A morphological and histochemical study has been made of the primordial and early growing oocytes in the ovaries of crow (Corvus splendens) and common myna (Acridotheres tristis). The primordial oocytes in the myna ovary are loosely arranged in groups or nests, whereas in crow they form compact nests surrounded by highly vascularized connective tissue bands or lie in layers beneath the surface epithelium. The primordial oocytes in both the species are surrounded by flat granulosa cells whose number, shape, and cytochemical properties change with the initiation of growth. The oocyte nucleus shows a single basophilic nucleolus and thick diplotene chromosomes. With the initiation of growth, the number of nucleoli increases; simultaneously the chromosomes attain lampbrush configuration. Crescent-shaped Balbiani's vitelline body consists of ribonucleoproteins, lipoproteins, and phospholipids. The amount of these substances increases with the oocyte growth. The nature of proteins and lipids in the ooplasm and follicular epithelium also changes with the oocyte growth. Some randomly distributed protein bodies are also present in the ooplasm of primordial follicles. They disappear with the initiation of oocyte growth. The enzyme activities of acid phosphatase, NADP-diaphorase and NAD-diaphorase, also increase in the Balbiani's vitelline body with the oocyte growth. Alkaline phosphatase and delta 5-3 beta-HSDH activities are not seen. The possible functional significance of these morphological and histochemical changes has been discussed in relation to the initiation of growth in quiescent oocytes.

In the cells of RH, SPEV and HEp-2 lines irradiated with 6.5 mm radiowaves of 1 mW/cm2 flux density the following phenomena were established: activation of succinate dehydrogenase and ATPase; reduction of cytochrome oxidase, NAD- and NADP-diaphorase, acid and alkaline phosphatase activities; repression of 3H-thymidine incorporation in DNA and of 3H-uridine incorporation in RNA; violation of ultrastructure; suppression of cellular proliferation; decrease of mitotic activity; occurrence of pathological forms of mitosis.

Rhodospirillum rubrum cell extracts have active NADP-reductase capable of catalyzing the diaphorase reaction in the presence of methyl viologene or benzyl viologene and NADPH-generating system. The greater part of NADP-reductase is localized in the soluble fraction of destroyed cells (90-10(3) g; 90 min). The purified preparation of NADP-reductase was found to contain 6 proteins, 4-5 of them possessing diaphorase activity. Partially purified NADP-reductase preparation with a period of half-inactivation of about two days has a molecular weight of 95 000 and absorption spectrum, characterized by two maxima at 410 and 430 nm. NADP-reductase preparation possesses also menadione-reductase activity, but showed no ability for transhydrogenase reaction and reduction of cytochrome c.

The dermal cells in grey, xanthic, and white goldfish integuments were cytochemically characterized for the following enzymatic activities: tyrosinase, DOPA-oxidase, cytochrome oxidase, monoamine oxidase, peroxidase, non-specific esterase, cholinesterase, NAD-diaphorase, NADP-diaphorase, aryl sulfatase, nucleotide phosphodiesterase, beta-glucuronidase, acid phosphatase, alkaline phosphatase, adenosine triphosphatase, thiamine pyrophosphatase, glucose-6-phosphatase, aldolase, as well as succinate, malate, isocitrate, glutamate, glucose-6-phosphate, 6-phosphogluconate, alpha-glycerophosphate, alcohol, lactate, and beta-hydroxybutyrate dehydrogenases. It was found that the epidermis was a significant barrier to the access of cytochemical reaction substrates. Removal of the epidermal barrier provided dermal cell localizations of enzymatic activities which were reproducible. Further, alterations in reaction times and temperatures from the mammalian methodology provided conditions fe various integumental cells were compared for possible interrelationships. The basic foundations for future work with the dermis of poikilothermic vertebrates on an experimental basis were established. In addition, a previously undescribed non-pigmented dermal cell, the "x"-cell, was found to have enzymatic characteristics similar to both melanophores and lipophores. The "x"-cell may be the common precursor of both types of pigment cells.

Cytochrome b-559 was isolated from spinach and the alga Bumilleriopsis filiformis (Xanthophyceae) and characterized by functional properties: (a) It was active as electron acceptor in a diaphorase system using NADPH as donor and ferredoxin and ferredoxin-NADP reductase as redox proteins. (b) It exhibited photooxidation with Photosystem-I particles, when illuminated with 707 nm light. (c) It was photooxidized by Photosystem-II particles and 652 nm light at room temperature. Light greater than 702 nm was ineffective. The data corroborate previous reports on redox reactions of bound cytochrome b-559.

The arcuate nucleus (AN) and the median eminence (ME) of the hypothalamus were investigated in young and ageing female rats. During the estral cycle (EC) the monoamine (MA) content, the monoaminoxidase (MAO), NADP and NAD-diaphorase activities were determined in the AN, and the MA content and the activity of alkaline phosphatase (AP) -- in the ME. In young rats in the proestrus-estrus there was an increase in the activity of the NADP and NAD-diaphorase and of the MA content, but a decrease of the MAO activity. This indicated an intensified function of the nucleus at these stages of the EC. Accumulation of the MA in the ME was noted in the diestrus, while in the proestrus their concentration sharply fell; on the other hand, the activity of the AP was considerably increased. In the ageing rats the dynamics of the indices under study during the EC were largely unchanged. However, the functional activity of the AN proved to increase, and in the ME and elevation of the MA concentration and disturbance of its release from the nerve terminals was seen.

The activities of alkaline phosphatase, NAD diaphorase and NADP diaphorase increased in infantile mouse ovaries in response to injected gonadotrophins. The distribution and activity of these enzymes were studied in detail in the ovaries of normal mice from 1 to 41 days after birth and in mice injected at various ages with FSH, LH and HCG. Granulosa cells contained NAD and NADP diaphorases. Thecal cells contained NADP diaphorase and alkaline phosphatase with NAD diaphorase first appearing in the thecae of larger follicles 11 days after birth. All three enzymes occurred in interstitial tissue, in the interfollicular stroma and in groups of gonadotrophin-responsive cells in the medulla. These medullary cells and the interstitial tissue were stimulated by exogenous LH and HCG but not by FSH. Granulosa, theca and interfollicular tissue were stimulated at some stage by each of the three injected hormones. The normal pattern of development is discussed in relation to the changing serum levels of endogenous gonadotrophin found in similar mice. It is concluded that the enzyme changes were closely and reciprocally related to endogenous hormone concentrations.

The activity of 19 enzymes (hexokinase, glucoso-6-phosphatisomerase, alpha-glycerophosphate-, lactate-, succinate-, isocitrate-, malate-, glucoso-6-phosphate-, 6-phosphogluconate-, glutamate-, alcohol-, inosine-5'-phosphate-, guanosine-5'-monophosphate-dehydrogenase, cytochromoxidase NAD.N2- and NADP.N2-diaphorase, monoaminoxidase, alkaline and acid phosphatase) was studied comparatively in the mucosa of control rats and in tumors of the small intestine (27), and large intestine (176), induced in 41 rats percutaneously by 1,2-dimethylhydrazine. A decreased level of the enzymes of tissue respiration and Krebs cycle was found with a simultaneous increase in the activity of the enzymes of glycolysis and pentoso-monophosphate shunt. These data evidence variations in tumor metabolism consisting in oxidizing phosphorylation, being replaced by aerobic glycosis, and also reflecting an intensive proliferation of tumor cells.

Mycotic foci were studied histochemically on various experimental models of candidiasis. NAD-H, NADP-H-diaphorase, acid phosphatase and ATPase were revealed in the fungi, the activity of these enzymes depended on the state of the fungus. Diaphorase activity in the mucous membrane epithelium falls only if it is damaged by massive invasion of pseudo-mycelium. Inhibition of the enzyme activity in the visceral foci (kidney, liver, heart) occurs only in case of pronounced destruction and is not observed at the distance from the fungi. The results do not confirm the idea of fungal secretion of mycotoxins penetrating into the surrounding tissues and damaging them.

An enzymatic cycling procedure for beta-NADP+ generated by the enzyme 3'-phosphodiesterase, 2':3'-cyclic nucleotide (EC 3.1.4.37) from its substrate 2':3'-cyclic NADP+ is described. The enzymes glucose-6-phosphate dehydrogenase (EC 1.1.1.49) and diaphorase (EC 1.8.1.4) are used to cycle the cofactor between its oxidized and reduced forms in the presence of glucose-6-phosphate and p-iodonitrotetrazolium violet (INT) with the concomitant production of colored INT-formazan, monitored at 492 nm. The amplification is about 400-fold per hour and is sensitive enough to detect 6 x 10(-13) mol of NADP(H). A simple procedure for the optimization of this cycling assay is also described. Conjugates to 3'-phosphodiesterase, 2':3'-cyclic nucleotide may be used in heterogeneous enzyme immunoassays for the detection of small quantities of haptens or proteins in biological fluids.
