The histochemical localization of six enzymic activities (acetylcholinesterase, pseudocholinesterase, monoamine oxidase, lactate dehydrogenase, succinate dehydrogenase and glucose-6-phosphate dehydrogenase) has been studied in the vagal and facial lobes of the goldfish, Carassius auratus. These encephalic centers are hypertrophic in Cyprinidae, corresponding to the dominance of gustatory function. Acetylcholinesterase shows a complex laminar distribution in the vagal lobes and a peculiar cellular localization in vagal motor neurons. Monoamine oxidase activity is mainly evident in fibrous tracts coming to or leaving from the lobes. Among oxidative enzymes examined, lactate dehydrogenase and succinate dehydrogenase exhibit distribution patterns respectively similar to those observed for acetylcholinesterase and monoamine oxidase. Some features on enzymes distribution in the gustatory centers of Carassius are in agreement with the enzymatic patterns well known in higher vertebrates.

A chromatolysis study, 14 to 21 days following denervation, showed the spinal cord representation of the nerve to the posterior latissimus dorsi muscle to be in the ventrolateral cell column between cervical ganglia 14 and 15. to characterize cevical neruos nt undergoing chromatolysis, histochemical stuies were done the cords of additional nondenervated animals. Staining reactions for beta-hydrocybutyrate dehydrogenase, succinic dehydrogenase and cholinesterase did not reveal any quantitative differences between motor neurons in cervical segments 14 and 15 of normal and dystrophic birds. Motor neurons are positive for beta-hydroxybutyrate dehydrogenase and succinic dehydrogenase, but the surrounding neuropil is positive for the latter only. No pseudocholinesterase activity is found in the ventral horn cells, but true cholinesterase is present in most of the neurons...

On the appearance in the animals (guinea pigs) of paralysis of the limbs and myasthenia after the administration of Cl. botulinum, type B, toxin, there was seen a considerable vascular hyperemia of the spinal cord, and in the neurons of the phrenic nerve nucleus there developed dystrophic-necrotic processes coursing with a marked swelling, hyperchromasia and tigrolysis. As revealed histochemically, at this stage of the botulin intoxication the neurons of the phrenic nerve nucleus displayed metabolic changes expressed in the altered activity of succinic dehydrogenase, acid phosphatase and cholinesterase.

When cervical segments 14 to 15 of the chicken spinal cord are cut transversely and studied by routine histological and histochemical methods, an onion-shaped region, filled with thread-like fibers, is seen to surround the ependymal cells of the central canal and to be bounded laterally by the neural elements of the spinal gray matter. This area is negative for succinic dehydrogenase, beta-hydroxybutyrate dehydrogenase and cholinesterase activity, but very strongly periodic acid-Schiff positive. Diastase controls show the positive material to be glycogen. Parasagittal sections through this cervical region and into the upper thoracic cord, show the glycogen-rich region to extend longitudinally throughout the region. Because of its location and histochemical characterization, which are similar to that of the ventral portion of the glycogen body, the term brachial glycogen body is proposed for this structure.

The thyroid gland of guinea pigs were studied morphologically. Histochemical methods were used for detection of lactate dehydrogenase, succinic dehydrogenase, cholinesterase, alkaline phosphatase and acid phosphatase. The distribution of "C"-cells in normal thyroid glands was proved to be uneven. In the center of the gland they were more numerous. For statistical investigations the method of silver impregnation of "C"-cells is more practicable, since they can not be obviously distinguished from acinar cells on the basis of glycerophosphate dehydrogenase only. The activity of cholinestarase in "C"-cells and in some other cells of folliculi epithelium is very high. A supposition is made that there exist two kinds of the follicular lining thyrocytes, having different histochemical properties and histogenesis as well.

In the structures of the nucleus supraopticus, changes of the activity of some enzymes (alkaline phosphatase, acid phosphatase, thiamine pyrophosphatase, butyrylcholinesterase, succinate dehydrogenase, glycerol-3-phosphate dehydrogenase) were studied in rat brains exposed to high supralethal doses of gamma radiation at early time interval after irradiation. The activity of alkaline phosphatase, acetylcholinesterase and butyrylcholinesterase increased in the wall of blood capillaries after irradiation with 50, 150, 500 Gy. The dose of 500 Gy induced the most pronounced activity. These membrane enzymes are highly sensitive to ionizing radiation. The activity of acid phosphatase, acid nonspecific esterase and thiamine pyrophosphatase increased in magnocellular neurons after irradiation with all doses of gamma radiation. Glycerol-3-phosphate dehydrogenase and succinate dehydrogenase showed a decreased activity in neurons, neuropil and capillaries.

The neurochemical changes induced by malathion, an organophosphate compound, were determined in rats. Maximal changes were found in the brain 2 h after the administration of malathion in a dose of 500 mg/kg ip. The activities of cholinesterase and succinic dehydrogenase were reduced whereas those of glycogen phosphorylase, phosphoglucomutase, and hexokinase were increased; the lactate content of brain was also increase. In malathion treated adrenalectomized animals, changes in the activities of cerebral cholinesterase and succinic dehydrogenase were still present; other changes were, however, abolished by adrenalectomy. Activities of certain enzymes, glucose-6-phosphatase, glucose-6-phosphate dehydrogenase, and lactate dehydrogenase were not significantly altered by malathion in normal or adrenalectomized animals. The results indicate that cerebral cholinergic mechanism in malathion treated animals was not modified by adrenalectomy which, however, abolished or reduced changes in the activities of certain glycolytic and glycogenolytic enzymes that are involved in the utilization or metabolism of glucose. The brain lactate content in malathion treated adrenalectomized animals was, also, not significantly different from the control values, suggesting that modification of induced changes by adrenalectomy.

Effect of injection in third ventricle of GABA, the GABA agonist muscimol, and the GABA antagonist picrotoxin on the activities of acetylcholinesterase (AChE), butyrylcholinesterase (BuChE) and monoamine oxidase (MAO) in serum and succinic dehydrogenase (SDH) in plasma has been studied. Surprisingly, the AChE, BuChE, MAO and SDH enzymes activity were inhibited by GABA and muscimol, while they were enhanced by picrotoxin.

The deltoid and gluteus maximus of the desert rat and the albino rat were examined histochemically for the distribution of succinic dehydrogenase (SDH) and cholinesterase (ChE). SDH activity showed that the deltoid and gluteus maximus muscles of the two animals consisted of three types of muscle fibres, with a predominance of muscle fibres that have higher SDH activity in both the deltoid and gluteus maximus muscles of the desert rat than in the albino rat. The mean diameter of all muscle fibres in the deltoid and gluteus maximus muscles and their ratios to the average body weight were determined in both animals. The desert rat showed a statistically significant increase in those ratios compared with the corresponding ratios for the albino rat. ChE activity showed that the deltoid and gluteus maximus muscles are richly innervated by intensely positive ChE motor end-plates with a predominance of plaque-like terminals. The mean diameters of the end-plates and the ratios of these diameters to the diameter of the muscle fibres together with their ratios to the body weight were determined. A correlation between these values and the histological findings is proposed.

In the present study, the effect of intracerebroventricular (icv) injection of GABA, its agonist--muscimol, and antagonist--picrotoxin, has been studied on histoenzymological alterations of acetylcholinesterase (AChE). butyrylcholinesterase (BuChE), monoamine oxidase (MAO), and succinic dehydrogenase (SDH) by cytophotometric technique. This study was conducted on medial preoptic area (mPOA), nucleus paraventricularis hypothalami (PVH), area lateralis hypothalami (LHA), nucleus dorsomedialis hypothalami (DMH), and nucleus ventromedialis hypothalami (VMH). Results showed that GABA and muscimol inhibited AChE, BuChE, MAO, and SDH in all the areas while picrotoxin stimulated these enzymes. These changes in enzyme activity by GABA, muscimol, and picrotoxin and their possible mode of action are discussed.
