Glomeruli from adult normal male Wistar rats were obtained by teasing a cortex slice with stainless steel needles. The enzyme content and the morphologic aspect of these glomeruli were assessed as a preliminary step to further metabolic studies. Robinson's medium appeared to be the most suitable medium. There was no loss of glutamic dehydrogenase, glucose-6-phosphate dehydrogenase or acid phosphatase. Lactate dehydrogenase was lost to about 50%. Electron microscopy showed morphologic signs of damage in the podocytes. The glomerular oxygen uptake was measured with the help of the Cartesian diver technique, using approximately 20 glomeruli per assay. The mean dry wt of lyophilized glomeruli was determined for 13 rats for which the glomerular oxygen uptake had been measured, and these data showed a glomerular Q-02 of 4 mul/hr/mg of dry wt. The following substances were tested for their influence on the oxygen uptake: acetate, alpha-oxoglutarate, citrate, oxalacetate, glutamate, alanine, all 10 mM; succinate, 2.5, 5 and 10 mM; glucose, 5, 10 and 20 mM; fructose 10 and 20 mM; and palmitate. Citrate increases the O-2 uptake/hr/glomerulus by 30%; glucose, 20 mM, by 30%; and succinate, 2.5 mM by 50% and 10 mM by 190%. In a Robinson's medium containing 35 mg of albumin/ml, the endogenous respiration is not different from that obtained in the inorganic medium but the oxygen uptake is increased 26% by glucose, 10 mM. From these data, it can be concluded that the oxygen uptake of the glomerulus is small. This fact explains its resistance to anoxia. The systematic investigation of possible substrates indicate that glucose, citrate and succinate may play a role in supporting this small oxidative metabolism.

The realtionship between growth rate and the metabolic activity of certain liver enzymes was studied using two strains of White Plymouth Rock chickens which had been selected in divergent directions for eight-week body weight. The activities of hexokinase, glucokinase, phosphofructokinase, glucose-6-phosphate dehydrogenase, citrate synthase, glycogen synthetase, glutamate dehydrogenase and aspartate transaminase were measured at 4, 8 and 20 weeks of age. The mean percentage rate of growth of the birds selected for high eight-week body weight exceeded that of the birds selected for low eight-week body weight only during the early growth period. Thereafter, and until sexual maturity, the low-line birds grew at a faster rate, relative to body size. The mature body weight of the high-line birds exceeded that of the low-line birds by a factor of approximately 1.5. A close similarity was noted between the metabolic activity of certain liver enzymes and the growth rate (relative to body size) of the birds studied. At four and eight weeks of age, the faster-growing birds (whether high- or low-line) generally exhibited a greater capacity for glucose phosphorylation and glycolysis, but a poorer capacity for glycogen synthesis, than the slower-growing birds. At twenty weeks, growth rate and metabolic activity were similar in both strains.

Electrophoretic mobilities in polyacrylamide gel of five dehydrogenases: NADP-dependent malate dehydrogenase (NADP-MDH), 6-phosphogluconate dehydrogenase (6PGD), alcohol dehydrogenase (ADH), glucose-6-phosphate dehydrogenase (G6PD) and glutamate dehydrogenase (GDH) were investigated in a series of mouse X Chinese hamster somatic cell hybrids. Seven hybrid lines with different ratio of chromosome sets of hamster and mouse: 1:1, 2:1, 3:1 and 1:2 respectively were studied. NADP-MDH and 6PGD of both parental species and intermediate hybrid bands were present in all hybrids except two lines. These lines had only hamster MDH due to the elimination of mouse chromosomes. A correlation was found between the gene dose and the intensity of the expression of the MDH bands. The mouse type ADH was detected in all hybrids. The hamster ADH was found in one of the hybrid lines that lost all mouse chromosomes during cultivation. It is suggested that hamster ADH activity was suppressed in hybrids by the mouse genome. The species origin of GDH and G6PD could not be established due to similarity of electrophoretic mobilities of respective enzymes in parental cells.

In vitro alterations induced by a 10 micrograms/ml and 50 micrograms/ml dose each of thiophenate and fenbendazole on the absorptive surfaces of Haemonchus contortus (Nematoda: Trichostrongylidae) were studied. The most significant changes were induced in the gut epithelium. Alkaline phosphatase and adenosine triphosphatase activities were decreased, succinic dehydrogenase activity was increased, while acid phosphatase and glucose-6-phosphatase were completely lost from the intestinal epithelium after treatment with either of the drugs. A stimulatory effect of these two anthelmintics was observe on lactic dehydrogenase and reduced nicotinamide adenine dinucleotide diaphorase distribution. Thiophenate caused an increase in the activities of glutamate dehydrogenase (GDH), glucose-6-phosphate dehydrogenase (G-6-PD) and nonspecific esterases and a decrease in reduced nicotinamide adenine dinucleotide phosphate diaphorase (NADPH-D) activity. Fenbendazole treatment led to the inhibition of GDH, while G-6-PD, NADPH-D, cytochrome oxidase, monoamine oxidase and nonspecific esterase activity remained unaltered in the epithelium.

The activities of six enzymes associated with carbohydrate metabolism were measured both in carcinomas and in normal breast tissues. The following differences were observed. 1. The carcinoma showed higher enzyme activities than the normal mammary tissue. 2. The ratios of glutamate dehydrogenase, hydroxybutyrate dehydrogenase, glutathione reductase and catalase to lactate dehydrogenase were lower in carcinomas than in normal tissues. Similarly, the ratios of glutamate dehydrogenase, hydroxybutyrate dehydrogenase, glutathione reductase and catalase to glucose-6-phosphate dehydrogenase were also significantly lower in carcinomas. 3. There were no significant differences in enzyme activities between I and II stage of the disease and the metastatic tissues, however, there were significant differences between I and III stage. The significance of these findings is discussed in terms of the alterations in the balance between the metabolic pathways.

The ubiquinone systems and electrophoretic comparison of enzymes were used to determine the relatedness among 64 isolates of seven Aspergillus spp. These were 31 clinical and 3 nonclinical isolates of Aspergillus fumigatus Fres., 2 isolates of A. nidulellus Samson & W. Gams, 8 isolates of A. terreus Thom, 4 isolates of A. flavus Link, 1 isolate of A. oryzae (Ahlburg) Cohn, 14 isolates of A. niger van Tieghem, and 1 isolate of A. japonicus Saito. The enzymes glucose 6-phosphate dehydrogenase, lactate dehydrogenase, glutamate dehydrogenase, fumarase, and malate dehydrogenase were examined. The relative mobilities were analyzed numerically. The results were presented as a dendrogram. Isolates from clinical and nonclinical sources within the same species had identical ubiquinone systems and identical or very similar enzyme patterns. In the dendrogram, 64 of the tested isolates were separated into seven major clusters at a 60% similarity level. Each major cluster corresponds to a single species. On the dendrogram, A. fumigatus isolates showed homogeneity, whereas A. niger isolates showed relative heterogeneity; in particular, A. niger MF-24 and the other A. niger isolates were distantly linked to each other. All A. fumigatus isolates had the Q-10 ubiquinone system and formed a single major cluster at a similarity level of 73% or greater. Glucose 6-phosphate dehydrogenase and glutamate dehydrogenase were key enzymes for differentiating all clinical and nonclinical isolates of A. fumigatus from the other Aspergillus spp. Ubiquinone systems and enzyme patterns appear to be objective and useful indicators for use in the precise identification of clinical isolates of Aspergillus spp.

Quantitative cytochemical methods in functionally different rat brain formations (sensomotor cortex, visual cortex, nucleus caudatus, hippocampus) showed the peculiarities of the effect of tuftsin on the activity of some enzymes (the oxidative, neurotransmitter and protein metabolism enzymes) 15 min and 3 days after its single administration. No changes of activity of neurotransmitter metabolism enzymes (monoamine oxidase, acetylcholinesterase) were registered cytochemically. The specificity of the neuro-tropical effect of tuftsin on protein (activity of aminopeptidase and acid phosphatase) and oxidative (activity of glutamate dehydrogenase and glucose-6-phosphate dehydrogenase) metabolism in different functional brain systems is discussed.

In two groups of Wistar rats differing in horizontal motor activity in an open field, there was a dramatic lowering of that parameter after 2 weeks of administration of L-DOPA in the daily dose 50 mg/kg bw. The lowering in each group turned out different. Interferometric and cytophotometric studies of layers III and IV neurons of the sensorimotor cortex, n. caudatus and n. accumbens, made it possible to detect changes in the area of cytoplasm and neuronal nuclei, in their protein content, in aminopeptidase, glutamate dehydrogenase and glucose 6-phosphate dehydrogenase activity. The intensity and tendencies of the changes varied in both groups animals. It is concluded that excess dopamine produces an effect on the plastic and oxidative types of neuronal metabolism in the indicated structures.

Haemonchus contortus, incubated in 10 micrograms/ml and 50 micrograms/ml concentrations of Nilzan and albendazole in Tyrode solution were stained for histoenzymatic demonstration of various phosphatases, oxido-reductases and esterases. The intestine showed major alterations after drug treatments. The alkaline phosphatases (AkPase), adenosine triphosphatase (ATPase), glucose-6-phosphatase, succinic dehydrogenase (SDH), glutamate dehydrogenase (GDH), reduced nicotinamide adenine dinucleotide phosphate diaphorase and reduced nicotinamide adenine dinucleotide diaphorase showed a decreased activity in intestine after Nilzan treatment, whereas lactic dehydrogenase (LDH), glucose-6-phosphate dehydrogenase (G-6-PD) and monoamine oxidase resisted increased reaction. The albendazole treatment resulted in altered distribution pattern of the AkPase, ATPase, SDH, and GDH; while LDH, G-6-PD, and non-specific esterases exhibited slightly enhanced activity in the epithelium. The functional significance of these changes has been fully discussed.
