Quantitative cytochemical investigations have detected individual variations between murine peritoneal macrophages and have shown distinct difference between resident and exudate populations. The latter generally contain greater amounts of protein, RNA, acid phosphatase, succinate dehydrogenase, lactate dehydrogenase, glucose-6-phosphate dehydrogenase, and NADH dehydrogenase. These estimates do not differ essentially from heterozygosity at these loci for mongoloid groups living further south. The test for interpopulation heterogeneity has permitted to estimate contribution of the loci to the differentiation of these populations. The least heterogeneity has been found at loci where gene frequency distribution is the most specific for these ethnic groups.

Polymethylmethacrylate was thermally degraded in air at 300 degrees C and the volatile decomposition products studied with gas chromatography-mass spectrometry. The main product was monomeric methacrylate, although many other compounds existed among the products. Electron spin resonance spectroscopy revealed the presence of free radicals. Wistar rats were exposed to the fumes of the plastic (300 degrees C) and their lungs and brain studied for biochemical effects. In the lung, the activity of 7-ethoxycoumarin O-deethylase decreased and an initial inhibition of glutathione peroxidase and superoxide dismutase was observed. The contents of reduced nonprotein sulfhydryl groups were decreased in the lung and brain. The exposures enhanced the activities of acetylcholine

in the brain. Scanning electron microscopy of the exposed lungs showed disorganization of ciliated cells, and the epithelial serous cells (Clara cells) were damaged.

Changes in the activity of 13 enzymes are described in the process of cytodifferentiation of the nerve cells of spinal ganglion, the motor neurons of spinal cord and large nerve cells of the III layer of tectum opticum in 7, 10 and 21 day old chick embryos. Cytophotometry was performed with MZFV-1 (LOMO) by means of plug-method. A relatively high activity of glucose-6-phosphat dehydrogenase,

was found already in the 7 days old embryo. The activity of monoamine oxidase, aldolase-glyceroaldehyde phosphate dehydrogenase, isocitrate dehydrogenase, glutamate dehydrogenase increased markedly on the 21st day. When studying the reciprocal distribution of two enzymes in separate cells, pairs of enzymes with a high value of correlation coefficient were found. The cytodifferentiation was found to be accompanied by changes in the coefficient of correlation of the same pair of enzymes.

Cultures of adult rabbit hepatocytes have been used to study the early toxic effects of 2 model hepatotoxins, dimethylnitrosamine and allyl alcohol. Leakage of glutamate oxaloacetate transaminase and glutamate pyruvate transaminase into the cell culture medium was a sensitive indicator of plasma membrane damage by these compounds and a dose-response relationship was observed. By contrast, gamma-glutamyltranspeptidase and alkaline phosphatase were insensitive markers. The effects of dimethylnitrosamine were slower to develop. Dimethylnitrosamine also produced a dose-related inhibition of protein synthesis after 4 h, a decrease in
NADPH diaphorase
and an increase in non-specific
esterase
after 20 h. Dimethylnitrosamine, unlike allyl alcohol, caused extensive disruption of ribosome association with the endoplasmic reticulum.

The quantitative cytochemical investigation of the prodigiozan effect on the enzymatic activity of the peritoneal macrophages was performed on mice. The drug was administered in a single dose of 150 microgram/kg 24 hours before the specimen collection. Prodigiozan promoted a reliable increase in the activity of the enzymes participating in glycolysis (lactate and cytoplasmic alpha-glycerophosphate dehydrogenases), hexoso-monophosphatic pathway of glucose oxidation (glucoso-6-phosphate dehydrogenase), succinate dehydrogenase, NADP. N-
diaphorase
and lysosomal enzymes, such as acid phosphatase and non-specific alpha-naphthyl acetate

. The changes indicate that activation of the macrophages is one of the significant mechanisms of increasing the host nonspecific resistance with prodigiozan.

Nascent microglia cells in the developing brain were studied by morphological, cytochemical, and autoradiographic methods. These cells are a well differentiated population, characterized by the presence of an activated nucleus, numerous ribosomes, a well developed Golgi system and associated structures including clear vesicles, dense granules, and membrane-bound polymorphous structures. Big clear vacuoles in the cytoplasm are a constant feature; filaments and microtubules are found in variable numbers. Finger-like projections and invaginations of the plasma membrane are usually found. The identification of these cells with the classical microglial cells stained by Rio Hortega's method was made by reconstruction of 20 cells using serial sections. These cells show strong

, ATPase, and acid phosphatase activities, in addition to nonspecific

activity which is inhibited by sodium fluoride. Transfusion of labeled bone marrow cells from a donor showed labeled cells only in those areas in which nascent microglia cells are more abundant. Taken together, these data suggest an exogenous, most probably monocytic, origin of nascent microglia. New studies with membrane markers are, however, needed for providing better characterization of these cells.

Reactive microglia in the developing brain after stab wound was studied by morphological, cytochemical, and autoradiographic methods. Morphologically, early reactive cells are of the "M" cell type (Matthews 1974). They show an activated nucleus, cytoplasm rich in ribosomes with wide Golgi complex and variable numbers of lipid inclusions. Big clear vacuoles are found in many of these cells. activities were demonstrated in many reactive cells. Intravenous infusion of labelled bone marrow cells from a donor showed labelled macrophages and labelled perivascular cells at the site of injury. Intracerebral injection of a small dose of tritiated thymidine at the time of injury resulted in the appearance of labelled macrophages in the following days. These data suggest that many of the reactive cells have an exogenous, more probably monocytic, origin; but a certain amount of endogenous cells also act as macrophages in brain injuries.

A range of enzymatic activities in cervical mucus-secreting, ciliated and subcolumnar basal cells were assessed using light and electron microscopic cytochemical techniques. Enzymes detected in all three cell types were those of the tricarboxylic acid cycle, pentose-phosphate and glycolytic pathways, other mitochondrial associated enzymes (NADH and
NADPH dehydrogenase
), acid phosphatase and non-specific

. Mucus-secreting and ciliated cells exhibited thiamine pyrophosphatase and 5' nucleotidase activities while leucine aminopeptidase was most convincingly demonstrated in mucus-secreting cells. Alkaline phosphatase, on the other hand, was detected only in mucus-secreting and subcolumnar basal cells. The profile of enzymatic activities in subcolumnar basal cells closely resembles that of mature lining cells and further supports the hypothesis that these cells differentiate into functioning columnar cells.

The fimbria-fornix (FF) fiber tract was unilaterally transected in adult rats by a stereotaxic knife cut. In the axotomized neurons of the medial septal nucleus (MS) and ventral diagonal band of Broca (VDB), the expression of Jun, Fos, Krox, CREB transcription factors, choline acetyltransferase (ChAT) and nitric oxide synthase (NOS) were studied by immunocytochemistry. In addition,

) and acetylcholine

(AChE) were visualized by activity assays. For retrograde tracing of axotomized neurons, either HRP-coupled gold was injected in the entorhinal cortex prior to axotomy, or Fast Blue was injected into the transection site subsequently to FF transection. Following FF transection c-Jun and in a less extend JunD were expressed in axotomized MS and VDB neurons. Expression levels rose at 24 h, but not at 18 h, postaxotomy, reached their maximal levels between 5 and 7 days, and then gradually declined. Up to 100 days, c-Jun was still present in a substantial number of septal neurons. JunB, Krox-20, Krox-24, c-Fos, and pan-Fos immunoreactivities (IR) were not detectable in axotomized septal neurons and CREB-IR did not change compared to the intact contralateral side. ChAT-IR dramatically declined over 36 h, and furthermore AChE reactivity had substantially fallen after 5 days. The number and intensity of cytoplasmic neuronal NOS-IR and

which generated congruent temporospatial patterns gradually fell between 3 and 5 days postaxotomy. The surviving neurons labeled by NOS and

showed a high coexpression of c-Jun, whereas c-Jun was almost completely absent in neurons stained for ChAT and AChE. Finally, ChAT-IR and
NDP
reaction labeled different subpopulations. Our findings demonstrate a lasting expression of the c-Jun transcription factor in axotomized MS and VDB neurons that might indicate the regenerative propensity of damaged neurons. The decrease of NOS and
NDP
in MS and VDB neurons demonstrates that neuronal populations respond to axotomy with an individual regulation of NOS expression.
