-dependent dehydrogenases in gastric adenocarcinoma and undifferentiated cancer cells were studied comparatively. The activity of
NADP
-dependent malate dehydrogenase, glutamate dehydrogenase and glucose-6-phosphate dehydrogenase was found to be high in gastric adenocarcinoma, while there was noted a more high activity of
succinate dehydrogenase
and NAD-dependent malate dehydrogenase in undifferentiated cancer. Differences ni the activity of oxido-reductive enzymes in adrenocarcinoma and undifferentiated cancer are discussed from the standpoint of various histogenesis of these forms of gastric cancer.

In a detailed study focused on the methodological problems in dehydrogenase histochemistry [e.g., fixation, diffusion of enzymes and of reduced inermediates, conversion of NADPH and
NADP
to NADH and NAD, respectively, penetration of tetrazolium salt and formazan substantivity, 'nothing dehydrogenase' reaction, use of exogenous CoQ10 and of flavoprotein substitute (PMS)], the distribution and activity of
succinate dehydrogenase
, NAD(P)H-tetrazolium reductase, glucose-6-phosphate dehydrogenase, lactate dehydrogenase (H and M types), and of L-glutamate dehydrogenase (E.C.1.4.1.2 and E.C.1.4.1.3) have been investigated in the rat cerebellum. It was evident from the study that reliable results could only be obtained if all the aforementioned factors had been considered. The image of actual concentration of SDH in the neuropil of the molecular layer could only be recorded by adding CoQ10, while other structures exhibited greater balance between SDH and endogenous mitochondrial CoQ. Contrary to previous studies, a reversed localization of the activity of G-6-PDH and LDH was noticed. The elements of molecular and Purkinje layers were rich in G-6-PDH, while the granular layer was nearly depleted. The actual level of LDH could only be recorded if NADH-tetrazolium reductase was bypassed with PMS. The H and M types of LDH coexisted in the three cortical layers, the H type being prevalent and the M type attaining its highest level in synaptic glomeruli followed by the structures of the molecular layer and the Purkinje cells. High activity of GDH was noticed in Bergmann glia followed by synaptic glomeruli, while most other structures showed weak to moderate activity. The two GDH types coexisted in all structures showing activity, except for Bergmann cells, which only showed presence of the E.C. 1.4.1.3 type. Furthermore, Bergmann glia was exceptional by showing no activity of SDH and LDH, but strong activity of G-6-PDH and NADPH-tetrazolium reductase. The granular cells were exceptional by showing weak or no activity of all enzymes in question.

The specific activity of human platelet monoamine oxidase from control subjects undergoing glucose tolerance tests is reduced drastically. Three hours after intake of 100 g of glucose only 25%-30% of the MAO-baseline activity was measured with tryptamine. beta-phenylethylamine and p-tyramine as substrates. At about 5 hr, platelet MAO activity has increased again. Inhibition was not due to small molecular weight inhibitors or other diffusible factors. Studies of other platelet enzymes, including
succinate dehydrogenase
and isocitrate dehydrogenase (
NADP+
dependent) showed no parallel reductions; hGH, insulin, blood glucose and platelet glycogen concentrations did not correlate with platelet MAO activity. The changes of MAO activity in respect with p-tyramine and tryptamine as substrates 24 hr after glucose ingestion suggest changes of the lipid microenvironment of this enzyme of the outer mitochondrial membrane.

The specific activity of a peroxisomal enzyme, lactate oxidase, and of pyruvate kinase and lactate dehydrogenase, which are not peroxisomal, increased rapidly when shaken cultures of Tetrahymena were transferred to conditions of oxygen restriction and supplemented with glucose. Two other peroxisomal enzymes, catalase and
TPN
-linked isocitrate dehydrogenase, did not increase substantially, nor did
succinate dehydrogenase
. The increases were reduced if glucose was not added at the time of transfer, and were prevented by actinomycin D or cycloheximide, but not by chloramphenicol. The results suggest an involvement of peroxisomes in the metabolism of glycolytic endproducts when the availability of oxygen to the cell is limiting.

The subcellular distribution of mitochondrial enzymes was studied in cerebral hemispheres of 15-day-old and adult rats. At both ages the synaptosomal fraction contained very little glutamate dehydrogenase (EC 1.4.1.2) but significant amounts of

(EC 1.3.99.1), glutaminase (EC 3.5.1.2), hexokinase (EC 2.7.1.1), malate

In immature brain, in the fraction enriched with free (perikaryal) mitochondria, the concentrations of these enzymes were 9.5, 1.8, 2.0, 0.92, 1.5, and 2.1 times higher, respectively, than in the synaptosomes. succinate dehydrogenase
and glutaminase was restricted to free mitochondria while hexokinase and malate
NADP
dehydrogenase accumulated and beta-hydroxybutyrate dehydrogenase diminished in both fractions. In adult brain, too, where the above ratios became 7.5, 5.2, 3.5, 0.84, 1.4, and 2.0, respectively, the concentrations of enzymes relative to each other distinguished clearly between free and synaptic mitochondria. The results substantiate previously noted signs of mitochondrial heteroeneity in adult brain, and extend them to immature brain. The chemical composition, the quantitative pattern of enzymes, of free and synaptic mitochondria is clearly different, and undergoes separate changes during postnatal differentiation.

), glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, NADH-oxydoreductase, mitochondrial glycerol-3-phosphate dehydrogenase, cytochrome c oxidase, phosphoglucomutase, glucosephosphate isomerase, glucose-6-phosphatase, acid phosphatase, beta-glucuronidase and non specific aryl esterase. Therefore, the results supply a sound foundation for those quantitative enzyme histochemical techniques in which tissue specimens are frozen or frozen-dried before enzyme estimations are performed.

The ultrastructure of feline yolk sacs from 11 stages between the 14th and the 66th day is described with reference to the endoderm and the mesothelium; supplementary histochemical and cytochemical studies are included. Despite the absence of yolk, the endodermal epithelium shows a high degree of differentiation and activity, especially in the period between the 25th and the 38th day. Large stacks of RER, abundant SER, mitochondria enveloped by RER cisternae, and a peculiar type of lysosome are the most prominent organelles. Acid phosphatase,

-diaphorases are found with high activity, whereas the 17beta-hydroxysteroid dehydrogenase assay stains the endothelium only moderately. Indications of reabsorption are less marked. In view of the apparent immaturity of the liver parenchymal cells at this stage, the yolk sac endoderm of cat is suggested to act as an important extraembryonic site of biosynthesis. As preliminary results of a chemical analysis show that the yolk sac fluid has nearly no nutritional value, the substances synthesized are believed to be transported directly to the fetus. The mesothelium shows relatively few alterations over the period studied, is less rich in organelles and is obviously far less active than the endoderm.

A change in the activity of
succinic dehydrogenase
, of the NAD- and
NADP
-diaphorases was studied in the dissociated culture of the cerebellum of neonatal rats under the effect of immune serum of the animals with an experimental allergic pertussis encephalomyelitis. There was revealed an increase in the activity of oligodendrocytes following a 3-hour exposure with the immune serum and a reduction of the activity of the enzymes under study in the same cells after a 12-hour exposure of the cultures with the serum. In difference from oligodendrocytes, the astrocytes displayed a high activity of all the enzymes under study after a 12-hour exposure.
