NADPH-diaphorase
positive cells were described in the visual cortex of the rat in layers II-VI and in the white matter. Their somata were large or medium-sized, oval or elongated, and their cytoplasm was accumulated at the poles. Some proximal thickened coarse dendrites formed a bitufted dendritic field. These features showed a cell type impregnated with the Golgi-Kopsch-method and a Golgi-deimpregnation-method described as Martinotti cell (sparsely spined polarized neurons with ascending axons). The immunocytochemical evidence of GABA in
NADPH-diaphorase
positive neurons (double labeling) showed the GABA ergic nature of these cells, but an attempt for a double labelling of
NADPH-diaphorase
and
Parvalbumin
was negative.

Parvalbumin
(PV)-immunoreactive neurons in rat neostriatum were studied under light and electron microscopes. A small number of neurons in the striatum were immunoreactive for PV (a Ca-binding protein). Most of them were also strongly immunoreactive for glutamate decarboxylase but were negative for
NADPH-diaphorase
activity. Light microscopic analysis revealed that PV-containing neurons have somata with fusiform or polygonal shape and are medium to large in size. The dendrites were smooth and cylindrical at the proximal portion but were varicose at the distal portion. Thin PV-immunoreactive fibers with large boutons were unevenly distributed in the striatum. Electron microscopy revealed that the somata of PV-immunoreactive neurons had a deeply indented nucleus with a nucleolus and often an intranuclear rod. These are the morphological features reported for interneurons of the striatum. Gap junctions formed between two neighboring PV-immunoreactive dendrites. A total of 175 boutons forming synapses with somata and dendrites of PV-immunoreactive neurons were examined. Of these, 115 were small in diameter (less than 1 micron), contained densely packed round vesicles and formed asymmetrical synapses mainly with dendrites. The other 60 boutons formed symmetrical synapses with somata and dendrites of PV-immunoreactive neurons. Both myelinated and unmyelinated axons with boutons were observed. PV-immunoreactive boutons had a diameter of 0.3-2 microns and contained round or elongated vesicles which were about 35 nm in diameter. The boutons formed symmetrical synapses with postsynaptic targets. Of the 100 PV-immunoreactive boutons, 51 were found on somata and proximal dendrites of medium-sized neurons containing a large, round, centrally located nucleus. The others formed synapses with dendrites of various sizes. It was occasionally observed that varicose dendrites free of spines were contacted by a large number of PV-immunoreactive boutons. The study indicates that, in the striatum, immunocytochemistry for PV selectively stains GABAergic interneurons and that the GABAergic interneurons are incorporated in a feed-forward inhibitory circuit of the striatum.

GABAergic neurons in laminae I-III of the spinal dorsal horn may contain one or more of the following compounds: glycine, acetylcholine, neuropeptide Y, enkephalin, nitric oxide synthase or parvalbumin. Although the pattern of co-localization of some of these compounds is understood, it is not known which types of GABAergic neurons contain parvalbumin, or whether nitric oxide synthase coexists with peptides, acetylcholine or parvalbumin in any of these neurons, and in this study we have used immunocytochemistry and enzyme histochemistry to resolve these issues.
Parvalbumin
-immunoreactivity was restricted to those GABA-immunoreactive neurons that also showed glycine-immunoreactivity and was not co-localized with neuropeptide Y-immunoreactivity or
NADPH diaphorase
activity. By combining

histochemistry with immunocytochemistry with an antiserum to nitric oxide synthase, we were able to show that
NADPH diaphorase
activity was a reliable marker for nitric oxide synthase in the spinal cord. Neurons that possess GABA- but not glycine-immunoreactivity may contain neuropeptide Y, enkephalin, acetylcholine or
NADPH diaphorase
, and all of the cholinergic neurons appear to contain
NADPH diaphorase
. By combining immunofluorescent detection of neuropeptide Y or enkephalin with
NADPH diaphorase
histochemistry, we showed that peptide-immunoreactivity did not coexist with
NADPH diaphorase
. This suggests that neither of these peptides coexists with nitric oxide synthase or with acetylcholine in neurons in the superficial dorsal horn. Several phenotypically distinct groups of GABA-immunoreactive neuron can therefore be identified in laminae I-III of the dorsal horn, and these may represent different functional types of inhibitory neuron.

The morphological organization of the tegmental pedunculopontine nucleus, midbrain extrapyramidal area, substantia nigra and subthalamic nucleus and their interrelationships were studied in rat organotypic culture using immunohistochemistry and
NADPH-diaphorase
histochemistry. Three coronal sections, one containing the tegmental pedunculopontine nucleus/midbrain extrapyramidal area, another with the substantia nigra and the third with the subthalamic nucleus, were obtained from postnatal 1-2-day-old rats. These sections were co-cultured for 3-4 weeks using the roller-tube technique. In the tegmental pedunculopontine nucleus/midbrain extrapyramidal area, the distribution pattern of cholinergic neurons was similar to that found in the in vivo study. We could, therefore, identify the subdivisions of the tegmental pedunculopontine nucleus (i.e., pars compacta and pars dissipata) and the midbrain extrapyramidal area. As in the in vivo situation, glutamate immunoreactive neurons were also located in these areas. Approximately 10% of
NADPH-diaphorase
positive neurons in the tegmental pedunculopontine nucleus, were glutamate immunoreactive. In the substantia nigra, as in the in vivo, tyrosine hydroxylase immunoreactive (putative dopaminergic) neurons were identified predominantly in the substantia nigra pars compacta, and parvalbumin immunoreactive neurons (putative GABAergic) mainly in the substantia nigra pars reticulata. The subthalamic nucleus was ladened with glutamate immunoreactive neurons.
NADPH-diaphorase
stained axons originating from the tegmental pedunculopontine nucleus were traced into the substantia nigra and subthalamic nucleus. They were often in close apposition to tyrosine hydroxylase immunoreactive neurons in the substantia nigra.
Parvalbumin
immunoreactive fibers from the substantia nigra projected heavily to the midbrain extrapyramidal area, but only sparsely to the tegmental pedunculopontine nucleus and the subthalamic nucleus. These findings indicate that the tegmental pedunculopontine nucleus/midbrain extrapyramidal area, substantia nigra and subthalamic nucleus in the organotypic culture have retained a basic morphological organization and connectivity similar to those seen in the in vivo situation. Therefore, this preparation could be a useful model to conduct further studies to investigate functional circuits among the structures represented.

We have examined the topography of the cerebral cortex of the Australian echidna (Tachyglossus aculeatus), using Nissl and myelin staining, immunoreactivity for parvalbumin, calbindin, and nonphosphorylated neurofilament protein (SMI-32 antibody), and histochemistry for acetylcholinesterase (AChE) and
NADPH diaphorase
. Myelinated fibers terminating in layer IV of the cortex were abundant in the primary sensory cortical areas (areas S1, R, and PV of somatosensory cortex; primary visual cortex) as well as the frontal cortex.
Parvalbumin
immunoreactivity was particularly intense in the neuropil and somata of somatosensory regions (S1, R, and PV areas) but was poor in motor cortex. Immunoreactivity with the SMI-32 antibody was largely confined to a single sublayer of layer V pyramidal neurons in discrete subregions of the somatosensory, visual, and auditory cortices, as well as a large field in the frontal cortex (Fr1). Surprisingly, SMI-32 neurons were absent from the motor cortex. In AChE preparations, S1, R, V1, and A regions displayed intense reactivity in supragranular layers. Our findings indicate that there is substantial regional differentiation in the expanded frontal cortex of this monotreme. Although we agree with many of the boundaries identified by previous authors in this unusual mammal (Abbie [1940] J. Comp. Neurol. 72:429-467), we present an updated nomenclature for cortical areas that more accurately reflects findings from functional and chemoarchitectural studies.

We have examined the cyto- and chemoarchitecture of the dorsal thalamus of the short beaked echidna (Tachyglossus aculeatus), using Nissl and myelin staining, immunoreactivity for parvalbumin, calbindin, calretinin and non-phosphorylated neurofilament protein (SMI-32 antibody), and histochemistry for acetylcholinesterase and
NADPH diaphorase
. Immunohistochemical methods revealed many nuclear boundaries, which were difficult to discern with Nissl staining.
Parvalbumin
immunoreactive somata were concentrated in the ventral posterior, reticular, posterior, lateral and medial geniculate nuclei, while parvalbumin immunoreactivity of the neuropil was present throughout all but the midline nuclei. Large numbers of calbindin immunoreactive somata were also found within the midline thalamic nuclei, and thalamic sensory relay nuclei. Immunoreactivity for calretinin was found in many small somata within the lateral geniculate "a" nucleus, with other labelled somata found in the lateral geniculate "b" nucleus, ventral posterior medial and ventral posterior lateral nuclei. Immunoreactivity with the SMI-32 antibody was largely confined to somata and neuropil within the thalamocortical relay nuclei (ventral posterior medial and lateral nuclei, lateral and medial geniculate nuclei and the posterior thalamic nucleus). In broad terms there were many similarities between the thalamus of this monotreme and that of eutheria (e.g. disposition of somatosensory thalamus, complementarity of parvalbumin and calbindin immunoreactive structures), but there were some unique features of the thalamus of the echidna. These include the relatively small size of the thalamic reticular nucleus and the preponderance of calbindin immunoreactive neurons over parvalbumin immunoreactive neurons in the ventral posterior nucleus.
