The fatigue characteristics and the activities of oxidative and glycolytic enzymes were determined in tetrodotoxin (TTX)-induced disuse muscles and in muscles of animals recovering from TTX-induced disuse (TTX-
rec
). In addition, the effects of additional daily exercise (grid-climbing and swimming) on the fatigue and metabolic profiles of muscles from TTX-
rec
and control animals were investigated. The activities of
citrate synthase
(CS), phosphofructokinase (PFK), and alpha-glycerophosphate dehydrogenase (alpha-GPD) were depressed following 28 days of inactivity produced by the chronic neural application of TTX. The response of these muscles to a pattern of stimulation that has been used to classify fast-twitch motor units according to their fatigability (6) (330 ms, 40 Hz, l/s, 4 min) was not affected to any great extent by inactivity, except for a loss in the ability to summate or maintain forces during each 330-ms burst, as fatigue developed. After 28 days of recovery, the concentration of CS had returned to normal, whereas the concentrations of PFK and alpha-GPD remained depressed. TTX-

muscles, on the other hand, appeared more resistant to fatigue than control muscles, based on several indices of muscle fatigue. Control and TTX muscles responded similarly to daily training. Swimming but not climbing increased the activity of CS and the fatigue resistance of the muscle. Neither exercise influenced the activity of PFK and alpha-GPD. Although the activity of CS was influenced by the level of neuromuscular usage, the former did not appear to play a dominant role in determining the fatigue resistance of the muscle, emphasizing the need to consider other factors as primary determinants of muscle fatigue.

Muscle biopsy samples were collected from the middle gluteal muscle of seven horses undergoing a nine-month endurance training programme. Samples were collected before the programme began and again after three, six and nine months of training. A fifth sample was collected three months after training ceased. Serial muscle sections were reacted histochemically for myosin adenosine triphosphatase after either acid (pH 4.3 and 4.6) or alkaline (pH 10.3) pre-incubation, and muscle fibres identified as type I, IIA, IIB or IIC. The oxidative capacity of individual fibres was assessed, using the reduced nicotinamide dinucleotide tetrazolium reductase stain, and the number of intermyofibrillar capillaries adjacent to each fibre was counted after staining, using the alpha-amylase periodic acid Schiff technique. , 3-hydroxy acyl CoA dehydrogenase and lactate dehydrogenase as markers of end terminal oxidative, beta oxidative and glycolytic potential, respectively. There was an increase in the percentage of type IIB fibres having high nicotinamide dinucleotide tetrazolium reductase staining after three months training. This increase persisted throughout the period of training and during the period without training. There was an increase in the number of capillaries adjacent to type IIB fibres after six and nine months training. These had returned to near pre-training numbers after three months without training. There were increases in the activities of
citrate synthase

Muscle cell fiber types in gracilis, rectus femoris, and long head of triceps brachii muscles of ferrets and dogs were identified on serial sections stained for myosin ATPase after preincubation at pH values of 9.8, 4.6, and 4.3 and for NADH-tetrazolium reductase (NADH-TR) activity. Although fiber types I and II were identified, the ATPase stain did not demonstrate classic type IIA/IIB fiber differences in either species. However, two type II fiber subtypes could be distinguished in the ferret because they differed slightly in staining intensity with ATPase at pH 4.3 and markedly with NADH-TR. One ferret type II fiber (designated II dark or IID) was smaller, slightly darker on ATPase, more oxidative on NADH-TR, and comprised more muscle volume than the other type II fiber (designated II light IIL). The IID fibers of ferret may represent the IID/X fibers of other authors. Both ferret type II fiber subtypes stained darker at pH 4.3 than canine II fibers. The NADH-TR staining indicated high oxidative activity in canine and ferret type I fibers. In contrast, type II fibers in the dog and IIL fibers in the ferret were moderately oxidative. Canine type IIC fibers were intermediate between type I and type II, whereas in the ferret, type IIC fibers were highly oxidative, as were type IID fibers. Ferret muscles are more oxidative than canine muscles according to NADH-TR staining. Also, ferret muscles possess 40-100% higher
citrate synthase
activity as compared to canine muscles.

Biopsies of the gluteus medius muscle were taken at three different depths from 36 endurance horses aged 8.42 +/- 2.85 years and of both sexes. Twenty of the horses were considered to be excellent performers on the basis of the mean speed of their three fastest records in endurance events over the previous two or three years, whereas 16 were moderate performers. The biopsy samples were analysed for the activities of the enzymes
citrate synthase
(an indicator of citric acid cycle activity), 3-OH-acyl-CoA-dehydrogenase (an indicator of lipid oxidation) and lactate dehydrogenase (an indicator of anaerobic metabolism). The 20 excellent performers had higher activities of
citrate synthase
(P < 0.001) and 3-OH-acyl-CoA-dehydrogenase (P < 0.02) than the 16 moderate performers. The activities of
citrate synthase
and 3-OH-acyl-CoA-dehydrogenase increased by 65 per cent and 75 per cent, respectively, and the activity of lactate dehydrogenase decreased by 23 per cent in the samples taken at successively greater depths. There was a strong linear relationship between the ratios of the activities of lactate dehydrogenase/
citrate synthase
and lactate dehydrogenase/3-OH-acyl-CoA-dehydrogenase and the depth from which the samples were taken for both performance groups (P < 0.001). The intercepts of the regression lines were higher in the moderate than in the excellent performers (P < 0.001 and P < 0.01 for the two ratios), showing that the endurance horses with the better performance record had a greater aerobic capacity and a relatively lower anaerobic capacity in the gluteus medius muscle than the horses with a poorer record.

In order to determine the effects of chronic, high-altitude hypoxia on the ovine fetal heart, we exposed pregnant ewes to 3,820 m beginning at 30 days gestation. We previously showed that following approximately 110 days of hypoxia the fetal heart showed significant reduction in cardiac output (76% of control) and contractility, and elevated levels of
citrate synthase
and lactate dehydrogenase. To investigate ultrastructural influences on these observed physiologic changes at altitude, we hypothesized that the volume densities of myofibrils and mitochondria, and glycogen content would be reduced in the ovine fetal heart and that this may contribute to contraction and cardiac output deficits in hypoxia. Mitochondria and myofibril volume density were determined by standard point-counting techniques and glycogen content was determined by biochemical analysis. The glycogen content from the hypoxic right ventricle (4.8 +/- 0.3%) was significantly lower than in control right ventricle (6.8 +/- 0.5%) and both left ventricles (hypoxia, 7.2 +/- 0.5; control, 7.8 +/- 0. 4%). Total mitochondrial volume density was also significantly reduced following hypoxia (15.5 +/- 0.7%) compared to controls (16.9 +/- 0.4%). As is common in the ovine fetal heart, the myofibril volume density of the right ventricle from both groups was significantly higher than the left ventricle (RV, 58.6 +/- 1.6; LV 54.3 +/- 0.9%). However, it was not different between control and high altitude. In support of our hypothesis, we may speculate that deficits in the quantity of myocyte glycogen and mitochondria contribute to the observed reduction in cardiac output and contractility, despite the upregulation of
citrate synthase
and lactate dehydrogenase. In contrast, myofibril volume density was unchanged.

Between October 1999 and February 2000, 691 blood samples examined routinely for either haematological or virological assessment were screened by culture for the presence of Bartonella species. They came from 615 animals: 360 cats, 211 dogs, 27 horses, 16 cattle and a gorilla. The samples were incubated for long periods on 10 per cent horse blood agar at 37 degrees C in an atmosphere containing 5 per cent carbon dioxide. Isolates were obtained from 35 samples from 34 (9.4 per cent) of the cats, but not from any of the other animals. Comparison of
citrate synthase
gene sequences from the isolates indicated that they were all Bartonella henselae. Analysis of 16S rRNA gene fragments indicated that 30 of the cats were infected solely with B henselae genotype II, two were infected solely with B henselae genotype I and two were infected with both genotypes.
