We have tested the ability of a representative range of dietary glucosinolates and their breakdown products, found in high concentrations in cruciferous vegetables, to act as blocking agents against carcinogenesis by inducing the activity of the anticarcinogenic phase II marker enzyme
quinone reductase
in murine hepa1c1c7 cells. Breakdown of glucosinolates was catalysed by the endogenous plant enzyme
thioglucoside glucohydrolase
at neutral and acid pH. Only two unmodified glucosinolates, p-hydroxybenzyl and 2-hydroxybut-3-enyl, significantly induced
quinone reductase
activity. However, after enzymic hydrolysis at near-neutral pH, some of the glucosinolates yielded breakdown products that significantly induced
quinone reductase
in the order: 3-methylsulphinylpropyl-->prop-2-enyl-->pent-4-enyl approximately 2-phenylethyl approximately benzyl-->all others tested. Incubation with
myrosinase
at acidic pH resulted in induction of
quinone reductase
activity by the hydrolysis products of only three of the tested glucosinolates:3-methylsulphinyl-propyl approximately 2-phenylethyl-->benzyl-->all others, activity due to the two alkenyl compounds being lost. The results show that the potential cancer-blocking action of both intact and
thioglucoside glucohydrolase

Induction of phase 2 detoxication enzymes [e.g., glutathione transferases, epoxide hydrolase, NAD(P)H:
quinone reductase
, and glucuronosyltransferases] is a powerful strategy for achieving protection against carcinogenesis, mutagenesis, and other forms of toxicity of electrophiles and reactive forms of oxygen. Since consumption of large quantities of fruit and vegetables is associated with a striking reduction in the risk of developing a variety of malignancies, it is of interest that a number of edible plants contain substantial quantities of compounds that regulate mammalian enzymes of xenobiotic metabolism. Thus, edible plants belonging to the family Cruciferae and genus Brassica (e.g., broccoli and cauliflower) contain substantial quantities of isothiocyanates (mostly in the form of their glucosinolate precursors) some of which (e.g., sulforaphane or 4-methylsulfinylbutyl isothiocyanate) are very potent inducers of phase 2 enzymes. Unexpectedly, 3-day-old sprouts of cultivars of certain crucifers including broccoli and cauliflower contain 10-100 times higher levels of glucoraphanin (the glucosinolate of sulforaphane) than do the corresponding mature plants. myrosinase
, by homogenization in a mixture of equal volumes of dimethyl sulfoxide, dimethylformamide, and acetonitrile at -50 degrees C. Extracts of 3-day-old broccoli sprouts (containing either glucoraphanin or sulforaphane as the principal enzyme inducer) were highly effective in reducing the incidence, multiplicity, and rate of development of mammary tumors in dimethylbenz(a)anthracene-treated rats. Notably, sprouts of many broccoli cultivars contain negligible quantities of indole glucosinolates, which predominate in the mature vegetable and may give rise to degradation products (e.g., indole-3-carbinol) that can enhance tumorigenesis. Hence, small quantities of crucifer sprouts may protect against the risk of cancer as effectively as much larger quantities of mature vegetables of the same variety.

Male, 4-week-old, Fischer 344 rats were administered SF or SF nitrile (200, 500, or 1000 micromol/kg) by gavage daily for 5 days. Hepatic, colonic mucosal, and pancreatic

and glutathione S-transferase activities were induced by high doses of SF, but not by SF nitrile. When Hepa 1c1c7 cells were exposed to increasing levels of each compound for 24 h,
quinone reductase
showed a 3-fold maximal induction over control at 2.5 microM SF and a 3.5-fold maximal induction over control at 2000 microM SF nitrile, the highest concentration tested. These results demonstrate that SF nitrile is substantially less potent than SF as an inducing agent of phase II detoxification enzymes. Therefore, glucoraphanin hydrolysis directed toward the production of SF rather than SF nitrile could increase the potential chemoprotective effects of broccoli.

We have compared the effects of aqueous extracts of cooked Brussels sprouts, isolated glucosinolates and their breakdown products on the activity of
quinone reductase
[NADPH:quinone-reductase] (QR) and on DNA strand breaks induced by hydrogen peroxide in murine hepa1c1c7 cells. QR activity was not significantly altered after incubation of the cells with Brussels sprouts extracts. However, some of the glucosinolates and in particular their
myrosinase
-catalysed hydrolysis products and the degradation product of indole-glucosinolates, indole-3-carbinole (I3C), di(indol-3-yl)-methane (DIM) and 2,3-bis(indol-3-ylmethyl)indole (TRI) effectively induced QR activity. Isolated isothiocyanates did not influence the QR activity. The extracts of cooked and autolysed Brussels sprouts and some glucosinolates inhibited the DNA strand breaks induced by 100 microM hydrogen peroxide. Maximum inhibition was by 20-38% after 24 h of preincubation. Hydrolysis of the glucosinolates by
myrosinase
decreased the inhibitory effects, whereas I3C, DIM or TRI had no effect on DNA damage. Accordingly, the protective effect of Brussels sprouts constituents against induction of oxidative DNA damage appears to be unrelated to enzyme inducing properties via the antioxidant responsive element. Both of these effects could be part of the suggested cancer preventive effect of cruciferous vegetables.

Rocket (Eruca sativa Mill. or Eruca vesicaria L.) is widely distributed all over the world and is usually consumed fresh (leafs or sprouts) for its typical spicy taste. Nevertheless, it is mentioned in traditional pharmacopoeia and ancient literature for several therapeutic properties, and it does contain a number of health promoting agents including carotenoids, vitamin C, fibers, flavonoids, and glucosinolates (GLs). The latter phytochemicals have recently gained attention as being the precursors of isothiocyanates (ITCs), which are released by
myrosinase
hydrolysis during cutting, chewing, or processing of the vegetable. ITCs are recognized as potent inducers of phase II enzymes (e.g., glutathione transferases, NAD(P)H:
quinone reductase
, epoxide hydrolase, etc.), which are important in the detoxification of electrophiles and protection against oxidative stress. The major GL found in rocket seeds is glucoerucin, GER (108 +/- 5 micromol g(-)(1) d.w.) that represents 95% of total GLs. The content is largely conserved in sprouts (79% of total GLs), and GER is still present to some extent in adult leaves. Unlike other GLs (e.g., glucoraphanin, the bio-precursor of sulforaphane), GER possesses good direct as well as indirect antioxidant activity. GER (and its metabolite erucin, ERN) effectively decomposes hydrogen peroxide and alkyl hydroperoxides with second-order rate constants of k(2) = 6.9 +/- 0.1 x 10(-)(2) M(-)(1) s(-)(1) and 4.5 +/- 0.2 x 10(-)(3) M(-)(1) s(-) , respectively, in water at 37 degrees C, thereby acting as a peroxide-scavenging preventive antioxidant. Interestingly, upon removal of H(2)O(2) or hydroperoxides, ERN is converted into sulforaphane, the most effective inducer of phase II enzymes among ITCs. On the other hand, ERN (and conceivably GER), like other ITCs, does not possess any chain-breaking antioxidant activity, being unable to protect styrene from its thermally (37 degrees C) initiated autoxidation in the presence of AMVN. The mechanism and relevance of the antioxidant activity of GER and ERN are discussed.

In this study, we sought to determine whether sulforaphane exerts a direct chemopreventive action on animal and human mammary tissue. enzymatic activity, as well as 4-fold elevated immunostaining of HO-1 in rat mammary epithelium, provides strong evidence of a pronounced pharmacodynamic action of sulforaphane. In a subsequent pilot study, eight healthy women undergoing reduction mammoplasty were given a single dose of a broccoli sprout preparation containing 200 mumol of sulforaphane. Following oral dosing, sulforaphane metabolites were readily measurable in human breast tissue enriched for epithelial cells. These findings provide a strong rationale for evaluating the protective effects of a broccoli sprout preparation in clinical trials of women at risk for breast cancer.

Spanish black radish (Raphanus sativus L. var. niger) is a member of the Cruciferae family that also contains broccoli and Brussels sprouts, well-known to contain health-promoting constituents. Spanish black radishes (SBR) contain high concentrations of a glucosinolate unique to the radish family, glucoraphasatin, which represents >65% of the total glucosinolates present in SBR. The metabolites of glucosinolates, such as isothiocyanates, are implicated in health promotion, although it is unclear whether glucosinolates themselves elicit a similar response. The crude aqueous extract from 0.3 to 3 mg of dry SBR material increased the activity of the phase II detoxification enzyme

These data demonstrate that the crude aqueous extract of SBR and the isothiocyanate metabolite of glucoraphasatin, MIBITC, are potent inducers of detoxification enzymes in the HepG2 cell line.

activity increased ( approximately 20%), but not EROD. Varying doses of semi-purified GRP (30, 60, or 120 mg/kg rat daily for 4 days) again caused no change in EROD activity, although a dose-dependent increase in
NQO1
was seen. Urinary excretion of mercapturic acids showed no difference between preparations, and recovery increased with decreasing dose. Histopathologic examination revealed no abnormal tissues other than cecum, where inflammation was dose dependent; mild at 120 mg/kg and severe at 240 mg/kg, a greatly supra-physiological dose.
