated in posterior root ganglion cells (PRGCs) of the spinal cord in patients suffering from olivopontocerebellar atrophy (OPCA), amyotrophic lateral sclerosis (ALS), and Duchenne muscular dystrophy (DMD) by means of the NAD, NADP and CoA cycling methods. In ALS and DMD, the enzyme activities examined were within normal ranges. In OPCA, PDC activity was significantly reduced and

Weanling and adult rats were subjected to left ventricular pressure overload induced by abdominal aortic constriction. At 5 days or 5 weeks postsurgery, the left ventricle (LV) was dissected, weighed, and metabolic marker enzyme activities (mumole/g/min) of tissue homogenates were measured. Enzymes representing glycolytic (phosphofructokinase (PFK] and mitochondrial (
citrate synthase
(CS) and malate dehydrogenase (MDH] metabolisms were evaluated. Five days of pressure overload had detectable, but statistically nonsignificant effects on left ventricles of both weanling and adult rats. Sustained pressure overload (5 weeks) increased LV weight by 52 and 39% in weanling and adult rats, respectively. PFK activity was 24 +/- 1 (mean +/- SE) in control weanlings and was unaltered in any of the other groups.
LDH
isoenzyme composition was estimated by substrate inhibition (ratio 0.33/10 mM pyruvate). With normal heart development, the
LDH
ratio increased from 1.89 +/- 0.06 to 2.03 +/- 0.08. Pressure overload had no influence on the adult
LDH
ratio. Developmental
LDH
responses were not observed in weanling LV after 5 weeks of aortic constriction (1.74 +/- 0.06). The product of CS activity and LV weight was used to estimate mitochondrial mass in the ventricle. Mitochondria accumulated at a rate of about 5% increase per day over the intervening 5-week period of normal heart growth. Pressure overload for 5 weeks in weanling rats elicited net accumulation of mitochondria at a rate of about 9% increase per day. Mitochondrial accumulation in the adapting adult rat heart amounted to less than 1% increase per day. The results indicate that qualitative and quantitative differences exist between young and adult animals in their heart enzyme adaptive responses to pressure overloading. Divergent metabolic adaptations may contribute to heart functional differences in the enlarged heart of weanlings and adults.

The renal cortex already differs metabolically from the medullar structures on the 10th day of life. It displays a high activity of aerobic breakdown of both fatty acids and carbohydrates. Its metabolic capacity further increases up to the 30th day of life. 3. The outer medullar structure is not grossly different from the inner medulla on the 10th day of life. Further it differentiates into a highly aerobic tissue mainly able to utilize carbohydrates. It can, however, to some extent, also utilize fatty acids aerobically and produce lactate from carbohydrates anaerobically. 4. The inner medullar structure is best equipped to utilize carbohydrates by anaerobic glycolysis, forming lactate. This feature is already pronounced on the 10th day of life, its capacity increases to some extent during postnatal development, being highest between the 10th and the 60th day of life.

In an earlier study, we reported close relationships between marathon running performance and the running velocity (V) at which the "onset of blood lactate accumulation" (VOBLA) occurs in a group of marathon runners. Using biopsy material from the m. vastus lateralis of the same subjects (n = 19), we have evaluated the relationship of VOBLA to different muscle enzyme activities together with muscle fiber composition and capillary density in the present study. The activities of lactate dehydrogenase (
LDH
EC 1.1.1.27), phosphofructokinase (PFK EC 2.7.1.11), and
citrate synthase
(CS EC 4.1.3.7) were determined. VOBLA was negatively correlated to
LDH
(r = -0.54) and PFK/CS (r = -0.68). Using multiple regression analysis, the PFK/CS ratio together with the capillary density accounted for 61% of the variation in VOBLA. Absolute training kilometrage was the most significant variable measured and accounted for 77% of the variation in VOBLA. Subjects were divided into elite runners (n = 6) and nonelite runners (n = 13) for an additional analysis of the relationship between VOBLA and the ratios of PFK/CS or
LDH
/CS. Significant relationships between VOBLA and the ratios were observed only in the nonelite runners (r = -0.77 and -0.66, respectively). The vertical distances between the regression lines for these two subject groups could not be explained only on the basis of the enzyme activity ratios. A greater adaptation to fat combustion in the elite runners might explain the disproportionally high VOBLA in relation to the PFK/CS or
LDH
/CS activity ratios.

Although some of these changes as well as the differences between the cultivation at 0 degree C and 37 degrees C, were statistically significant, they were not greater than the variations between different samples of any tissue taken at different times. They were not, therefore considered to be of major significance. However, all the aggregates exhibited "profiles" characteristic for the nervous tissue, with relatively very high activity of HK, high activity of MDH and CS (carbohydrate breakdown) and low activity of GPDH and HOADH (lipid catabolism).

Muscle hypertrophy was induced in the soleus muscle of young rats by tenotomy of the gastrocnemius and plantaris muscles. Three and 7 days afterwards the sciatic nerve was sectioned. The loss of weight of muscles subjected to this combined procedure three days after denervation was 30-40%. , CS) were determined 3 days after denervation, 3, 7 and 10 days after hypertrophy had been induced and 3 days after denervation of hypertrophying muscles on day 3 and 7. Normal non-operated rats of corresponding body weight served as controls and their enzyme activities were estimated on the same day. In the course of muscle hypertrophy, the 4 lysosomal enzyme activities increased progressively. Although 3 days' denervation of control muscles did not alter lysosomal enzyme activities, denervation of hypertrophying muscles greatly enhanced the activity of these enzymes. Enzymes of energy metabolism were affected to a lesser degree. The results suggest that denervation of hypertrophying muscles causes more extreme changes in muscle weight and lysosomal enzyme activities than denervation alone. The possible implications of this finding are discussed in relation to the rapid atrophy.

The metabolic activities of skeletal muscles were studied in male rats exposed to hypobaric-hypoxia at about 550 Torr for 8 h per day for 2 weeks. Rats were divided into three groups; control (normoxic control), diurnal hypoxic (DH) exposure, and nocturnal hypoxic (NH) exposure groups. The changes in body weight and daily diet intake of the NH group were lower than the other groups (p < 0.01). The weights of fat in the abdominal cavity of both NH and DH groups were less than that of the control group. The red blood cell count, hemoglobin concentration, and hematocrit values were significantly increased in the hypoxic groups. The plasma glucose level in the NH group was significantly less than the control group (p < 0.05). The lactate dehydrogenase/

/CS) activity ratios in the skeletal muscle tended to be lower in both hypoxic groups than in the control group. The swimming times to exhaustion at mild and high intensities that were measured after 2 weeks, loaded with a weight equivalent to 2.5% of the body weight, improved in the DH group. There were insignificant differences in the metabolic activity of skeletal muscles and blood characteristics between the NH and DH groups, but endurance swimming times in the DH group tended to be improved as compared to the NH group. We conclude that the DH group became competent in endurance work, which is believed to be driven from the combined effects of increased O2 transport capacity of the blood and enhanced O2 utilization capability by mitochondrial enzyme activity.

To study the effects of nandrolone laureate (19-nortestosterone) on muscle hypertrophy and concentration of androgen receptors (AR), biopsy specimens were taken from the middle gluteal muscle of 6 Finnhorse trotters (geldings and mares) undergoing training before, immediately after, and 13 weeks after a 14-week treatment with nandrolone. Another 6 similarly trained horses served as controls. An additional 10 mares and 10 geldings were used to study annual variation in muscle concentration of AR. AR was immunohistochemically localized in the nuclei. AR concentration remained constant during the first 14 weeks of the study, but increased significantly during the 13-week follow-up period in both groups. This finding can be explained by the annual variation in AR. In the anabolic steroid (AS)-treated horses, but not in the controls (C), the cross-sectional area of the type I fibres increased significantly during the treatment period, and the percentage of type IIA fibres correlated positively with AR concentration at the end of nandrolone treatment. In the AS group, the concentration of DNA decreased during the 13-week follow-up period, and the percentage of H-chains in the isoenzymes of
LDH
increased. Protein concentration increased in both groups during the follow-up period. Glycogen content and the activity of
citrate synthase
in muscle during the study remained unchanged. It can thus be concluded that AS produce differing effects on type I and type II fibres, and the AR concentration in equine muscle may contribute to the change observed in the middle gluteal muscle.
