By differential hybridization, we have isolated 14 cDNA clones corresponding to genes that are more highly expressed in the flat revertant cell line R1 than in the parental human Ha-ras oncogene-transformed NIH/3T3 cell line (EJ-NIH/3T3). From cross-hybridization experiments, we determined that 5 sequence families accounted for the 14 clones. DNA sequencing revealed that four out of five selected cDNA clones represented mitochondrial genes (cytochrome b, cytochrome c oxidase subunit II, NADH dehydrogenase subunits 1 and 4, respectively), whereas one cDNA clone was homologous to the alpha 2 (type I collagen gene. Although a Southern blot analysis of the studied cell lines showed similar copy numbers of mitochondrial genomes, the transcript levels of the mitochondrial genes were high in R1, intermediate in NIH/3T3 and low in EJ-NIH/3T3 and partially revertant R2 cell lines. alpha 2 (type I) collagen mRNA levels were high in R1 and NIH/3T3, intermediate in R2 and low in EJ-NIH/3T3 cells. These results suggest that a complex alteration of the expression of mitochondrial and extracellular matrix components may be closely associated with the flat reversion of the transformed cells.

The histochemical profile of intrafusal fibres of muscle spindles and muscle spindle capsule were analysed in normal sural muscles of a rat. Weak activity of oxidative enzymes (NADH diaphorase, succinic dehydrogenase, lactate dehydrogenase) and creatine phosphokinase were found in bag1 and bag2 fibres, but the oxidative enzyme activity was moderate in chain fibres. High phosphorylase activity was demonstrated in bag fibres as well as in chain fibres. No differences could be detected in adenosine desaminase activity between these various intrafusal fibres. In the outer capsule of muscle spindles, high amount of type III collagen and elastin, but only small amount of type I collagen and fibronectin could be observed.

The neurons of whole cardiac atria were stained using a NADH-diaphorase technique in young adult (3 months old) (GI) and aging rats (20 months old) (GII). Light microscopy revealed differences in the appearance of the neurons in the two groups. In GI, most ganglia contained 50-100 neurons while in GII, most ganglia usually contained 20 neurons. The mean total number of neurons in the atria of GII was 245+/-31, i.e. only 23% of the mean value in GI (1086+/-203). The mean size of the ganglionic neurons (area of maximum cell profile) was 702 microm2 in GI and 1065 microm2 in GII. Histological sections of the ganglia revealed that a capsule of collagen fibers sheaths each ganglion in both groups. In GII, the density of collagen fibers increases in the capsule and in the septa within the ganglia; yellow or red, type I collagen fibers predominate in this group. No elastic fibers were present in the cardiac ganglia of either group. It is suggested that in aging rats, structural changes and reorganization of the remnant neurons accompany neuron reduction.

A quantitative morphometric evaluation of the intramural plexus of the urinary bladder of adult and aged guinea-pigs was performed by histological analysis, scanning electron microscopy, and hystochemical methods, such as NADH-diaphorase and acetylcholinesterase (AChE). The round or oval shaped intramural neurons were revealed among the bundles of the smooth detrusor muscle in clusters containing a variable number of cells in the groups. In both adult control and aged animals, the ganglia were enveloped by a ganglionar capsule of connective tissue mainly composed of type I collagen fibers. The number of neurons NADH-diaphorase positives estimated in the intramural plexus was 1433+/-187.71 and 1107+/-120.67 in the adult control and aged groups, respectively. The perikaryon areas of the NADH-diaphorase neurons reactives ranged from 216.40 to 1809.30 microm(2) in adult control group and from 198.20 to 2096.25 microm(2) in aged group. The nuclear area showed an increase in aged animals. The number of AChE-positive neurons estimated in the intramural plexus was 3294.67+/-415 microm(2) in the adult control group and 1960.33+/-526 microm(2) in the aged group, showing a significant decrease in the latter group. This age-related morphological change in intramural neurons may contribute to changes in urinary bladder activities in the elderly.
