Oxidized and reduced manganese cytochromes c, Mn Cyt c+ and Mn Cyt c, have been synthesized. Mn Cyt c+ and Fe Cyt c+ have identical electrophoretic and ion exchange mobilities. Mn Cyt c+ does not bind F-, CN-, or N3- ions; Mn Cyt c does not bind CO or O2. Mn Cyt c is very rapidly autooxidized by O2 even at -50 degrees. The manganese ion is readily dissociated from Mn Cyt c at acidic pH values. Both Mn Cyt c and Mn Cyt c+ are high spin complexes with 3d5 S = 5/2 and 3d4 S = 2 electronic configurations, respectively. The epr spectrum of Mn Cyt c is rhombic with (formula: see text). Both oxidized and reduced Mn Cyt c react with NO; the former reaction is reversible and the product has the following epr spectral parameters: (formula: see text). There is no superhyperfine interaction observable with the NO ligand, and the unpaired electron density is estimated to be mostly in the metal ion d xy orbital. The structure is best formulated as Mn Cyt c (NO)+. The half-reduction potential of Mn Cyt c is + 60 +/- 40 mV. It is neither oxidized by
cytochrome oxidase
nor reduced by NADH,
NADPH
, or succinate cytochrome reductase. These physical, chemical, and enzymic properties of manganese cytochromes c suggest a five-coordinate metalloporphyrin prosthetic group with the manganese ion situated significantly out-of-plane toward the side of His-18.

An improved procedure for the preparation of cobalt-cytochrome c has been developed. Various factors influencing the cobalt insertion process are discussed. The optical spectra of cobalt-cytochrome c suggest a six-coordinated species. The spectral shifts occurring with oxidation-reduction are compared with those observed for deoxy-cobaltohemoglobin and ferrocytochrome c and attributed to the effect of d(z2) electron on stereoelectronic interactions between the axial ligands and the porphyrin pi systems. Cobalt-cytochrome c has Em,7 = -140 +/- 20 mV as compared to an Em,7 of +250mV for ferrocytochrome c. An explanation for this negative Em,7 is offered. Cobaltocytochrome c is oxidized by
cytochrome oxidase
at about 45% of the rate for native cytochrome c. On the other hand cobalticytochrome c was not reduced by microsomal NADH or
NADPH
cytochrome c reductase nor by mitochondrial NADH or succinate cytochrome c reductase. It appears that the integrity of the reductase binding site is destroyed and the oxidase binding site has been modified by cobalt substitution.

The presence and localization of the enzymes in various periods of development of the redia were detected with histochemical methods. Out of the studied oxidases and dehydrogenases only

was found to be absent from the stages of young rediae. It was ascertained that the redia uses all three paths of release of energy i.e. the glycolytic, Krebs, and pentose cycles, glycolysis being presumably the principal mode of energy production.

The object of the study was the investigation of the occurrence and localization of oxidative enzymes in the 4th and 5th larval stages of the liver fluke, i. e. in the cercaria and metacercaria. The following enzymes were detected:

tetrazolium reductases/diaphorases) as well as succinate, isocitrate, malate, lactate, alpha-glycerophosphate, glyceraldehyde phosphate, glucose-6-phosphate, 6-phosphogluconate, beta-hydroxybutyrate, L-glutamate and alcohol dehydrogenases. The occurrence and localization of the enzymes were investigated histochemically in the cercaria still unreleased from the snail tissues, in the free natatorial cercaria, and in the encysted specimen, i.e. metacercaria. Among the enzymes studied only peroxidase was found to be absent from the cercaria and metacercaria, the latter larva being deprived of alcohol and L-glutamate dehydrogenases as well. The aerobic path, i.e. the Krebs cycle, was ascertained as the principal mode of obtaining energy in the free natatorial cercaria and metacercaria, and glycolysis as the main energy path for the undetached larva. The analysis of the above-mentioned oxidative enzymes was the basis for the description, within the available range, of the function and metabolism of individual organs of cercaria in different periods of its development. In the recapitulation the author discusses the effect of the parasitic way of life of the larval forms of Fasciola hepatica on their energy metabolism.

1. Cells of the hydrogen bacterium Alcaligenes eutrophus are broken by gentle lysis using lysozyme treatment in hypertonic sucrose followed by osmotic shock. By this method, 93% of the in vivo activity of the H2 oxidase is recovered and the ATPase remains particle bound. In contrast, cell disruption in a French pressure cell diminishes the in vivo activity of the H2 oxidase by 50% and solubilizes the bulk of the ATPase. 2. The bacterium contains a periplasmic cytochrome c with bands at 418, 521 and 550 nm (difference spectrum). In addition to

, succinate, formate and endogenous substrate (NAD linked) at rates 2--3-fold higher than membranes obtained from cells disrupted in a French pressure cell. With the exception of the H2 oxidase all oxidase activities in lysozyme membranes are sensitive to carbonylcyanide m-chlorophenylhydrazone (20-100% stimulation of oxygen uptake). 4. The cytoplasmic fraction contains a B-type cytochrome with absorption maxima at 436 and 560 nm, capable of combining with CO; it contains non-covalently bound protohaem. In alkaline solutions a spectral transition to the haemochrome type with bands at 423, 526 and 556 nm occurs. The addition of NADH to an aerobic suspension of this cytochrome elicits new absorption maxima at 418, 545 and 577 nm (difference spectrum), which are believed to represent an oxygenated form of the reduced cytochrome.

-nitrobluetetrazolium reductases. This is true of preparations exhibiting heme aa3 to protein ratios considered indicative of an excellent purity. An apparent association of
cytochrome-c oxidase
and one or more of the contaminants persists through immunodiffusion and nondenaturing electrophoresis and, in addition, in one instance copurification of NADH-K3Fe(CN)6 reductase and
cytochrome-c oxidase
to a constant ratio of specific activities was demonstrated. Cytochrome-c oxidase can be freed of the contaminants by equilibration with an NAD+-affinity matrix. As aconcomitant of equilibration with the matrix, the KM of
cytochrome-c oxidase
for ferrocytochrome-c is invariably decreased. Rat constants at low ferrocytochrome-c concentrations are consistently enhanced in all oxidase preparations upon equilibration with the NAD+ matrix. However, the effects of such equilibrations on the extrapolated Vmax varies from one preparation to another. Polyacrylamide gel electrophoresis in SDS-urea systems establishes that each of the preparations contains a minimum of three contaminants, each of an apparent formula weight of greater than 40,000 Daltons. NADH-NBT reductase was found to have a formula weight of approximately 46,000 Daltons. Their properties establish that NADH-K3Fe(CN)6 and NADH-NBT reductases are separate proteins; the separate identity of
NADPH
-NBT reductase has not yet been determined.

In the paper the author is concerned with the histochemical estimation of the metabolic adaptation of the heart muscle of albino rats during an early experimental alloxan diabetes. It has been found that the state of experimentally produced insulin deficiency directly influences metabolism of the heart muscle and the changes observed in the histochemical reactions prove this. An increase in the intensity of histochemical reactions concerns the PAS-positive reaction and the reactions to the NADH and

tetrazole reductase activities. Alkaline phosphatase shows a decrease in the enzymatic activity, whose nature is transitional and reversible with regard to

The circum ventricular region of C. batrachus is highly vascular and the ependymal cells appear differently when stained with haematoxylin, silver impregnation and Golgi-Cox techniques. The ventricule has PAS and AF positive material and some ependymal cells themselves are PAS positive. Few AF positive peptidergic and several AF negative small neurons have liquor contacting terminals. Golgi-Cox preparations reveal a variety of forms among the tanycytes. Their basal processes which are barbed or studded with varicosities, usually end on blood vessels and other neuronal elements. These basal processes themselves are often seen in direct morphological contact. Smaller silver positive cells without basal processes are also evident. Some tanycytes have apical processes resembling broadened endfeet. Few neurosecretory tracts are Golgi-Cox positive and can be differentiated from the tanycytic processes by their smooth surface. Varying degrees of ascorbic acid activity are noticed inside the ventricle, among the tanycytes and in the neurons of the NLT. Some of the latter neurons have liquor contacting terminals as well. The ChE activity noticed in some parts of the ependyma and in some NLT cells suggest their probable differential cholinergic control. Presence of SDH,

in varying quantities in the ependymal cells suggests that they are metabolically active. Presence of MAO positive tracts bridging the subependyma and ventricle suggests the degradation of monoamines at these sites. The presence of various enzymes and the morphological relationship of the tanycytes described in this species are comparable to those of the mammals. It is significant as this species is reported to have a median eminence morphologically resembling the tetrapods.

In populations of cultured arterial endothelial and smooth muscle cells grown under the same conditions, we have measured the total activity per cell of 10 enzymes commonly used as "markers" for subcellular organelles: NADH: ferricyanide reductase, NADH:cytochrome c reductase (rotenone insensitive).

:cytochrome c reductase, alpha-glucosidase, 5'-nucleotidase, alkaline phosphodiesterase I,
cytochrome oxidase
, monoamine oxidase, cathepsin D, and N-acetyl-beta-glucosaminidase. Significant differences between the cell types were found for 7 of the 10 enzymes tested. The total activity of 5'-nucleotidase in cultured smooth muscle cells was 17 times that of cultured endothelial cells. Comparison of the activities in the two cell types freshly collected and in culture showed that the difference in 5'-nucleotidase in cultured cells is due principally to loss of activity from endothelial cells, suggesting that this activity is regulated differently in the two cell types. In both cell types cathepsin D activity rose during culture.

Activities of delta amino levulinic synthetase (DALS),
cytochrome oxidase
(E. C. 1.9.3.1.), NADH cytochrome b5 reductase (NADH red.),

red.), contents of cytochrome P450 (cyt. P450) and cytochrome b5 (cyt. b5), and levels of hemoglobin and hematocrit were studied in three groups of rats: a) malnourished, b) during recovery from malnutrition, and c) controls. During severe protein malnutrition blood levels of hemoglobin and hematocrit were found to be decreased as well as DALS's activity in homogenized bone marrow and liver. The activity of NADH red, and contents of cyt. P.450 and cyt. b5 in hepatic microsomes were also found significantly depressed. The microsomal activity of

red. as well as mitochondrial
cytochrome oxidase
did not present significant changes, since values obtained in malnourished rats were similar to those found for the control group. While recovering from malnutrition, when rats were fed a casein based diet (10 NDpCalo/o) supplemented with Fe and Cu, the hepatic enzymatic activities, the cytochrome contents of P450 and b5, and hematocrit experienced a spectacular increase, reaching towards the end of the refeeding period values which could be compared to those found in the control group. Nevertheless, DALS' activity in homogenized bone marrow and hemoglobin levels remained low. Results are discussed in relation to depressed activities and contents of enzymes, coenzymes, metabolites and subtrates involved in the hemoglobin synthesis in the rat bone marrow, during recovery from malnutrition.
