The corresponding enzyme activities were measured when possible. The histochemically demonstrable activities of NADH-

and MDH decreased during the first hour of ischaemia. The time course of inactivation of biochemically detectable NADH-ferricyanide oxidoreductase was much the same as that of NADH-
diaphorase
. Both histochemically and biochemically detectable beta-HBDH gradually decreased by about 6 h of ischaemia. NADH-
diaphorase
but not MDH itself proved to be the rate-limiting factor when demonstrating MDH histochemically with nitroblue tetrazolium (NBT), whereas in the case of beta-HBDH the situation was probably the reverse. CytO and
SDH
activities did not change during the experimental period. Histochemistry clearly demonstrated ischaemic cellular injury, even though no significant diagnostic changes of ischaemia were visible by light microscopy. Even though this shows that enzyme-histochemical methods can be sensitive indicators of early ischaemic injury, in practice the time between the onset of injury and death as well as between death and autopsy must be taken into consideration when interpreting the results.

A histological and histochemical study of ingested food material, energy stores and enzymes in the monogenean Pseudodactylogyrus anguillae, parasitizing the gills of the European eel (Anguilla anguilla) is presented. It was found that mucus, epithelial cells and blood from the gills were ingested. Glycogen deposits were small and primarily located in the parenchyma and to a minor extent in the vitellariae. Numerous globules of neutral lipids were found in the vitellariae. A marked esterase activity was found in the gut and a less marked activity in the vitellariae. Acid phosphatase activity was found throughout the body whereas alkaline phosphatase and leucine-amino-peptidase were not detected. Marked activity of

and NADH-
diaphorase
was found in all cells, indicating a predominantly aerobic metabolism in this monogenean.

), beta-hydroxybutyrate dehydrogenase (HBDH) and malate dehydrogenase (MDH) were demonstrated histochemically and the corresponding enzyme activities were measured biochemically in isolated perfused rats hearts after global ischemia from 0 to 12 h. The present data show that the enzyme-histochemical methods when used properly are more sensitive indicators of early ischemic injury than classical histological staining procedures. From the enzymes tested here the histochemical demonstration of HBDH turned out to be best suited for use when suspecting ischemic myocardial injury at autopsy.

in cardiomyocytes of dogs with acute experimental arterial occlusion in ischemic and postischemic periods are reviewed. An increased activity of dehydrogenases in the early periods (3,6 h) of ischemia and during recirculation was established, with its noticeable reduction at later terms (9,12 h). Medical correction of postischemic disorders was shown to improve cardiomyocyte metabolism.

, alkaline phosphatase, and acid phosphatase in the liver, kidneys, lymph nodes, and heart muscle of a total of 20 cows with positive serologic response for leukosis as well as in organs of 2 normal controls. It was found that the lymphoid cells of the leukotic proliferations and the activated endothelial and adventitial cells of the blood vessels had high alkaline phosphatase activity and negligibly expressed acid phosphatase activity. Dehydrogenase activity was low in the lymphoid-cell proliferations and in the activated cell of the vessels. The cell metabolism of the leukotic proliferations was like-wise disturbed. Histochemical methods of investigations could be used test-like in the diagnosis of bovine leukosis.

Histochemical study of enzymatic activity in the myocardium was performed in sudden cardiac death. Human hearts in which there were no macroscopic and histological focal or diffuse changes served as material. The following enzymes were studied in the anterior or posterior walls of the left ventricle or in the interventricular septum:

, lactate dehydrogenase (LDH), beta-hydroxybutyrate dehydrogenase (OHBDH), alpha-glycerophosphate- and glucose-6-phosphate dehydrogenase, NAD-

and phosphorylase. Increased activity of OHBDH and LDH was found: 36,0 and 22,6% higher than in trauma and brain hemorrhage that served as control. These alterations seem to be connected with the increase of blood content of fatty acids, and lactate as a response to the catecholamine excess. Foci of an acute ischemia were found in the interventricular septum in 80% of cases in which phosphorylase was revealed. The appearance of the ischemic foci was obviously due to the coronary arteries contraction.

By means of the light and electron microscopy, histochemical and cytophotometrical methods the anterior subarea of the cerebral limbic cortex has been studied in 30 experimental and 30 control rabbits. The experimental animals have been given 3, 15 and 30 sessions (1 h per day) of electric irritation (0.05 mA, 50 Hz, 1 msec) on the posterior hypothalamic field (PHF). Twelve rabbits from 30 control animals make an intact group and 18--a group with inactive electrodes inserted into the PHF. A single administration of chlorophos (trichlorophon) solution (600 mg/kg) (LD50) results in vacuolar distrophy appearing in the white rat liver and is especially pronounced in 2-4 days. Thirty minutes after the poisonous chemical is administered, butyrilcholinesterase (BChE) activity is inhibited by 90%, somewhat later oxidation-reduction enzymes activity decreases and alkaline phosphatase (APh) activity increases. Cytoplasm of hepatocytes is filled with glycogene and nearly deprived of RNA. Owing to the cytophotometric analysis of the enzymatic activity and the stereologic morphometry method, it has been possible to reveal a certain synchronism in the development of distrophic processes, in a decreasing activity of the oxidation-reduction enzymes and in a disturbed synthesis of glycogene and RNA. On the 6th day after chlorophos has been administered,

activity, as well as contents and distribution of RNA in hepatocytes reach their control values. BChE and APh activity does not restore. During the whole experiment there is not any statistically significant change in the volumetric part of the sinusoid capillaries and in the stellate reticuloendotheliocytes. Thus, the main effect of chlorophos action is a specific inhibition of ChE, that results in certain structural changes and in changes of the histoenzymatic parameters of the liver.

We studied the morphologic appearance of alcoholic hyalin (AH)-containing hepatocytes in liver biopsies from 14 patients with alcoholic liver disease. Most hepatocytes had a characteristic appearance. The cells were swollen and hydropic with an intact cell membrane. The mitochondria had variable-sized cristae which were both shortened and elongated. The smooth endoplasmic reticulum was markedly decreased. The rough endoplasmic reticulum was bizarre, with detachment of the ribosomes that surrounded the AH. The hepatocytes that contained AH bodies had lost almost all the glucose-6-phosphate activity but had variable amounts of

and diphosphopyridine nucleotide
diaphorase
activities. The neutrophils admixed with mononuclear cells attached themselves to the hepatocytes and then invaginated into the hepatocytic cytoplasm with focal lysis of the cell membrane mediated via the release of neutrophilic lysosomes. The distortion of protein-synthesizing organelles and decrease in glucose-6-phosphatase activity suggest that the AH-containing hepatocyte is metabolically decompensated. The final cell death may be related to the neutrophilic attack, rather than the metabolic derangement.
