The quality and stability of sibling relationships during adulthood were assessed by comparing TAT themes of sibling affiliation, conflict, and separation at the period of active child-rearing (CR) and the empty nest (EN). Thirty men and thirty women, divided equally between the two life stages, were randomly selected from a pool of individuals who had a same-sex, closely-spaced sibling. The Sibling Thematic Apperception Test (S-TAT) was developed to elicit underlying feelings about the relationship, because traditional methods of assessment have failed to tap its negative qualities. Contrary to previous findings, results showed no differences in affiliation or conflict theme frequencies between the CR and EN periods. Separation themes, however, were more frequent during the CR than the EN period, suggesting that concerns over separation from siblings reflect a developmental process which might be related to bonding with the family of procreation. Although there were no sex differences within life periods, women's stories about sisters across life period had more conflict themes than did men's stories about brothers.

Is there a covert, middle-age shift towards passivity and lowered ego energy? Age differences were investigated in dreams recalled from REM sleep by fifty-eight well-educated men age twenty-seven to sixty-four, and also in their TAT stories and dreams recalled from sleep at home. There was no confirmation of previous findings, although there was evidence of a slight age-related decline of aggression in dreams. These results raise the possibility that previous findings reflect cohort differences rather than age changes. Dreams and TAT stories failed to correlate sufficiently to warrant any assumption that they are interchangeable or that they tap covert processes.

Cell-penetrating peptides (CPP) or membrane-translocating peptides such as penetratin from Antennapedia homeodomain or TAT from human immunodeficiency virus are useful vectors for the delivery of protein antigens or their cytotoxic (Tc) or helper (Th) T cell epitopes to antigen-presenting cells. Mice immunized with CPP containing immunogens elicit antigen-specific Tc and/or Th responses and could be protected from tumor challenges. In the present paper, we investigate the mechanism of class I and class II antigen presentation of ovalbumin covalently linked to penetratin (AntpOVA) by bone marrow-derived dendritic cells with the use of biochemical inhibitors of various pathways of antigen processing and presentation. Results from our study suggested that uptake of AntpOVA is via a combination of energy-independent (membrane fusion) and energy-dependent pathways (endocytosis). Once internalized by either mechanism, multiple tap-dependent or independent antigen presentation pathways are accessed while not completely dependent on proteasomal processing but involving proteolytic trimming in the ER and Golgi compartments. Our study provides an understanding on the mechanism of antigen presentation mediated by CPP and leads to greater insights into future development of vaccine formulations.

The Social Cognition and Object Relations Scales-Global Rating Method (SCORS-G) contains 8 scales for coding narrative content. This study explores the factor structure of this measure using college (n = 171), outpatient (n = 239), and inpatient (n = 78) samples. Participants told stories to the Thematic Apperception Test (TAT; Murray, 1943) cards. Stories were transcribed and coded by blind raters using the SCORS-G. Cases were randomly assigned to an exploratory or validation group. Exploratory factor analysis with the exploratory group suggested 2- and 3-factor models. The Emotional Investment in Relationships (EIR) scale did not obtain a primary loading on any factor and was not included in subsequentmodels. After modifications, confirmatory factor analysis indicated good-to-adequate fit for 2- and 3-factor models. Both models showed good fit in the validation group and met criteria for invariance across models. Findings indicated that some SCORS-G scales tap cognitive-structural elements, whereas others assess affective-relational components of narratives. We found mild support separating the affective-relational scales in terms of internal representations for the self and others and relationships. The results reported here indicate that clinicians and researchers can calculate a separate cognitive-structural composite score and an affective-relational composite score when using the SCORS-G to rate TAT stories.
